    Effect of Porcine circovirus 2 (PCV-2) maternally derived antibodies on performance and PCV-2 viremia in vaccinated piglets under field conditions

    Background: Nowadays, the most common presentation of PCV-2 is the subclinical infection in piglets after weaning. The success of PCV-2 vaccination is associated with the control of the clinical disease as well as the improvement of production parameters. In consequence, the objective of the present study was to analyse the effect of PCV-2 maternally derived antibody (MDA) levels on vaccine efficacy in piglets vaccinated at three weeks of age with a commercial PCV-2 subunit vaccine. The study was performed analysing a database with 6112 wean-to-slaughter piglets from 4 different European regions. Results: Results showed that the use of the vaccine was able to decrease the PCV-2 viremia calculated as area under the curve (AUC = 60.29 ± 3.73), increase average daily weight gain (ADWG = 0.65 ± 0.01 kg/day) and reduce mortality (7%) in vaccinated piglets compared to non-vaccinated ones (AUC of 198.27 ± 6.14, 0.62 ± 0.01 kg/day and 11% respectively). The overall difference of ADWG between both groups was close to 30 g per day (p < 0.05), also when they were split for low and high levels of MDA titres. Moreover, the animals with the highest ADWG were observed in the group of piglets vaccinated with high or extremely high antibody titres (0.66 and 0.65 kg/day respectively). Considering only animals with extremely high antibody titres, both study groups performed similar, however there was a numerical difference of 10 g/day in favour of vaccinated piglets. Likewise, lack of correlation between ADWG and MDA was observed suggesting that no maternal antibody interference was present with the tested vaccine because the vaccinated animals grew faster compared to unvaccinated control animals, regardless of the level of maternal antibodies present at the time of vaccination. Conclusions: The results of the present study demonstrated that the MDA against PCV-2 transferred through the colostrum intake has a protective effect against this viral infection. The vaccine used in the present study (Ingelvac CircoFLEX®) was effective when applied at three weeks of age and was not affected by the level of MDA at the time of vaccination

Las directrices PRISMA-P se componen de una lista de verificación de 17 elementos que pretenden facilitar la preparación y presentación de un protocolo sólido para la revisión sistemática. Las instituciones financiadoras y quienes encargan las revisiones podrían considerar la posibilidad de exigir la lista de verificación, a fin de facilitar la presentación de la información relevante del protocolo, para solicitar financiación. De forma similar, los revisores por pares y editores pueden utilizar las directrices para determinar la integridad y transparencia de un protocolo de revisión sistemática presentado a una revista u otro medio para su publicación.Systematic reviews should build on a protocol that describes the rationale, hypothesis, and planned methods of the review; few reviews report whether a protocol exists. Detailed, well-described protocols can facilitate the understanding and appraisal of the review methods, as well as the detection of modifications to methods and selective reporting in completed reviews. We describe the development of a reporting guideline, the Preferred Reporting Items for Systematic reviews and Meta-Analyses for Protocols 2015 (PRISMA-P 2015). PRISMA-P consists of a 17-item checklist intended to facilitate the preparation and reporting of a robust protocol for the systematic review. Funders and those commissioning reviews might consider mandating the use of the checklist to facilitate the submission of relevant protocol information in funding applications. Similarly, peer reviewers and editors can use the guidance to gauge the completeness and transparency of a systematic review protocol submitted for publication in a journal or other medium

    P82 238. ¿Es segura la reintervención dejando los injertos arteriales permeables sin clampar?

    ObjetivoEl objetivo de no dañar la arteria mamaria evitando su disección y clampaje supone un reto para la protección miocárdica debido al lavado de la cardioplejía.Presentamos nuestra experiencia en reintervenciones sin disecar ni clampar los injertos arteriales.Material y métodosDesde septiembre de 2000 hasta febrero de 2010 se realizaron 29 reintervenciones en 28 pacientes, (89,7% varones), edad media 73,17±7,38años. Las causas de reoperación fueron: progresión de valvulopatía 17 pacientes (58,6%), endocarditis 10 (34,4%), disfunción protésica no estructural 2 (6,9%). La mediana del EuroS-CORE logístico fue 14,84 (4-77,25). La cirugía fue urgente en 7 pacientes. Se realizó sustitución valvular aórtica aislada en 18 y se asoció revascularización en 4, sustitución/plastia mitral en 4, sustitución de aorta en 2; sustitución mitral aislada en 1.La protección miocárdica se realizó con cardioplejía hemática con esmolol, K+ y Mg+, administrándola siempre que no dificultaba el trabajo quirúrgico (intervalos nunca > 20min). Temperatura sistémica media 32,26±3,23 °C.ResultadosLa mediana del tiempo de circulación extracorpórea (CEC) fue 153 (91-494) y de clampaje 103 (71-430)min. Presentaron infarto postoperatorio 1 paciente (3,4%) y síndrome de bajo gasto postoperatorio 2 pacientes (6,5%). La mediana de troponina I postoperatoria fue 7,03ng/ml (1,84-109,5). La mortalidad hospitalaria fue 3 pacientes (10,3%) (cirugía urgente por endocarditis). Las causas de mortalidad fueron: daño neurológico irreversible (1 paciente), sepsis y bajo gasto (2 pacientes). La mediana de estancia hospitalaria fue 7 (1-33) días.ConclusiónEn nuestra experiencia, sin clampar los injertos arteriales y con la estrategia descrita, la protección miocárdica parece adecuada y la mortalidad hospitalaria aceptable

    Magnetorheology of alginate ferrogels

    Magnetorheological (MR) effect is a phenomenon typical of suspensions of magnetizable particles in a liquid carrier, characterized by strong changes of their mechanical properties in response to applied magnetic fields. Its origin is on the migration of magnetized particles and their aggregation into chain-like structures. However, for ferrogels, consisting of dispersions of magnetic particles in a polymer matrix, migration of particles is hindered by the elastic forces of the polymer network, preventing from strong MR effect. Interestingly, we demonstrate in this manuscript that strong MR effect in robustly cross-linked polymer ferrogels is still possible. Experimental results showed enhancement of the storage modulus of more than one order of magnitude for alginate ferrogels containing less than about 10 vol.% of iron particles under moderate magnetic fields. The differential feature of these ferrogels is that, instead of individual particles, the disperse phase consisted of large clusters of iron microparticles homogeneously distributed within the polymer networks. These clusters of magnetic particles were formed at the stage of the preparation of the ferrogels and their presence within the polymer networks had two main consequences. First, the volume fraction of clusters was considerably larger than this of individual particles, resulting in a larger effective volume fraction of solids. Second, since the force of magnetic attraction between magnetic bodies is roughly proportional to the cube of the body size, the existence of such clusters favored inter-cluster interaction under a magnetic field and the appearance of strong MR effect. On this basis, we demonstrated by theoretical modeling that the strong MR effect displayed by the alginate ferrogels of the present work can be quantitatively explained by assuming the existence of large, roughly spherical particle aggregates formed at the stage of the preparation of the ferrogels.     Astrocytic glycogen accumulation drives the pathophysiology of neurodegeneration in Lafora disease

    The hallmark of Lafora disease, a fatal neurodegenerative disorder, is the accumulation of intracellular glycogen aggregates, called Lafora bodies. Until recently, it was widely believed that brain Lafora bodies were present exclusively in neurons and thus that Lafora disease pathology derived from their accumulation in this cell population. However, recent evidence indicates that Lafora bodies are also present in astrocytes. To define the role of astrocytic Lafora bodies in Lafora disease pathology, we deleted glycogen synthase specifically from astrocytes in a mouse model of the disease (malinKO). Strikingly, blocking glycogen synthesis in astrocytes-thus impeding Lafora bodies accumulation in this cell type-prevented the increase in neurodegeneration markers, autophagy impairment, and metabolic changes characteristic of the malinKO model. Conversely, mice that overaccumulate glycogen in astrocytes showed an increase in these markers. These results unveil the deleterious consequences of the deregulation of glycogen metabolism in astrocytes and change the perspective that Lafora disease is caused solely by alterations in neuron

    Comparison of fiber effect on glycemic index and glycemic load in differents types of bread

Numerosos estudios se han llevado a cabo con el fin de reducir su índice glicémico, sin embargo, aún existe controversia sobre la acción de la fibra dietética en la disminución del IG en este alimento. Este estudio determinó el efecto de la fibra dietética sobre el índice glicémico y carga glicémica en dos tipos de panes comerciales en 23 individuos sanos quienes consumieron aleatoriamente 3 diferentes productos, de 50 g de carbohidratos cada uno, durante 6 días: pan blanco (PH), pan integral (PF), y solución glucosada como producto de referencia (SG). Se midió glicemia en ayunas y post-prandial a los tiempos 15, 30, 45, 60, 90 y 120 min. La insulina fue medida en el minuto 0 y 120 min. El área bajo la curva de glicemia resultó más baja para ambos tipos de pan PH 13589 ±1557, PF 12005 ±1254 que para el producto de referencia SG 14089 ±1245. Los valores del índice glicémico PH 68,55 ±1,2 y PF 62,10 ±1,3 y carga glicémica PH 16,45 ±1,4 resultaron más bajos para el pan con mayor aporte de fibra 9,93 ± 1,1, sin diferencias en la concentración de insulina, sugiriendo que la cantidad de carbohidratos y tipo de fibra contenidos en el pan integral, pueden considerarse factores intrínsecos en su composición nutricional, capaces de afectar la respuesta glicémica post- ingesta de estos productos en individuos sanos.There are several foods that contain carbohydrates as the main nutrient, being one of the most important the bread for its massive worldwide consumption. Numerous studies have been done in order to reduce its glycemic index, however there is still controversy about the action of dietary fiber in the decrease of GI in this product. In this study, it was determined the effect of dietetic fiber on glycemic index and glycemic load in two types of commercial breads in 23 healthy individuals who randomly consumed 3 different products during 6 days of 50g of carbohydrates each: white bread (PH), whole wheat bread (PE) and glucose solution as reference product (SG). Fasting and postprandial glycemia was measured at times 15, 30, 45, 60, 90 and 120 minutes. Insuline was measured at 0 min and 120 min. The area under de glycemia curve was lower for both bread types PH 13589 ±1557, PF 12005 ±1254 than for the reference product SG 14089 ±1245. The values of the glycemic index PH 68,55 ±1,2 and PF 62,01 ±1,3 and glycemic load PH 16.45 ±1,4 were lower for bread with more amount fiber 9,93 ± 1,1, with no difference in insulin concentration, suggesting that the amount of carbohydrates and fiber type contained in whole wheat bread can be considered intrinsic factors in bread composition, affecting the post-intake glycemic response of this type of products in healthy individuals.http://www.redalyc.org/articulo.oa?id=5594990800

    Effect of oat β-glucan on glycemic index and glycemic load of a nutritional supplement sweetened with sucralose in healthy adults: A randomized clinical trial

Existen suplementos nutricionales específicos para diabéticos edulcorados con sucralosa cuyo índice glicémico y carga glicémica aún no han sido establecidos. El efecto del β-glucano de avena sobre el índice glicémico y carga glicémica de un suplemento nutricional edulcorado con sucralosa, fue determinado en 13 adultos sanos (6 hombres y 7 mujeres), quienes consumieron aleatoriamente 4 alimentos en días distintos, de 50 g de carbohidratos cada uno: suplemento nutricional para diabéticos (FN), suplemento nutricional con β-glucano (FN- β), y como productos de referencia: solución glucosada (SG) y pan blanco (PB). Se midió glicemia en ayunas y post- prandial a los tiempos 15, 30, 45, 60, 90 y 120 min. El área bajo la curva de glicemia resultó más baja para ambas fórmulas (FN) 12697±993, (FN-β) 11584 ±1171, que para los productos de referencia:(SG) 13900±1245, y (PB) 13267 ± 1557. Los valores de índice glicémico (FN) 67,02 ± 5,69, así como la carga glicémica resultaron intermedios y más bajos para el suplemento con β-glucano incorporado (FN –β) 59,8 ± 6,2; sin diferencias en la concentración de insulina, sugiriendo que la adición del β-glucano derivado de la avena reduce la velocidad de absorción intestinal de la glucosa, efecto que podría estudiarse en diabéticos.The hypoglycemic properties of oat β-glucan is of interest for the food industry and clinical area, for potencial health benefits by reducing glycemic response, serum low-density lipoprotein cholesterol, and glycemic index of meals. There are specific nutritional supplements for diabetics sweetened with sucralose whose glycemic index and glycemic load has not been established. Effect of oat β-glucan on glycemic index and glycemic load of a nutritional supplement sweetened with sucralose in healthy adults was determined in 13 healthy subjects (6 men and 7 women) old that consumed randomly 4 meals of 50 g of carbohydrates each in different days: a nutritional supplement for diabetics (FN), the nutritional supplement with β-glucan incorporated (FN-β) and two reference food, glucose solution (SG) and white bread (PB). Fasting and postprandial glycemia was measured at times 15, 30, 45, 60, 90 and 120 min. The area under the glycemia curve was lower for both formulas (FN) 12697±993, (FN-β) 11584 ±1171 than for reference products (SG) 13900±1245, y (PB) 13267 ± 1557. The values of glycemic index (GI) (FN) 67, 02 ± 5,69 and glycemic load were intermediate and more lower for the supplement with β-glucan incorporated (FN –β) 59, 8 ± 6,2, with no difference of insulin concentration .     Molecular map of chronic lymphocytic leukemia and its impact on outcome

    Recent advances in cancer characterization have consistently revealed marked heterogeneity, impeding the completion of integrated molecular and clinical maps for each malignancy. Here, we focus on chronic lymphocytic leukemia (CLL), a B cell neoplasm with variable natural history that is conventionally categorized into two subtypes distinguished by extent of somatic mutations in the heavy-chain variable region of immunoglobulin genes (IGHV). To build the ‘CLL map,’ we integrated genomic, transcriptomic and epigenomic data from 1,148 patients. We identified 202 candidate genetic drivers of CLL (109 new) and refined the characterization of IGHV subtypes, which revealed distinct genomic landscapes and leukemogenic trajectories. Discovery of new gene expression subtypes further subcategorized this neoplasm and proved to be independent prognostic factors. Clinical outcomes were associated with a combination of genetic, epigenetic and gene expression features, further advancing our prognostic paradigm.
