    Nuclear Surveillance and Degradation of Hypomodified Initiator tRNA\u3csup\u3eMet\u3c/sup\u3e in \u3cem\u3eS. cerevisiae\u3c/em\u3e

    The tRNA m1A58 methyltransferase is composed of two subunits encoded by the essential genes TRM6 and TRM61 (formerly GCD10 and GCD14). The trm6-504 mutation results in a defective m1A methyltransferase (Mtase) and a temperature-sensitive growth phenotype that is attributable to the absence of m1A58 and consequential tRNAiMet instability. We used a genetic approach to identify the genes responsible for tRNAiMet degradation in trm6 cells. Three recessive extragenic mutations that suppress trm6-504 mutant phenotypes and restore hypomodified tRNAiMet to near normal levels were identified. The wild-type allele of one suppressor, DIS3/RRP44, encodes a 3′-5′ exoribonuclease and a member of the multisubunit exosome complex. We provide evidence that a functional nuclear exosome is required for the degradation of tRNAiMet lacking m1A58. A second suppressor gene encodes Trf4p, a DNA polymerase (pol σ) with poly(A) polymerase activity. Whereas deletion of TRF4 leads to stabilization of tRNAiMet, overexpression of Trf4p destabilizes the hypomodified tRNAiMet in trm6 cells. The hypomodified, but not wild-type, pre-tRNAiMet accumulates as a polyadenylated species, whose abundance and length distribution both increase upon Trf4p overexpression. These data indicate that a tRNA surveillance pathway exists in yeast that requires Trf4p and the exosome for polyadenylation and degradation of hypomodified pre-tRNAiMet

    Realeasy: Real-Time capable Simulation to Reality Domain Adaptation

    We address the problem of insufficient quality of robot simulators to produce precise sensor readings for joint positions, velocities and torques. Realistic simulations of sensor readings are particularly important for real time robot control laws and for data intensive Reinforcement Learning of robot movements in simulation. We systematically construct two architectures based on Long Short-Term Memory to model the difference between simulated and real sensor readings for online and offline application. Our solution is easy to integrate into existing Robot Operating System frameworks and its formulation is neither robot nor task specific. The collected data set, the plug-and-play Realeasy model for the Panda robot and a reproducible real-time docker setup are shared alongside the code. We demonstrate robust behavior and transferability of the learned model between individual Franka Emika Panda robots. Our experiments show a reduction in torque mean squared error of at least one order of magnitude

    SWI/SNF-like chromatin remodeling factor Fun30 supports point centromere function in S. cerevisiae

    Budding yeast centromeres are sequence-defined point centromeres and are, unlike in many other organisms, not embedded in heterochromatin. Here we show that Fun30, a poorly understood SWI/SNF-like chromatin remodeling factor conserved in humans, promotes point centromere function through the formation of correct chromatin architecture at centromeres. Our determination of the genome-wide binding and nucleosome positioning properties of Fun30 shows that this enzyme is consistently enriched over centromeres and that a majority of CENs show Fun30-dependent changes in flanking nucleosome position and/or CEN core micrococcal nuclease accessibility. Fun30 deletion leads to defects in histone variant Htz1 occupancy genome-wide, including at and around most centromeres. FUN30 genetically interacts with CSE4, coding for the centromere-specific variant of histone H3, and counteracts the detrimental effect of transcription through centromeres on chromosome segregation and suppresses transcriptional noise over centromere CEN3. Previous work has shown a requirement for fission yeast and mammalian homologs of Fun30 in heterochromatin assembly. As centromeres in budding yeast are not embedded in heterochromatin, our findings indicate a direct role of Fun30 in centromere chromatin by promoting correct chromatin architecture

This team is composed of undergraduate and graduate students with a goal to design and build the most attractive, effective, and energy-efficient solar-powered house for the 2009 Solar Decathlon. The Solar Decathlon, an international design competition, exists to educate the public on solar energy, energy efficiency, and the best in home design. While this effort is well over the budget available for the current funding opportunity, we have selected certain components for submission in this application. Specifically, this effort includes architecture, interior design, architectural engineering, electrical engineering, civil engineering, history, and business disciplines across two campuses in a unique experience for all involved and utilizes technology and IT communications to make it happen. Most of the components of the project are done through computer and the virtual world. The students must rely on the ability to communicate via the internet for most decisions because only occasional face-to-face meetings take place. In addition, students on both campuses utilize computer programs related to their discipline and learn programs associated with other disciplines due to the multidisciplinary efforts required. For example, MU students often use Google Sketch?up to convey the architectural elements of the home. S&T students, new to Google Sketch-up, have found that with a tutorial by MU that they can better understand the dialogue going on between the teams on the design. We believe that the Show-Me Solar team�s proposal for the Interdisciplinary Innovation Fund meets the requirements from the MU Information Technology Committee. The student-led team is working hard to make the University of Missouri system a leader in the next wave of home design through the use of innovative technology and strong relationships between disciplines.     The effects of entrepreneurship education

    Entrepreneurship education ranks high on policy agendas in Europe and the US, but little research is available to assess its impact. To help close this gap we investigate whether entrepreneurship education a?ects intentions to be entrepreneurial uniformly or whether it leads to greater sorting of students. The latter can reduce the average intention to be entrepreneurial and yet be socially beneficial. This paper provides a model of learning in which entrepreneurship education generates signals to students. Drawing on the signals, students evaluate their aptitude for entrepreneurial tasks. The model is tested using data from a compulsory entrepreneurship course. Using ex ante and ex post survey responses from students, we find that intentions to found decline somewhat although the course has significant positive e?ects on students’ self-assessed entrepreneurial skills. The empirical analysis supports the hypothesis that students receive informative signals and learn about their entrepreneurial aptitude. We outline implications for educators and public policy

    Cyanobacteria net community production in the Baltic Sea as inferred from profiling pCO(2) measurements

    Organic matter production by cyanobacteria blooms is a major environmental concern for the Baltic Sea, as it promotes the spread of anoxic zones. Partial pressure of carbon dioxide (pCO(2)) measurements carried out on Ships of Opportunity (SOOP) since 2003 have proven to be a powerful tool to resolve the carbon dynamics of the blooms in space and time. However, SOOP measurements lack the possibility to directly constrain depth-integrated net community production (NCP) in moles of carbon per surface area due to their restriction to the sea surface. This study tackles the knowledge gap through (1) providing an NCP best guess for an individual cyanobacteria bloom based on repeated profiling measurements of pCO(2) and (2) establishing an algorithm to accurately reconstruct depth-integrated NCP from surface pCO(2) observations in combination with modelled temperature profiles.Goal (1) was achieved by deploying state-of-the-art sensor technology from a small-scale sailing vessel. The low-cost and flexible platform enabled observations covering an entire bloom event that occurred in July-August 2018 in the Eastern Gotland Sea. For the biogeochemical interpretation, recorded pCO(2) profiles were converted to C-T*, which is the dissolved inorganic carbon concentration normalised to alkalinity. We found that the investigated bloom event was dominated by Nodularia and had many biogeochemical characteristics in common with blooms in previous years. In particular, it lasted for about 3 weeks, caused a C-T* drawdown of 90 mu mol kg(-1), and was accompanied by a sea surface temperature increase of 10 degrees C. The novel finding of this study is the vertical extension of the C-T* drawdown up to the compensation depth located at around 12 m. Integration of the C-T* drawdown across this depth and correction for vertical fluxes leads to an NCP best guess of similar to 1:2 mol m(-2) over the productive period.Addressing goal (2), we combined modelled hydrographical profiles with surface pCO(2) observations recorded by SOOP Finnmaid within the study area. Introducing the temperature penetration depth (TPD) as a new parameter to integrate SOOP observations across depth, we achieve an NCP reconstruction that agrees to the best guess within 10 %, which is considerably better than the reconstruction based on a classical mixed-layer depth constraint.Applying the TPD approach to almost 2 decades of surface pCO(2) observations available for the Baltic Sea bears the potential to provide new insights into the control and long-term trends of cyanobacteria NCP. This understanding is key for an effective design and monitoring of conservation measures aiming at a Good Environmental Status of the Baltic Sea

    Ten-eleven-translocation (TET) proteins catalyze DNA hydroxylation, playing an important role in demethylation of DNA in mammals. Remarkably, although hydroxymethylation levels are high in the mouse brain, the potential role of TET proteins in adult neurogenesis is unknown. We show here that a non-catalytic action of TET3 is essentially required for the maintenance of the neural stem cell (NSC) pool in the adult subventricular zone (SVZ) niche by preventing premature differentiation of NSCs into non-neurogenic astrocytes. This occurs through direct binding of TET3 to the paternal transcribed allele of the imprinted gene Small nuclear ribonucleoprotein-associated polypeptide N (Snrpn), contributing to transcriptional repression of the gene. The study also identifies BMP2 as an effector of the astrocytic terminal differentiation mediated by SNRPN. Our work describes a novel mechanism of control of an imprinted gene in the regulation of adult neurogenesis through an unconventional role of TET3

    Growth in densely populated Asia: implications for primary product exporters

    Economic growth and integration in Asia is rapidly increasing the global economic importance of the region. To the extent that this growth continues and is strongest in natural resource-poor Asian economies, it will add to global demand for imports of primary products, to the benefit of (especially nearby) resource-abundant countries. How will global production, consumption and trade patterns change by 2030 in the course of such economic developments and structural changes? We address this question using the GTAP model and Version 8.1 of the 2007 GTAP database, together with supplementary data from a range of sources, to support projections of the global economy from 2007 to 2030 under various scenarios. Factor endowments and real gross domestic product are assumed to grow at exogenous rates, and trade-related policies are kept unchanged to generate a core baseline, which is compared with an alternative slower growth scenario. We also consider the impact of several policy changes aimed at increasing China's agricultural self-sufficiency relative to the 2030 baseline. Policy implications for countries of the Asia-Pacific region are drawn out in the final section

    “Peers, parents and phones”—Swedish adolescents and health promotion

    Many unhealthy behaviors are created during adolescence and follow the individual into adulthood. In addition, health behaviors often occur in clusters as those who are inactive are more likely to eat unhealthy food and smoke. This makes the early foundation of healthy behaviors vital. The aim was to describe and develop an understanding of adolescents’ awareness and experiences concerning health promotion. Data was collected using focus groups with a total of 28 seventh graders and was analysed with latent qualitative content analysis. One main theme was identified; being competent, ambivalent and creative at the same time. The following three subthemes also emerged: being a digital native for better and for worse, knowing what is healthy, and sometimes doing it, and considering change and having ideas of how change could be supported. The main theme elucidates how the majority of students were informed and able but they did not always prioritize their health. The concept of health promotion relies upon the engagement of the individual; however, although the students had clear ideas about how they would like to change their own behaviors, they felt a need for support. Interestingly, the students were able to make several suggestions about the kind of support that would make a difference to their adoption to more healthy modes of living. They suggested information and communication technology (ICT), for example encouraging text messages (SMS), and social support, for example parents setting rules and peers inspiring them to adhere to a healthy behavior. The knowledge gained from this study echoes our view of inclusion and this could be helpful for those who encounter the challenge of promoting health among adolescents
