    Drugs, Depression, and Dating Violence: Partnering With Schools to Collect and Use Data on Adolescent Risky Behaviors

    In 2005, the MetroWest Health Foundation launched a 10-year initiative to conduct the federal Centers for Disease Control and Preventionā€™s Youth Risk Behavior Survey biennially with every high school and middle school student in the foundationā€™s 25-town region. The survey asks students about substance use, violence, sexual behaviors, mental health, and nutrition. In the first year of the initiative, about two-thirds of public high schools and half of the middle schools in the region participated. By 2012, every public high school and middle school participated. Encompassing some 40,000 students, the survey is considered to be one of largest, if not the largest, regional census of risky behavior among youth in the country. Schools and communities use the data to change school and law enforcement policies, implement new curriculum, educate parents, expand time for school wellness programs, and secure federal grants. The foundation has used the data to measure the impact of its substance abuse and antibullying initiatives as well as to develop new funding programs, such as adolescent mental health. The foundation\u27s research partner, Education Development Center, was asked to share the data with New York City, which used the data to demonstrate that increasing the minimum age to purchase tobacco is associated with reduced rates of tobacco use among youth

    Simulating Reionization in Numerical Cosmology

    The incorporation of radiative transfer effects into cosmological hydrodynamical simulations is essential for understanding how the intergalactic medium (IGM) makes the transition from a neutral medium to one that is almost fully ionized. Here, we present an approximate numerical method designed to study in a statistical sense how a cosmological density field is ionized by a set of discrete point sources. A diffuse background radiation field is also computed self-consistently in our procedure. The method requires relatively few time steps and can be employed with simulations having high resolution. We describe the details of the algorithm and provide a description of how the method can be applied to the output from a pre-existing cosmological simulation to study the systematic reionization of a particular ionic species.     Campylobacter infection of broiler chickens in a free-range environment

    Campylobacter jejuni is the most common cause of bacterial gastroenteritis worldwide, with contaminated chicken meat considered to represent a major source of human infection. Biosecurity measures can reduce C. jejuni shedding rates of housed chickens, but the increasing popularity of free-range and organic meat raises the question of whether the welfare benefits of extensive production are compatible with food safety. The widespread assumption that the free-range environment contaminates extensively reared chickens has not been rigorously tested. A year-long survey of 64 free-range broiler flocks reared on two sites in Oxfordshire, UK, combining high-resolution genotyping with behavioural and environmental observations revealed: (i) no evidence of colonization of succeeding flocks by the C. jejuni genotypes shed by preceding flocks, (ii) a high degree of similarity between C. jejuni genotypes from both farm sites, (iii) no association of ranging behaviour with likelihood of Campylobacter shedding, and (iv) higher genetic differentiation between C. jejuni populations from chickens and wild birds on the same farm than between the chicken samples, human disease isolates from the same region and national samples of C. jejuni from chicken meat

    A multiplex assay for the sensitive detection and quantification of male and female Plasmodium falciparum gametocytes.

    BACKGROUND: The transmission of malaria to mosquitoes depends on the presence of gametocytes that circulate in the peripheral blood of infected human hosts. Sensitive estimates of the densities of female gametocytes (FG) and male gametocytes (MG) may allow the prediction of infectivity to mosquitoes and thus a molecular estimate of the human infectious reservoir for transmission. METHODS: A novel multiplex qRT-PCR assay with intron-spanning primers was developed for the parallel quantification of FG and MG. CCp4 (PF3D7_0903800) transcripts specific for FG and PfMGET (PF3D7_1469900) transcripts specific for MG were quantified in total nucleic acids. The assay was validated on sex-sorted gametocytes from culture material and on samples from clinical trials with gametocytocidal drugs. Synthetic RNA standards were generated for the two targets genes and calibrated against known gametocyte quantities. RESULTS: The limit of detection was determined at 0.1 male and 0.1 female gametocyte/ĀµL, which was equal to the limit of quantification (LOQ) for MG, while the LOQ for FG was 1Ā FG/ĀµL. Results from previously reported clinical trials that used separate gametocyte qRT-PCR assays for FG (targeting Pfs25) and MG (targeting PfMGET) were reproduced with the multiplex assay. High levels of agreement between separate assays and the multiplex approach were observed (R2ā€‰=ā€‰0.9473, 95% CI 0.9314-0.9632, for FG measured by transcript levels of Pfs25 in qRT-PCR or CCp4 in multiplex; R2ā€‰=ā€‰0.8869, 95% CI 0.8541-0.9197, for MG measured by PfMGET in either single or multiplex qRT-PCR). FG and MG transcripts were detected in pure ring stage parasites at 10,000- and 100,000-fold reduced frequency for CCp4 and PfMGET, respectively. The CCp4 and PfMGET transcripts were equally stable under suboptimal storage conditions. CONCLUSIONS: Gametocyte densities and their sex ratios can be determined in the presented one-step multiplex assay with higher throughput than single assays. The interpretation of low gametocyte densities at asexual parasite densities above 1000Ā parasites/ĀµL requires caution to avoid false positive gametocyte signals from spurious transcript levels in ring stage parasites
