    Maternal effects in vulnerability to eye-parasites and correlations between behavior and parasitism in juvenile Arctic charr

    Hatchery-reared fish show high mortalities after release to the wild environment. Explanations for this include potentially predetermined genetics, behavioral, and physiological acclimation to fish farm environments, and increased vulnerability to predation and parasitism in the wild. We studied vulnerability to Diplostomum spp. parasites (load of eye flukes in the lenses), immune defense (relative spleen size) and antipredator behaviors (approaches toward predator odor, freezing, and swimming activity) in hatchery-reared juvenile Arctic charr (Salvelinus alpinus) using a nested mating design. Fish were exposed to eye-fluke larvae via the incoming water at the hatchery. Fish size was positively associated with parasite load, but we did not find any relationship between relative spleen size and parasitism. The offspring of different females showed significant variation in their parasite load within sires, implying a dam effect in the vulnerability to parasites. However, the family background did not have any effect on spleen size. In the mean sire level over dams, the fish from the bolder (actively swimming) families in the predator trials suffered higher loads of eye flukes than those from more cautiously behaving families. Thus, the results indicate potentially maternally inherited differences in vulnerability to eye-fluke parasites, and that the vulnerability to parasites and behavioral activity are positively associated with each other at the sire level. This could lead to artificial and unintentional selection for increased vulnerability to both parasitism and predation if these traits are favored in fish farm environments.Peer reviewe

    Does enriched rearing during early life affect sperm quality or skin colouration in the adult brown trout?

    Enriched rearing has been demonstrated to shape the phenotype of hatchery-reared salmonids and improve their post-release survival in the wild, thus having an important applied value in conservation. However, it is unclear if rearing conditions or survival selection during the early life stages induce long-term fitness effects on adult phenotypes. Using a paired full-sib set-up, we investigated the influence of the environmental enrichment at the egg and fry stages on the milt quality and skin colouration of the adult brown trout (Salmo trutta L.). Overall, males had a higher number of skin spots than females. Notably, the total numbers of spots and black spots were significantly lower in fish raised in an enriched environment than in their full siblings reared in a conventional hatchery environment. However, neither sperm motility nor sperm swimming behaviour differed between full sib males reared in different environments. Our results suggest that rearing method may shape the colouration of brown trout, either by ecological carry-over effects or by selective survival during the rearing process. This, in turn, indicates that ecological conditions at early life can have long-prevailing phenotypically plastic or microevolutionary effects on the adult traits of fish. These effects should be taken into consideration to better understand the ecological role of rearing methodology in salmonid conservation.Peer reviewe

    Donor genetic determinant of thymopoiesis, rs2204985, and stem cell transplantation outcome in a multipopulation cohort

    \ua9 2024 The Author(s)Background: A genetic polymorphism, rs2204985, has been reported to be associated with the diversity of T-cell antigen receptor repertoire and TREC levels, reflecting the function of the thymus. As the thymus function can be assumed to be an important factor regulating the outcome of stem cell transplantation (SCT), it was of great interest that rs2204985 showed a genetic association to disease-free and overall survival in a German SCT donor cohort. Tools to predict the outcome of SCT more accurately would help in risk assessment and patient safety. Objective: To evaluate the general validity of the original genetic association found in the German cohort, we determined genetic associations between rs2204985 and the outcome of SCT in 1,473 SCT donors from four different populations. Study design: Genetic associations between rs2204985 genotype AA versus AG/GG and overall survival (OS) and disease-free survival (DFS) in 1,473 adult, allogeneic SCT from Finland, the United Kingdom, Spain, and Poland were performed using the Kaplan-Meier analysis and log-rank tests. We adjusted the survival models with covariates using Cox regression. Results: In unrelated SCT donors (N = 425), the OS of genotype AA versus AG/GG had a trend for a similar association (p = 0.049, log-rank test) as previously reported in the German cohort. The trend did not remain significant in the Cox regression analysis with covariates. No other associations were found. Conclusion: Weak support for the genetic association between rs2204985, previously also associated with thymus function, and the outcome of SCT could be found in a cohort from four populations

    Unique transcriptional and protein-expression signature in human lung tissue-resident NK cells

    Human lung tissue-resident NK cells (trNK cells) are likely to play an important role in host responses towards viral infections, inflammatory conditions and cancer. However, detailed insights into these cells are still largely lacking. Here we show, using RNA sequencing and flow cytometry-based analyses, that subsets of human lung CD69(-)CD16(-) NK cells display hallmarks of tissue-residency, including high expression of CD49a, CD103, and ZNF683, and reduced expression of SELL, S1PR5, and KLF2/3. CD49a(+)CD16(-) NK cells are functionally competent, and produce IFN-gamma, TNF, MIP-1 beta, and GM-CSF. After stimulation with IL-15, they upregulate perforin, granzyme B, and Ki67 to a similar degree as CD49a(-) CD16(-) NK cells. Comparing datasets from trNK cells in human lung and bone marrow with tissue-resident memory CD8(+) T cells identifies core genes co-regulated either by tissue-residency, cell-type or location. Together, our data indicate that human lung trNK cells have distinct features, likely regulating their function in barrier immunity.Peer reviewe

    The Safety and Efficacy of Live Viral Vaccines in Patients With Cartilage-Hair Hypoplasia

    Background: Live viral vaccines are generally contraindicated in patients with combined immunodeficiency including cartilage-hair hypoplasia (CHH); however, they may be tolerated in milder syndromes. We evaluated the safety and efficacy of live viral vaccines in patients with CHH.Methods: We analyzed hospital and immunization records of 104 patients with CHH and measured serum antibodies to measles, mumps, rubella, and varicella zoster virus (VZV) in all patients who agreed to blood sampling (n= 50). We conducted a clinical trial (identifier: NCT02383797) of live VZV vaccine on five subjects with CHH who lacked varicella history, had no clinical symptoms of immunodeficiency, and were seronegative for VZV; humoral and cellular immunologic responses were assessed post-immunization.Results: A large proportion of patients have been immunized with live viral vaccines, including measles-mumps-rubella (MMR) (n= 40, 38%) and VZV (n= 10, 10%) vaccines, with no serious adverse events. Of the 50 patients tested for antibodies, previous immunization has been documented with MMR (n= 22), rubella (n= 2) and measles (n= 1) vaccines. Patients with CHH demonstrated seropositivity rates of 96%/75%/91% to measles, mumps and rubella, respectively, measured at a medium of 24 years post-immunization. Clinical trial participants developed humoral and cellular responses to VZV vaccine. One trial participant developed post-immunization rash and knee swelling, both resolved without treatment.Conclusion: No serious adverse events have been recorded after immunization with live viral vaccines in Finnish patients with CHH. Patients generate humoral and cellular immune response to live viral vaccines. Immunization with live vaccines may be considered in selected CHH patients with no or clinically mild immunodeficiency

    Modelling of the LTDE-SD radionuclide diffusion experiment in crystalline rock at the Aspo Hard Rock Laboratory (Sweden)

    This study shows a comparison and analysis of results from a modelling exercise concerning a field experiment involving the transport and retention of different radionuclide tracers in crystalline rock. This exercise was performed within the Swedish Nuclear Fuel and Waste Management Company (SKB) Task Force on Modelling of Groundwater Flow and Transport of Solutes (Task Force GWFTS). Task 9B of the Task Force GWFTS was the second subtask within Task 9 and focused on the modelling of experimental results from the Long Term Sorption Diffusion Experiment in situ tracer test. The test had been performed at a depth of about 410m in the Aspo Hard Rock Laboratory. Synthetic groundwater containing a cocktail of radionuclide tracers was circulated for 198 days on the natural surface of a fracture and in a narrow slim hole drilled in unaltered rock matrix. Overcoring of the rock after the end of the test allowed for the measurement of tracer distribution profiles in the rock from the fracture surface (A cores) and also from the slim hole (D cores). The measured tracer activities in the rock samples showed long profiles (several cm) for non-or weakly-sorbing tracers (Cl-36, Na-22), but also for many of the more strongly-sorbing radionuclides. The understanding of this unexpected feature was one of the main motivations for this modelling exercise. However, re-evaluation and revision of the data during the course of Task 9B provided evidence that the anomalous long tails at low activities for strongly sorbing tracers were artefacts due to cross-contamination during rock sample preparation. A few data points remained for Cs-137, Ba-133, Ni-63 and Cd-109, but most measurements at long distances from the tracer source (>10mm) were now below the reported detection limits. Ten different modelling teams provided results for this exercise, using different concepts and codes. The tracers that were finally considered were Na-22, Cl-36, Co-57, Ni-63, Ba-133, Cs-137, Cd-109, Ra-226 and Np-237. Three main types of models were used: i) analytical solutions to the transport-retention equations, ii) continuum -porous-medium numerical models, and iii) microstructure-based models accounting for small-scale heterogeneity (i.e. mineral grains, porosities and/or microfracture distributions) and potential centimetre-scale fractures. The modelling by the different teams led to some important conclusions, concerning for instance the presence of a disturbed zone (a few mm in thickness) next to the fracture surface and to the wall of the slim hole and the role of micro-fractures and cm-scale fractures in the transport of weakly sorbing tracers. These conclusions could be reached after the re-evaluation and revision of the experimental data (tracer profiles in the rock) and the analysis of the different sets of model results provided by the different teams.Peer reviewe

The development of thymus-derived Tregs is known to require high-avidity interaction with MHC-self peptides leading to the generation of self-reactive Tregs fundamental for the maintenance of self-tolerance. Notwithstanding their crucial role in the control of immune responses, human thymic Treg differentiation remains poorly understood. In this mini-review, we will focus on the developmental stages at which Treg lineage commitment occurs, and their spatial localization in the human thymus, reviewing the molecular requirements, including T-cell receptor and cytokine signaling, as well as the cellular interactions involved.     Association between television viewing and the risk of metabolic syndrome in a community-based population

    <p>Abstract</p> <p>Background</p> <p>As a result of metabolic syndrome becoming an important issue during recent decades, many studies have explored the risk factors contributing to its development. However, less attention has been paid to the risk associated with sedentary behavior, especially television viewing. This study examined the association between television viewing time and the risk of having metabolic syndrome in a population of Taiwanese subjects.</p> <p>Methods</p> <p>This community-based cross-sectional study included 2,353 subjects (1,144 men and 1,209 women) aged 40 and over from October, 2004 to September, 2005. Information about the time spent watching TV was obtained using a self-administered questionnaire. The definition of metabolic syndrome was according to the Third Report of the National Cholesterol Education Program's Adult Treatment Panel modified for Asians.</p> <p>Results</p> <p>Compared to subjects who viewed TV < 14 hr/week, those who viewed TV > 20 hr/week had a 1.50-fold (95% confidence intervals (CI): 1.10, 2.03) risk for men and a 1.93-fold (95% CI: 1.37, 2.71) risk for women of having metabolic syndrome, after adjusting for physical activity and other covariates. Stratifying by the three categories of total activity levels, TV viewing time > 20 hr/week was found to still hold a significant risk for having metabolic syndrome in the lowest of the three categories of total activity level for men and in all three categories of total activity level for women.</p> <p>Conclusion</p> <p>The findings suggest that TV viewing is an independent risk factor associated with metabolic syndrome in Taiwanese people.</p
