    Rheology of living cells

    The mechanical behavior of living cells, during planktonic growth, has been thoroughly explored combining common biological techniques with rheology and rheo-imaging measurements. Under a shear flow, bacterial cultures of Staphylococcus aureus revealed a complex and rich rheological behavior not usually accessed in biological studies. In particular, in stationary shear flow, the viscosity increased during the exponential phase and returned close to its initial value at the late phase of growth, accompanied by the stabilization of the bacterial population. In oscillatory flow, the elastic and viscous moduli exhibited power-law behaviors whose exponents are dependent on the bacteria growth stage, and can be associated to a Soft Glassy Material behavior. These behaviors were framed in a microscopic model that suggests the formation of a dynamic web-like structure, where specific aggregation phenomena may occur, depending on growth stage and cell density. Furthermore, systematic measurements combining optical density and dry weight techniques presented new evidences, which confirmed that the observed cell aggregation patterns developed during growth, under shear, can not only be cell density dependent.info:eu-repo/semantics/publishedVersio

    Review: Genomics of interactions between phytoplasmas, host plants and vector insects

     Los fitoplasmas son bacterias de la clase Mollicutes sin pared celular, que se caracterizan por tener un genoma reducido (600-900 Kb) comparado con el de otras bacterias fitopatógenas. Tienen un genoma plástico que les permite multiplicarse exitósamente en dos tipos de hospederos biológicamente distantes, como insectos vectores y plantas. Actualmente se han secuenciado seis genomas completos de fitoplasmas y hay 15 más en proceso de anotación, lo que ha permitido el estudio de los mecanismos de interacción de los fitoplasmas con sus hospederos insectos y plantas. Estas interacciones están gobernadas por proteínas inmunodominantes de membrana de los fitoplasmas como Imp, Amp y Vmp1, que están implicadas en el reconocimiento de moléculas como las cadenas de actina, miosina y la ATP sintasa del intestino y glándulas salivales de los insectos vectores, y con las cadenas de actina de las células de las plantas. También se ha demostrado que los fitoplasmas afectan la idoneidad biológica de los insectos vectores, aumentando o disminuyendo su longevidad y tiempos de oviposición. Por otra parte, las proteínas efectoras como SAP11, SAP54 y TENGU codificadas por los genomas de los fitoplasmas, se encuentran codificadas principalmente en unidades repetitivas del genoma denominadas Unidades Potencialmente Moviles (PMUs), interfieren en las rutas de síntesis de ácido jasmónico y auxinas, lo que afecta la fisiología y arquitectura de las plantas, causando síntomas asociados a fitoplasmas como virescencía y proliferación de brotes vegetativos, entre otros.Palabras Clave: efectores, interacciones moleculare

    Nevoid Basal-cell Syndrome: Literature Review And Case Report In A Family.

    The Nevoid Basal-Cell Carcinoma Syndrome (NBCC), or as it is also referred to, basal-cell nevus syndrome or Gorlin-Goltz syndrome, is characterized by multiple early-appearing basal cell carcinomas, keratocytosis of the mandible, and anomalies of the ocular, skeletal reproductive system. We describe four patients in the same family, all of them possessing a large number of skin tumors associated with other typical clinical and X-Ray anomalies of NBCC. The definitive treatment of NBCC has yet to be established, however, early diagnosis is very important as well as the periodical follow-up examination of ten patients, mainly due to the transformations in the skin lesions that may occur.113391792

    Evaluation of an integrated system for classification, assessment and comparison of services for long-term care in Europe: the eDESDE-LTC study

    The harmonization of European health systems brings with it a need for tools to allow the standardized collection of information about medical care. A common coding system and standards for the description of services are needed to allow local data to be incorporated into evidence-informed policy, and to permit equity and mobility to be assessed. The aim of this project has been to design such a classification and a related tool for the coding of services for Long Term Care (DESDE-LTC), based on the European Service Mapping Schedule (ESMS). Methods. The development of DESDE-LTC followed an iterative process using nominal groups in 6 European countries. 54 researchers and stakeholders in health and social services contributed to this process. In order to classify services, we use the minimal organization unit or "Basic Stable Input of Care" (BSIC), coded by its principal function or "Main Type of Care" (MTC). The evaluation of the tool included an analysis of feasibility, consistency, ontology, inter-rater reliability, Boolean Factor Analysis, and a preliminary impact analysis (screening, scoping and appraisal). Results: DESDE-LTC includes an alpha-numerical coding system, a glossary and an assessment instrument for mapping and counting LTC. It shows high feasibility, consistency, inter-rater reliability and face, content and construct validity. DESDE-LTC is ontologically consistent. It is regarded by experts as useful and relevant for evidence-informed decision making. Conclusion: DESDE-LTC contributes to establishing a common terminology, taxonomy and coding of LTC services in a European context, and a standard procedure for data collection and international comparison

    Genome-Wide Analysis in Brazilian Xavante Indians Reveals Low Degree of Admixture

    Characterization of population genetic variation and structure can be used as tools for research in human genetics and population isolates are of great interest. the aim of the present study was to characterize the genetic structure of Xavante Indians and compare it with other populations. the Xavante, an indigenous population living in Brazilian Central Plateau, is one of the largest native groups in Brazil. A subset of 53 unrelated subjects was selected from the initial sample of 300 Xavante Indians. Using 86,197 markers, Xavante were compared with all populations of HapMap Phase III and HGDP-CEPH projects and with a Southeast Brazilian population sample to establish its population structure. Principal Components Analysis showed that the Xavante Indians are concentrated in the Amerindian axis near other populations of known Amerindian ancestry such as Karitiana, Pima, Surui and Maya and a low degree of genetic admixture was observed. This is consistent with the historical records of bottlenecks experience and cultural isolation. By calculating pair-wise F-st statistics we characterized the genetic differentiation between Xavante Indians and representative populations of the HapMap and from HGDP-CEPH project. We found that the genetic differentiation between Xavante Indians and populations of Ameridian, Asian, European, and African ancestry increased progressively.     Toll-like receptor stimulation induces higher TNF-alpha secretion in peripheral blood mononuclear cells from patients with hyper IgE syndrome

    Hyper IgE syndromes (HIES) are primary immunodeficiency disorders of unknown pathogenesis. Patients are typically affected with `cold' abscesses of the skin, recurrent cyst-forming pneumonia, chronic mucocutaneous candidiasis and other less frequent features such as progressive skeletal abnormalities. Defective signaling in the Toll-like receptor (TLR) pathways has been suggested as a responsible pathologic mechanism, however, in previous reports, 10 patients revealed no defect in inflammatory cytokine responses to different TLR ligands. Here, we report the increase in pro-inflammatory cytokines TNF-alpha and IL-8, following TLR2 and TLR4 stimulation in a larger cohort of 25 additional patients with HIES, and provide a meta-analysis of the TLR data in HIES.     The Parker Instability in 3-D: Corrugations and Superclouds Along the Carina-Sagittarius Arm

    Here we present three-dimensional MHD models for the Parker instability in a thick magnetized disk, including the presence of a spiral arm. The BB-field is assumed parallel to the arm, and the model results are applied to the optical segment of the Carina-Sagittarius arm. The characteristic features of the undular and interchange modes are clearly apparent in the simulations. The undular mode creates large gas concentrations distributed along the arm. This results in a clear arm/inter-arm difference: the instability triggers the formation of large interstellar clouds inside the arms, but generates only small structures with slight density enhancements in the inter-arm regions. The resulting clouds are distributed in an antisymmetric way with respect to the midplane, creating an azimuthal corrugation along the arm. For conditions similar to those of the optical segment of the Carina-Sagittarius arm, it has a wavelength of about 2.4 kpc. This structuring can explain the origin of both HI superclouds and the azimuthal corrugations in spiral arms. The wavelength matches the corrugation length derived with the young stellar groups located in the optical segment of the Carina-Sagittarius arm.     Increased diversity of libraries from libraries: chemoinformatic analysis of bis-diazacyclic libraries

    Combinatorial libraries continue to play a key role in drug discovery. To increase structural diversity, several experimental methods have been developed. However, limited efforts have been performed so far to quantify the diversity of the broadly used diversity-oriented synthetic (DOS) libraries. Herein we report a comprehensive characterization of 15 bis-diazacyclic combinatorial libraries obtained through libraries from libraries, which is a DOS approach. Using MACCS keys, radial and different pharmacophoric fingerprints as well as six molecular properties, it was demonstrated the increased structural and property diversity of the libraries from libraries over the individual libraries. Comparison of the libraries to existing drugs, NCI Diversity and the Molecular Libraries Small Molecule Repository revealed the structural uniqueness of the combinatorial libraries (mean similarity < 0.5 for any fingerprint representation). In particular, bis-cyclic thiourea libraries were the most structurally dissimilar to drugs retaining drug-like character in property space. This study represents the first comprehensive quantification of the diversity of libraries from libraries providing a solid quantitative approach to compare and contrast the diversity of DOS libraries with existing drugs or any other compound collection
