Deep RNA sequencing of muscle tissue reveals absence of viral signatures in dermatomyositis
Objective: To explore a possible connection between active viral infections and manifestation of Dermatomyositis (DM).
Methods: Skeletal muscle biopsies were analyzed from patients diagnosed with juvenile (n=10) and adult (n=12) DM. Adult DM patients harbored autoantibodies against either TIF-1γ (n=7) or MDA5 (n=5). Additionally, we investigated skeletal muscle biopsies from non-diseased controls (NDC, n=5). We used an unbiased high-throughput sequencing (HTS) approach to detect viral sequences. To further increase sequencing depth, a host depletion approach was applied.
Results: In this observational study, no relevant viral sequences were detected either by native sequencing or after host depletion. The absence of detectable viral sequences makes an active viral infection of the muscle tissue unlikely to be the cause of DM in our cohorts.
Discussion: Type I interferons (IFN) play a major role in the pathogenesis of both juvenile and adult dermatomyositis (DM). The IFN response is remarkably conserved between DM subtypes classified by specific autoantibodies. Certain acute viral infections are accompanied by a prominent type I IFN response involving similar downstream mechanisms as in DM. Aiming to elucidate the pathogenesis of DM in skeletal muscle tissue, we used an untargeted high-throughput sequencing and a host depletion approach to detect possible causative viruses
Influence of production variables and starting material on charcoal stable isotopic and molecular characteristics
We present a systematic study on the effect of starting species, gas composition, temperature, particle size and duration of heating upon the molecular and stable isotope composition of high density (mangrove) and low density (pine) wood. In both pine and mangrove, charcoal was depleted in o13C relative to the starting wood by up to 1.6% and 0.8%, respectively. This is attributed predominantly to the progressive loss of isotopically heavier polysaccharides, and kinetic effects of aromatization during heating. However, the pattern of o13C change was dependant upon both starting species and atmosphere, with different structural changes associated with charcoal production from each wood type elucidated by Solid-State o13C Nuclear Magnetic Resonance Spectroscopy. These are particularly evident at lower temperatures, where variation in the oxygen content of the production atmosphere results in differences in the thermal degradation of cellulose and lignin. It is concluded that production of charcoal from separate species in identical conditions, or from a single sample exposed to different production variables, can result in significantly different o13C of the resulting material, relative to the initial wood. These results have implications for the use of charcoal isotope composition to infer past environmental change
Sustainable Retailing – Influencing Consumer Behaviour on Food Waste
The aim of this research was to examine the influence of a UK national retailer on its customers' food waste behaviour. Using six communication channels (in‐store magazine, e‐newsletter, Facebook site, product stickers and in‐store demonstrations), Asda presented standard food waste reduction messages to its customers during two time limited periods in 2014 and 2015. Six national surveys over 21 months tracked customers' self‐reported food waste. Our results showed that the combined communication channels and repeated messages over time had a significant effect on reducing food waste of customers. Surprisingly, customers who said they did not recall seeing the messages also reduced their food waste, showing the wider influence of interventions. Those who saw a food waste reduction message saved an estimated £81 annually from reducing food waste. The main conclusion of this paper is that retailers can influence the pro‐environmental behaviour of customers using conventional communication channels; however, repeat messages are needed in order to have a long‐term impact
A tiered approach to the use of alternatives to animal testing for the safety assessment of cosmetics: Eye irritation
AbstractThe need for alternative approaches to replace the in vivo rabbit Draize eye test for evaluation of eye irritation of cosmetic ingredients has been recognised by the cosmetics industry for many years. Extensive research has lead to the development of several assays, some of which have undergone formal validation. Even though, to date, no single in vitro assay has been validated as a full replacement for the rabbit Draize eye test, organotypic assays are accepted for specific and limited regulatory purposes. Although not formally validated, several other in vitro models have been used for over a decade by the cosmetics industry as valuable tools in a weight of evidence approach for the safety assessment of ingredients and finished products. In light of the deadlines established in the EU Cosmetics Directive for cessation of animal testing for cosmetic ingredients, a COLIPA scientific meeting was held in Brussels on 30th January, 2008 to review the use of alternative approaches and to set up a decision-tree approach for their integration into tiered testing strategies for hazard and safety assessment of cosmetic ingredients and their use in products. Furthermore, recommendations are given on how remaining data gaps and research needs can be addressed
Luminosities and mass-loss rates of SMC and LMC AGB stars and Red Supergiants
(Abridged) Dust radiative transfer models are presented for 101 carbon stars
and 86 oxygen-rich evolved stars in the Magellanic Clouds for which 5-35 \mum\
{\it Spitzer} IRS spectra are available. The spectra are complemented with
available optical and infrared photometry to construct the spectral energy
distribution. A minimisation procedure is used to fit luminosity, mass-loss
rate and dust temperature at the inner radius. Different effective temperatures
and dust content are also considered. Periods from the literature and from new
OGLE-III data are compiled and derived. The O-rich stars are classified in
foreground objects, AGB stars and Red Super Giants.
For the O-rich stars silicates based on laboratory optical constants are
compared to "astronomical silicates". Overall, the grain type by Volk & Kwok
(1988) fit the data best. However, the fit based on laboratory optical
constants for the grains can be improved by abandoning the small-particle
limit. The influence of grain size, core-mantle grains and porosity are
explored.
Relations between mass-loss rates and luminosity and pulsation period are
presented and compared to the predictions of evolutionary models, those by
Vassiliadis & Wood (1993) and their adopted mass-loss recipe, and those based
on a Reimers mass-loss law with a scaling of a factor of five. The Vassiliadis
& Wood models describe the data better, although there are also some
deficiencies, in particular to the maximum adopted mass-loss rate.
The OGLE-III data reveal an O-rich star in the SMC with a period of 1749
days. Its absolute magnitude of makes it a good candidate
for a super-AGB star.Comment: A&A accepte
On the Carriers of the 21 Micron Emission Feature in Post-Asymptotic Giant Branch Stars
The mysterious 21micron emission feature seen in sixteen C-rich
proto-planetary nebulae (PPNe) remains unidentified since its discovery in
1989. Over a dozen of materials are suggested as the carrier candidates. In
this work we quantitatively investigate eight inorganic and one organic carrier
candidates in terms of elemental abundance constraints, while previous studies
mostly focus on their spectral profiles (which could be largely affected by
grain size, shape and clustering effects). It is found that: (1) five
candidates (TiC nanoclusters, fullerenes coordinated with Ti atoms, SiS,
doped-SiC, and SiO-coated SiC dust) violate the abundance constraints (i.e.
they require too much Ti, S or Si to account for the emission power of the
21micron band, (2) three candidates (carbon and silicon mixtures, FeO,
and FeO),while satisfying the abundance constraints, exhibit secondary
features which are not detected in the 21micron sources, and (3) nano FeO,
neither exceeding the abundance budget nor producing undetected secondary
features, seems to be a viable candidate, supporting the suggestions of Posch

Cortical depth dependent functional responses in humans at 7T: improved specificity with 3D GRASE
Ultra high fields (7T and above) allow functional imaging with high contrast-to-noise ratios and improved spatial resolution. This, along with improved hardware and imaging techniques, allow investigating columnar and laminar functional responses. Using gradient-echo (GE) (T2* weighted) based sequences, layer specific responses have been recorded from human (and animal) primary visual areas. However, their increased sensitivity to large surface veins potentially clouds detecting and interpreting layer specific responses. Conversely, spin-echo (SE) (T2 weighted) sequences are less sensitive to large veins and have been used to map cortical columns in humans. T2 weighted 3D GRASE with inner volume selection provides high isotropic resolution over extended volumes, overcoming some of the many technical limitations of conventional 2D SE-EPI, whereby making layer specific investigations feasible. Further, the demonstration of columnar level specificity with 3D GRASE, despite contributions from both stimulated echoes and conventional T2 contrast, has made it an attractive alternative over 2D SE-EPI. Here, we assess the spatial specificity of cortical depth dependent 3D GRASE functional responses in human V1 and hMT by comparing it to GE responses. In doing so we demonstrate that 3D GRASE is less sensitive to contributions from large veins in superficial layers, while showing increased specificity (functional tuning) throughout the cortex compared to GE
