    Effect of claw horn lesion type and severity at the time of treatment on outcome of lameness in dairy cows

    Prompt diagnosis and treatment of claw horn lesions in cattle affects the likelihood of recovery; however, it is unknown if the type of lesion influences the likelihood of recovery. The aim of this study was to investigate whether the type, severity and frequency of claw horn lesions in newly lame cows (lame for no more than 2 weeks) at the time of corrective foot trimming affects the probability of recovery from lameness after treatment. The images of 112 feet (224 claws) from newly lame cows (n = 112; lame in only one hind foot), which were treated with a standardised therapeutic hoof trim only, were used to score claw horn lesions (sole ulcer, sole haemorrhage, white line haemorrhage or white line separation). Most cows (n = 107/112; 95.5%) were classified as mildly lame at the time of treatment. The proportion of cows that recovered 2 weeks after therapeutic hoof trimming was 88/112 (78.6%). Results of a multilevel logistic regression model indicated that severely lame cows were less likely to recover than those that were mildly lame (odds ratio, OR, 0.16; P = 0.04). White line haemorrhage had a significant negative impact on the likelihood of recovery from lameness (OR 0.14; P > 0.01); however, recovery of cows with white line haemorrhage was positively associated with the length of the lesion (OR 1.05; P = 0.03). This latter finding may be associated with the severity of the lesion, since mild claw horn lesions affected a significantly larger area of the claw than more severe lesions.     Assessing the feasibility of retrospective and prospective clinical audit in farm animal veterinary practice

    As a quality improvement tool, clinical audit has been extensively described in the medical literature. There is scant literature on the use of clinical audit in the farm animal veterinary setting. This study describes the process and feasibility of prospective and retrospective data collection for farm animal clinical audit performed at three different farm animal practices in the United Kingdom. Retrospective clinical audit was difficult in all three practices due to barriers in establishing diagnosis and patient identity from clinical records. Prospective data collection proved feasible but depended on adequate case accrual and practitioner engagement. The decision to conduct clinical audit retrospectively or prospectively will depend on the topic to audit, the availability of data and the wishes of the team members involved. Further work is required looking at the use of clinical coding and alternatives to using practice management software to improve retrospective data availability for clinical audit

    Behavioural changes in dairy cows with lameness in an automatic milking system

    There is a tendency worldwide for the automation of farms; this has included the introduction of automatic milking systems (AMS) in the dairy industry. Lameness in dairy cows is highly prevalent and painful. These impacts potentially affect not only animal welfare, but also farm economies. Three independent observational studies were carried out to assess the impact of lameness on the behaviour of zero grazed high yielding Holstein cows managed in an AMS. The aim of the first study was to examine the impact of lameness on rumination time, the second study investigated differences between lame and sound dairy cows in total eating time and the third study assessed the impact of lameness on milking behaviour (frequency and time of visits to the AMS). In the first study data from 150 cows were used to analyse rumination (collected using rumination collars) for the 48hr following locomotion scoring. A multilevel linear regression demonstrated that lameness had a small but significant negative association (coefficient: -7.88 (SE: 3.93)) with rumination. In the second study the behaviour of eleven matched lame and sound pairs of cows at the feed face was analysed for 24 hours after locomotion scoring. Each feeding behaviour variable (total duration time, frequency of feeding bouts and length of bouts) was analysed using individual single level regression models. There was a significant negative association between total feeding time and lameness (coefficient: -73.65 (SE: 25.47)) and the frequency of feeding bouts and lameness (-9.93 (2.49)). Finally, the third observational study used 38 matched pairs of lame and sound cows. Data on the number and timings of visits to the AMS were collected for 24 hours after each locomotion score and analysed using a binomial logistic regression model. There was a significant difference in AMS visits between groups; lame animals visiting the robot less frequently than sound cows (median difference 0.50 milking visits; T = 256.0; N = 25; p = 0.01) and lame cows were 0.33 times less likely to visit the AMS between 24:01 and 06:00. Results from these studies reveal that lameness in an AMS affected feeding behaviour, rumination and AMS visits. All of these impacts are likely to have negative consequences for farm profitability, but also implications for the health and welfare of the animals

    Using simulation to interpret a discrete time survival model in a complex biological system: fertility and lameness in dairy cows

    The ever-growing volume of data routinely collected and stored in everyday life presents researchers with a number of opportunities to gain insight and make predictions. This study aimed to demonstrate the usefulness in a specific clinical context of a simulation-based technique called probabilistic sensitivity analysis (PSA) in interpreting the results of a discrete time survival model based on a large dataset of routinely collected dairy herd management data. Data from 12,515 dairy cows (from 39 herds) were used to construct a multilevel discrete time survival model in which the outcome was the probability of a cow becoming pregnant during a given two day period of risk, and presence or absence of a recorded lameness event during various time frames relative to the risk period amongst the potential explanatory variables. A separate simulation model was then constructed to evaluate the wider clinical implications of the model results (i.e. the potential for a herd’s incidence rate of lameness to influence its overall reproductive performance) using PSA. Although the discrete time survival analysis revealed some relatively large associations between lameness events and risk of pregnancy (for example, occurrence of a lameness case within 14 days of a risk period was associated with a 25% reduction in the risk of the cow becoming pregnant during that risk period), PSA revealed that, when viewed in the context of a realistic clinical situation, a herd’s lameness incidence rate is highly unlikely to influence its overall reproductive performance to a meaningful extent in the vast majority of situations. Construction of a simulation model within a PSA framework proved to be a very useful additional step to aid contextualisation of the results from a discrete time survival model, especially where the research is designed to guide on-farm management decisions at population (i.e. herd) rather than individual level

    A randomised controlled trial to evaluate the impact of indoor living space on dairy cow production, reproduction and behaviour

    As a global society, we have a duty to provide suitable care and conditions for farmed livestock to protect animal welfare and ensure the sustainability of our food supply. The suitability and biological impacts of housing conditions for intensively farmed animals is a complex and emotive subject, yet poorly researched, meaning quantitative evidence to inform policy and legislation is lacking. Most dairy cows globally are housed for some duration during the year, largely when climatic conditions are unfavourable. However, the impact on biology, productivity and welfare of even the most basic housing requirement, the quantity of living space, remains unknown. We conducted a long-term (1-year), randomised controlled trial (CONSORT 10 guidelines) to investigate the impact of increased living space (6.5m2 vs 3m2 per animal) on critical aspects of cow biology, behaviour and productivity. Adult Holstein dairy cows (n = 150) were continuously and randomly allocated to a high or control living space group with all other aspects of housing remaining identical between groups. Compared to cows in the control living space group, cows with increased space produced more milk per 305d lactation (primiparous: 12,235L vs 11,592L, P < 0.01; multiparous: 14,746L vs 14,644L, P < 0.01) but took longer to become pregnant after calving (primiparous: 155d vs 83d, P = 0.025; multiparous: 133d vs 109d). In terms of behaviour, cows with more living space spent significantly more time in lying areas (65min/d difference; high space group: 12.43h/day, 95% CI = 11.70-13.29; control space group: 11.42h/day, 95% CI = 10.73-12.12) and significantly less time in passageways (64min/d), suggesting enhanced welfare when more space was provided. A key physiological difference between groups was that cows with more space spent longer ruminating each day. This is the first long term study in dairy cows to demonstrate that increased living space results in meaningful benefits in terms of productivity and behaviour and suggests that the interplay between farmed animals and their housed environment plays an important role in the concepts of welfare and sustainability of dairy farming

    Effects of lameness treatment for claw horn lesions on lying behaviour in dairy cows

    Lameness affects lying behaviour in dairy cattle, increasing total lying time and the number of lying bouts. However, there is limited information about the effect of lameness treatment on dairy cow behaviour. This study investigated the effect of four lameness treatments on lying behaviour (total lying time, number of bouts, average bout duration and laterality of lying) in dairy cows. Forty-four newly lame cows were treated randomly with one of four treatment protocols: trim only, trim + block, trim + NSAID, and trim + block + NSAID. Thirty-four non-lame control cows were matched by parity, days in milk, and farm-pen. Each cow had an accelerometer attached to the hind leg, lying behaviour data was collected over 5 days immediately after treatment and analysed using multilevel regression models. Lame cows in three of the four treatment groups demonstrated no increase in lying time compared to non-lame controls. This finding is contrary to previous work and may reflect the enrolment criteria which favoured the selection of cows with mild disease i.e. before the behavioural impacts of lameness had manifested. Only cows in the treatment group which received a therapeutic trim and a foot block saw higher lying times post treatment. As this effect was not apparent in the group which received a NSAID in addition to a trim and a foot block, we hypothesise that this effect is caused by discomfort associated with the block. Where foot blocks are administered as part of treatment protocols, we propose that NSAIDs should be administered concurrently to alleviate the behavioural changes and likely discomfort associated with this treatment

    Factors affecting phage D29 infection: a tool to investigate different growth states of mycobacteria

    Bacteriophages D29 and TM4 are able to infect a wide range of mycobacteria, including pathogenic and non pathogenic species. Successful phage infection of both fast- and slow-growing mycobacteria can be rapidly detected using the phage amplification assay. Using this method, the effect of oxygen limitation during culture of mycobacteria on the success of phage infection was studied. Both D29 and TM4 were able to infect cultures of M. smegmatis and Mycobacterium avium subspecies paratuberculosis (MAP) grown in liquid with aeration. However when cultures were grown under oxygen limiting conditions, only TM4 could productively infect the cells. Cell attachment assays showed that D29 could bind to the cells surface but did not complete the lytic cycle. The ability of D29 to productively infect the cells was rapidly recovered (within 1 day) when the cultures were returned to an aerobic environment and this recovery required de novo RNA synthesis. These results indicated that under oxygen limiting conditions the cells are entering a growth state which inhibits phage D29 replication, and this change in host cell biology which can be detected by using both phage D29 and TM4 in the phage amplification assay

    Evaluation of the limitations and methods to improve rapid phage-based detection of viable Mycobacterium avium subsp. paratuberculosis in the blood of experimentally infected cattle

    Background Disseminated infection and bacteraemia is an underreported and under-researched aspect of Johne’s disease. This is mainly due to the time it takes for Mycobacterium avium subsp. paratuberculosis (MAP) to grow and lack of sensitivity of culture. Viable MAP cells can be detected in the blood of cattle suffering from Johne’s disease within 48 h using peptide-mediated magnetic separation (PMMS) followed by bacteriophage amplification. The aim of this study was to demonstrate the first detection of MAP in the blood of experimentally exposed cattle using the PMMS-bacteriophage assay and to compare these results with the immune response of the animal based on serum ELISA and shedding of MAP by faecal culture. Results Using the PMMS-phage assay, seven out of the 19 (37 %) MAP-exposed animals that were tested were positive for viable MAP cells although very low numbers of MAP were detected. Two of these animals were positive by faecal culture and one was positive by serum ELISA. There was no correlation between PMMS-phage assay results and the faecal and serum ELISA results. None of the control animals (10) were positive for MAP using any of the four detection methods. Investigations carried out into the efficiency of the assay; found that the PMMS step was the limiting factor reducing the sensitivity of the phage assay. A modified method using the phage assay directly on isolated peripheral blood mononuclear cells (without PMMS) was found to be superior to the PMMS isolation step. Conclusions This proof of concept study has shown that viable MAP cells are present in the blood of MAP-exposed cattle prior to the onset of clinical signs. Although only one time point was tested, the ability to detect viable MAP in the blood of subclinically infected animals by the rapid phage-based method has the potential to increase the understanding of the pathogenesis of Johne’s disease progression by warranting further research on the presence of MAP in blood
