    Lipidomic Evaluation of Feline Neurologic Disease after AAV Gene Therapy

    GM1 gangliosidosis is a fatal lysosomal disorder, for which there is no effective treatment. Adeno-associated virus (AAV) gene therapy in GM1 cats has resulted in a greater than 6-fold increase in lifespan, with many cats remaining alive at \u3e 5.7 years of age, with minimal clinical signs. Glycolipids are the principal storage product in GM1 gangliosidosis whose pathogenic mechanism is not completely understood. Targeted lipidomics analysis was performed to better define disease mechanisms and identify markers of disease progression for upcoming clinical trials in humans. 36 sphingolipids and subspecies associated with ganglioside biosynthesis were tested in the cerebrospinal fluid of untreated GM1 cats at a humane endpoint ( approximately 8 months), AAV-treated GM1 cats ( approximately 5 years old), and normal adult controls. In untreated GM1 cats, significant alterations were noted in 16 sphingolipid species, including gangliosides (GM1 and GM3), lactosylceramides, ceramides, sphingomyelins, monohexosylceramides, and sulfatides. Variable degrees of correction in many lipid metabolites reflected the efficacy of AAV gene therapy. Sphingolipid levels were highly predictive of neurologic disease progression, with 11 metabolites having a coefficient of determination (R(2)) \u3e 0.75. Also, a specific detergent additive significantly increased the recovery of certain lipid species in cerebrospinal fluid samples. This report demonstrates the methodology and utility of targeted lipidomics to examine the pathophysiology of lipid storage disorders

    Genome-wide association study identifies six new loci influencing pulse pressure and mean arterial pressure.

    Numerous genetic loci have been associated with systolic blood pressure (SBP) and diastolic blood pressure (DBP) in Europeans. We now report genome-wide association studies of pulse pressure (PP) and mean arterial pressure (MAP). In discovery (N = 74,064) and follow-up studies (N = 48,607), we identified at genome-wide significance (P = 2.7 × 10(-8) to P = 2.3 × 10(-13)) four new PP loci (at 4q12 near CHIC2, 7q22.3 near PIK3CG, 8q24.12 in NOV and 11q24.3 near ADAMTS8), two new MAP loci (3p21.31 in MAP4 and 10q25.3 near ADRB1) and one locus associated with both of these traits (2q24.3 near FIGN) that has also recently been associated with SBP in east Asians. For three of the new PP loci, the estimated effect for SBP was opposite of that for DBP, in contrast to the majority of common SBP- and DBP-associated variants, which show concordant effects on both traits. These findings suggest new genetic pathways underlying blood pressure variation, some of which may differentially influence SBP and DBP

    Search for a right-handed W boson and a heavy neutrino in proton-proton collisions at s \sqrt{s} = 13 TeV

    A search is presented for a right-handed W boson (WR) and a heavy neutrino (N), in a final state consisting of two same-flavor leptons (ee or μμ) and two quarks. The search is performed with the CMS experiment at the CERN LHC using a data sample of proton-proton collisions at a center-of-mass energy of 13 TeV corresponding to an integrated luminosity of 138 fb−1. The search covers two regions of phase space, one where the decay products of the heavy neutrino are merged into a single large-area jet, and one where the decay products are well separated. The expected signal is characterized by an excess in the invariant mass distribution of the final-state objects. No significant excess over the standard model background expectations is observed. The observations are interpreted as upper limits on the product of WR production cross sections and branching fractions assuming that couplings are identical to those of the standard model W boson. For N masses mN equal to half the WR mass mWR (mN = 0.2 TeV), mWR is excluded at 95% confidence level up to 4.7 (4.8) and 5.0 (5.4) TeV for the electron and muon channels, respectively. This analysis provides the most stringent limits on the WR mass to date

    Search for flavor-changing neutral current interactions of the top quark and the Higgs boson decaying to a bottom quark-antiquark pair at s√ = 13 TeV

    A search for flavor-changing neutral current interactions of the top quark (t) and the Higgs boson (H) is presented. The search is based on a data sample corresponding to an integrated luminosity of 137 fb−1 recorded by the CMS experiment at the LHC in proton-proton collisions at s√ = 13 TeV. Events containing exactly one lepton (muon or electron) and at least three jets, among which at least two are identified as originating from the hadronization of a bottom quark, are analyzed. A set of deep neural networks is used for kinematic event reconstruction, while boosted decision trees distinguish the signal from the background events. No significant excess over the background predictions is observed, and upper limits on the signal production cross sections are extracted. These limits are interpreted in terms of top quark decay branching fractions (B ) to the Higgs boson and an up (u) or a charm quark (c). Assuming one nonvanishing extra coupling at a time, the observed (expected) upper limits at 95% confidence level are B (t → Hu) < 0.079 (0.11)% and B (t → Hc) < 0.094 (0.086)%

    Inclusive and differential cross section measurements of single top quark production in association with a Z boson in proton-proton collisions at √s = 13 TeV

    Inclusive and differential cross sections of single top quark production in association with a Z boson are measured in proton-proton collisions at a center-of-mass energy of 13 TeV with a data sample corresponding to an integrated luminosity of 138 fb−1 recorded by the CMS experiment. Events are selected based on the presence of three leptons, electrons or muons, associated with leptonic Z boson and top quark decays. The measurement yields an inclusive cross section of 87.9−7.3+7.5(stat)−6.0+7.3(syst) fb for a dilepton invariant mass greater than 30 GeV, in agreement with standard model (SM) calculations and represents the most precise determination to date. The ratio between the cross sections for the top quark and the top antiquark production in association with a Z boson is measured as 2.37−0.42+0.56(stat)−0.13+0.27(syst). Differential measurements at parton and particle levels are performed for the first time. Several kinematic observables are considered to study the modeling of the process. Results are compared to theoretical predictions with different assumptions on the source of the initial-state b quark and found to be in agreement, within the uncertainties. Additionally, the spin asymmetry, which is sensitive to the top quark polarization, is determined from the differential distribution of the polarization angle at parton level to be 0.54 ± 0.16 (stat) ± 0.06 (syst), in agreement with SM predictions.     Combined searches for the production of supersymmetric top quark partners in proton–proton collisions at √s=13Te

    A combination of searches for top squark pair production using proton–proton collision data at a center-of-mass energy of 13TeV at the CERN LHC, corresponding to an integrated luminosity of 137fb1^{-1} collected by the CMS experiment, is presented. Signatures with at least 2 jets and large missing transverse momentum are categorized into events with 0, 1, or 2 leptons. New results for regions of parameter space where the kinematical properties of top squark pair production and top quark pair production are very similar are presented. Depending on the model, the combined result excludes a top squark mass up to 1325GeV for a massless neutralino, and a neutralino mass up to 700GeV for a top squark mass of 1150GeV. Top squarks with masses from 145 to 295GeV, for neutralino masses from 0 to 100GeV, with a mass difference between the top squark and the neutralino in a window of 30GeV around the mass of the top quark, are excluded for the first time with CMS data. The results of theses searches are also interpreted in an alternative signal model of dark matter production via a spin-0 mediator in association with a top quark pair. Upper limits are set on the cross section for mediator particle masses of up to 420GeV

    Observation of the Bc+_\mathrm{c}^+ Meson in Pb-Pb and pp Collisions at sNN\sqrt{s_{\mathrm{NN}}} = 5.02 TeV and Measurement of its Nuclear Modification Factor

    The Bc+_\mathrm{c}^+ meson is observed for the first time in heavy ion collisions. Data from the CMS detector are used to study the production of the Bc+_\mathrm{c}^+ meson in lead-lead (Pb-Pb) and proton-proton (pp) collisions at a center-of-mass energy per nucleon pair of sNN\sqrt{s_{\mathrm{NN}}} = 5.02 TeV , via the Bc+_\mathrm{c}^+ → (J/ψ → μ+^+μ^−+^+νμ decay. The Bc+_\mathrm{c}^+ nuclear modification factor, derived from the PbPb-to-pp ratio of production cross sections, is measured in two bins of the trimuon transverse momentum and of the PbPb collision centrality. The Bc+_\mathrm{c}^+meson is shown to be less suppressed than quarkonia and most of the open heavy-flavor mesons, suggesting that effects of the hot and dense nuclear matter created in heavy ion collisions contribute to its production. This measurement sets forth a promising new probe of the interplay of suppression and enhancement mechanisms in the production of heavy-flavor mesons in the quark-gluon plasma

    Measurement of the production cross section for Z + b jets in proton-proton collisions at s\sqrt{s} = 13 TeV

    The measurement of the cross section for the production of a Z boson, decaying to dielectrons or dimuons, in association with at least one bottom quark jet is performed with proton-proton collision data at √s=13  TeV. The data sample corresponds to an integrated luminosity of 137  fb1^{-1}, collected by the CMS experiment at the LHC during 2016–2018. The integrated cross sections for Z+≥1 b jet and Z+≥2 b jets are reported for the electron, muon, and combined channels. The fiducial cross sections in the combined channel are 6.52±0.04(stat)±0.40(syst)±0.14(theo)  pb for Z+≥1 b jet and 0.65±0.03(stat)±0.07(syst)±0.02(theo)  pb for Z+≥2 b jets. The differential cross section distributions are measured as functions of various kinematic observables that are useful for precision tests of perturbative quantum chromodynamics predictions. The ratios of integrated and differential cross sections for Z+≥2b  jets and Z+≥1 b jet processes are also determined. The value of the integrated cross section ratio measured in the combined channel is 0.100±0.005(stat)±0.007(syst)±0.003(theo). All measurements are compared with predictions from various event generators

    Search for heavy resonances decaying to ZZ or ZW and axion-like particles mediating nonresonant ZZ or ZH production at S\sqrt{S} = 13 TeV

    A search has been performed for heavy resonances decaying to ZZ or ZW and for axion-like particles (ALPs) mediating nonresonant ZZ or ZH production, in final states with two charged leptons (ℓ = e, μ) produced by the decay of a Z boson, and two quarks produced by the decay of a Z, W, or Higgs boson H. The analysis is sensitive to resonances with masses in the range 450 to 2000 GeV. Two categories are defined corresponding to the merged or resolved reconstruction of the hadronically decaying boson. The search is based on data collected during 2016–2018 by the CMS experiment at the LHC in proton-proton collisions at a center-of-mass energy of 13 TeV, corresponding to an integrated luminosity of 138 fb−1. No significant excess is observed in the data above the standard model background expectation. Upper limits on the production cross section of heavy, narrow spin-2 and spin-1 resonances are derived as functions of the resonance mass, and exclusion limits on the production of bulk graviton particles and W′ bosons are calculated in the framework of the warped extra dimensions and heavy vector triplet models, respectively. In addition, upper limits on the ALP-mediated diboson production cross section and ALP couplings to standard model particles are obtained in the framework of linear and chiral effective field theories. These are the first limits on nonresonant ALP-mediated ZZ and ZH production obtained by the LHC experiments
