SummaryThe active site of recombinant hexa-histidine-tagged human monoacylglycerol lipase (hMGL) is characterized by mass spectrometry using the inhibitors 5-((biphenyl-4-yl)methyl)-N,N-dimethyl-2H-tetrazole-2-carboxamide (AM6701), and N-arachidonylmaleimide (NAM) as probes. Carbamylation of Ser129 by AM6701 in the putative hMGL catalytic triad demonstrates this residue's essential role in catalysis. Partial NAM alkylation of hMGL cysteine residues 215 and/or 249 was sufficient to achieve ∼80% enzyme inhibition. Although Cys215 and/or Cys249 mutations to alanine(s) did not affect hMGL hydrolytic activity as compared with nonmutated hMGL, the C215A displayed heightened NAM sensitivity, whereas the C249A evidenced reduced NAM sensitivity. These data conclusively demonstrate a sulfhydryl-based mechanism for NAM inhibition of hMGL in which Cys249 is of paramount importance. Identification of amino acids critical to the catalytic activity and pharmacological modulation of hMGL informs the design of selective MGL inhibitors as potential drugs

The results obtained after evaluating attitudes towards the inclusion of students with special educational needs in ordinary classrooms are presented. In this preliminary study the sample is composed of 91 participants, all of them students of the Master's degree. , of which 22% are men and 78% are women, aged between 22 and 45 years (M = 28.13, Dt = 6.4). The Opinion Questionnaire was applied to Integration, which consists of 30 items with a Likert type response format of 5 points and higher scores denoting more favorable attitudes. The results indicated that women and those who have contact with the attitudinal reference consider that the teaching staff is better prepared to face the challenges of inclusion. However, there does not seem to be a linear relationship between contact frequency and attitudes toward integration, so it is necessary to deepen the type of contact and the types of educational needs with which you are familiar, in order to determine the weight of these factors in the formation of more or less favorable attitudes. The present findings as well as the continuation of this study will allow us to establish guidelines for intervention with teachers in training, who will be key elements in the promotion of inclusion in the classrooms of the future.

In our society, employment represents one of the most important aspects that condition our life and relationships and even our health. Work is one of the fundamental activities of life. However, access to it is conditioned by a series of personal and contextual variables that, in interaction, become barriers to participation in society. A traditionally excluded group is composed of people with disabilities. The objective of this work was to: 1) Evaluate the perceptions of workers with intellectual disabilities about their quality of work life; 2) Know the main attitudes, barriers and opportunities that favor and / or hinder employment in these workers; and 3) Identify areas of intervention to improve the quality of work life in workers and organizations. To this end, the opinions expressed by 12 workers with intellectual disabilities who currently worked in Supported Employment (ECA) using qualitative methodologies through the focus group procedure and through the analysis of textual fragments were analyzed. The results obtained showed that the quality of working life is a multidimensional construct formed by objective and subjective indicators, and that the attitudes, barriers and opportunities perceived by the workers impact on their well-being. Finally, strategies are proposed to improve the services oriented towards the standardized employment of people with intellectual disabilities.

The Effect of Antioxidant Supplementation on Fatigue during Exercise: Potential Role for NAD+(H)

This study compared serum pyridine levels (NAD+ /NADH) in trained (n = 6) and untrained (n = 7) subjects after continuous progressive exercise at 50%, 70% then 95% of physical work capacity until fatigue (TTF) after consuming a placebo or antioxidant (AO) cocktail (Lactaway©). An increase of 17% in TTF was observed in AO as compared to placebo (p = 0.032). This was accompanied by a significant increase in serum NAD+ levels (p = 0.037) in the AO supplemented group post exercise. The increases in NAD+ and improved endurance reflect lower oxidative stress-induced suppression of aerobic respiration

Background: Cactus pear (Opuntia ficus-indica) fruit contains vitamin C and characteristic betalain pigments, the radical-scavenging properties and antioxidant activities of which have been shown in vitro. Objective: We investigated the effects of short-term supplementation with cactus pear fruit compared with vitamin C alone on total-body oxidative status in healthy humans. Design: In a randomized, crossover, double-treatment study, 18 healthy volunteers received either 250 g fresh fruit pulp or 75 mg vitamin C twice daily for 2 wk, with a 6-wk washout period between the treatments. Before (baseline) and after each treatment, 8-epiprostaglandin F2α (8-epi-PGF2α) and malondialdehyde in plasma, the ratio of reduced to oxidized glutathione (GSH:GSSG) in erythrocytes, and lipid hydroperoxides in LDL were measured as biomarkers of oxidative stress; plasma Trolox-equivalent antioxidant activity (TEAC) and vitamins A, E, and C were evaluated as indexes of antioxidant status. Results: Both treatments caused comparable increases compared with baseline in plasma concentrations of vitamin E and vitamin C (P < 0.05); vitamin A and TEAC did not change significantly. After supplementation with cactus pear fruit, 8-epi-PGF2α and malondialdehyde decreased by ≈30% and 75%, respectively; GSH:GSSG shifted toward a higher value (P < 0.05); and LDL hydroperoxides were reduced by almost one-half. Supplementation with vitamin C did not significantly affect any marker of oxidative stress. Conclusions: Consumption of cactus pear fruit positively affects the body's redox balance, decreases oxidative damage to lipids, and improves antioxidant status in healthy humans. Supplementation with vitamin C at a comparable dosage enhances overall antioxidant defense but does not significantly affect body oxidative stress. Components of cactus pear fruit other than antioxidant vitamins may play a role in the observed effects. © 2004 American Society for Clinical Nutrition

Lutein decreases oxidative stress and inflammation in liver and eyes of guinea pigs fed a hypercholesterolemic diet

Guinea pigs were fed a hypercholesterolemic diet (0.25 g/100 g cholesterol) and randomly allocated either to a Control group (n = 9) or to a Lutein (0.1 g/100 g) group (n = 10) for 12 weeks to evaluate oxidative stress and inflammation in both liver and eyes. Malondialdehyde (MDA) concentrations and inflammatory cytokines were measured as well as hepatic nuclear factor-kappaB (NF-κB) binding. Lutein concentrations were greater in eyes (P < 0.01) and liver (P < 0.001) in the Lutein group. All guinea pigs had high concentrations of hepatic cholesterol as well as high plasma ALT and AST levels indicative of liver injury. However, the Lutein group had 43% lower hepatic free cholesterol than the Controls (P < 0.05). Hepatic MDA and MDA in the eye were lower in the Lutein compared to the Control group (P < 0.05). Hepatic tumor necrosis factor-α was 32% lower in the Lutein group (P < 0.05). Lastly, the Lutein group presented lower NF-κB DNA binding activity than the Control group (P < 0.001). These results suggest that in the presence of high cholesterol, lutein exerts both antioxidant and anti-inflammatory effects, which can be explained by attenuated NF-κB DNA binding activity. Furthermore, results also suggest that lutein accumulates in the eyes of guinea pigs to protect against oxidative stress
