Probing Dark Matter
Recent novel observations have probed the baryonic fraction of the galactic
dark matter that has eluded astronomers for decades. Late in 1993, the MACHO
and EROS collaborations announced in this journal the detection of transient
and achromatic brightenings of a handful of stars in the Large Magellanic Cloud
that are best interpreted as gravitational microlensing by low-mass foreground
objects (MACHOS). This tantalized astronomers, for it implied that the
population of cool, compact objects these lenses represent could be the elusive
dark matter of our galactic halo. A year later in 1994, Sackett et al. reported
the discovery of a red halo in the galaxy NGC 5907 that seems to follow the
inferred radial distribution of its dark matter. This suggested that dwarf
stars could constitute its missing component. Since NGC 5907 is similar to the
Milky Way in type and radius, some surmised that the solution of the galactic
dark matter problem was an abundance of ordinary low-mass stars. Now Bahcall et
al., using the Wide-Field Camera of the recently repaired Hubble Space

Expression of CXCL10 is associated with response to radiotherapy and overall survival in squamous cell carcinoma of the tongue
Five-year survival for patients with oral cancer has been disappointingly stable during the last decades, creating a demand for new biomarkers and treatment targets. Lately, much focus has been set on immunomodulation as a possible treatment or an adjuvant increasing sensitivity to conventional treatments. The objective of this study was to evaluate the prognostic importance of response to radiotherapy in tongue carcinoma patients as well as the expression of the CXC-chemokines in correlation to radiation response in the same group of tumours. Thirty-eight patients with tongue carcinoma that had received radiotherapy followed by surgery were included. The prognostic impact of pathological response to radiotherapy, N-status, T-stage, age and gender was evaluated using Cox's regression models, Kaplan-Meier survival curves and chi-square test. The expression of 23 CXC-chemokine ligands and their receptors were evaluated in all patients using microarray and qPCR and correlated with response to treatment using logistic regression. Pathological response to radiotherapy was independently associated to overall survival with a 2-year survival probability of 81 % for patients showing a complete pathological response, while patients with a non-complete response only had a probability of 42 % to survive for 2 years (p = 0.016). The expression of one CXC-chemokine, CXCL10, was significantly associated with response to radiotherapy and the group of patients with the highest CXCL10 expression responded, especially poorly (p = 0.01). CXCL10 is a potential marker for response to radiotherapy and overall survival in patients with squamous cell carcinoma of the tongue
Transit Photometry as an Exoplanet Discovery Method
Photometry with the transit method has arguably been the most successful
exoplanet discovery method to date. A short overview about the rise of that
method to its present status is given. The method's strength is the rich set of
parameters that can be obtained from transiting planets, in particular in
combination with radial velocity observations; the basic principles of these
parameters are given. The method has however also drawbacks, which are the low
probability that transits appear in randomly oriented planet systems, and the
presence of astrophysical phenomena that may mimic transits and give rise to
false detection positives. In the second part we outline the main factors that
determine the design of transit surveys, such as the size of the survey sample,
the temporal coverage, the detection precision, the sample brightness and the
methods to extract transit events from observed light curves. Lastly, an
overview over past, current and future transit surveys is given. For these
surveys we indicate their basic instrument configuration and their planet
catch, including the ranges of planet sizes and stellar magnitudes that were
encountered. Current and future transit detection experiments concentrate
primarily on bright or special targets, and we expect that the transit method
remains a principal driver of exoplanet science, through new discoveries to be
made and through the development of new generations of instruments.Comment: Review chapte
The Phospholipid Scramblases 1 and 4 Are Cellular Receptors for the Secretory Leukocyte Protease Inhibitor and Interact with CD4 at the Plasma Membrane
Secretory leukocyte protease inhibitor (SLPI) is secreted by epithelial cells in all the mucosal fluids such as saliva, cervical mucus, as well in the seminal liquid. At the physiological concentrations found in saliva, SLPI has a specific antiviral activity against HIV-1 that is related to the perturbation of the virus entry process at a stage posterior to the interaction of the viral surface glycoprotein with the CD4 receptor. Here, we confirm that recombinant SLPI is able to inhibit HIV-1 infection of primary T lymphocytes, and show that SLPI can also inhibit the transfer of HIV-1 virions from primary monocyte-derived dendritic cells to autologous T lymphocytes. At the molecular level, we show that SLPI is a ligand for the phospholipid scramblase 1 (PLSCR1) and PLSCR4, membrane proteins that are involved in the regulation of the movements of phospholipids between the inner and outer leaflets of the plasma membrane. Interestingly, we reveal that PLSCR1 and PLSCR4 also interact directly with the CD4 receptor at the cell surface of T lymphocytes. We find that the same region of the cytoplasmic domain of PLSCR1 is involved in the binding to CD4 and SLPI. Since SLPI was able to disrupt the association between PLSCR1 and CD4, our data suggest that SLPI inhibits HIV-1 infection by modulating the interaction of the CD4 receptor with PLSCRs. These interactions may constitute new targets for antiviral intervention
Acute mucosal pathogenesis of feline immunodeficiency virus is independent of viral dose in vaginally infected cats
<p>Abstract</p> <p>Background</p> <p>The mucosal pathogenesis of HIV has been shown to be an important feature of infection and disease progression. HIV-1 infection causes depletion of intestinal lamina propria CD4+ T cells (LPL), therefore, intestinal CD4+ T cell preservation may be a useful correlate of protection in evaluating vaccine candidates. Vaccine studies employing the cat/FIV and macaque/SIV models frequently use high doses of parenterally administered challenge virus to ensure high plasma viremia in control animals. However, it is unclear if loss of mucosal T cells would occur regardless of initial viral inoculum dose. The objective of this study was to determine the acute effect of viral dose on mucosal leukocytes and associated innate and adaptive immune responses.</p> <p>Results</p> <p>Cats were vaginally inoculated with a high, middle or low dose of cell-associated and cell-free FIV. PBMC, serum and plasma were assessed every two weeks with tissues assessed eight weeks following infection. We found that irrespective of mucosally administered viral dose, FIV infection was induced in all cats. However, viremia was present in only half of the cats, and viral dose was unrelated to the development of viremia. Importantly, regardless of viral dose, all cats experienced significant losses of intestinal CD4+ LPL and CD8+ intraepithelial lymphocytes (IEL). Innate immune responses by CD56+CD3- NK cells correlated with aviremia and apparent occult infection but did not protect mucosal T cells. CD4+ and CD8+ T cells in viremic cats were more likely to produce cytokines in response to Gag stimulation, whereas aviremic cats T cells tended to produce cytokines in response to Env stimulation. However, while cell-mediated immune responses in aviremic cats may have helped reduce viral replication, they could not be correlated to the levels of viremia. Robust production of anti-FIV antibodies was positively correlated with the magnitude of viremia.</p> <p>Conclusions</p> <p>Our results indicate that mucosal immune pathogenesis could be used as a rapid indicator of vaccine success or failure when combined with a physiologically relevant low dose mucosal challenge. We also show that innate immune responses may play an important role in controlling viral replication following acute mucosal infection, which has not been previously identified.</p

Anomaly Mediation and Cosmology
We consider an extension of the MSSM wherein anomaly mediation is the source
of supersymmetry breaking, and the tachyonic slepton problem is solved by a
gauged U(1) symmetry, which is broken at high energies in a manner preserving
supersymmetry, thereby also facilitating the see-saw mechanism for neutrino
masses and a natural source for the Higgs mu-term. We show that these
favourable outcomes can occur both in the presence and the absence of a large
Fayet-Iliopoulos (FI) D-term associated with the new U(1). We explore the
cosmological consequences of the model, showing that it naturally produces a
period of hybrid inflation, terminating in the production of cosmic strings. In
spite of the presence of a U(1) (even with an FI term), inflation is effected
by the F-term, with a D-flat tree potential (the FI term, if present, being
cancelled by non-zero squark and slepton fields). Calculating the 1-loop
corrections to the inflaton potential, we estimate the constraints on the
parameters of the model from Cosmic Microwave Background data. We will see that
a consequence of these constraints is that the Higgs mu-term necessarily small.
We briefly discuss the mechanisms for baryogenesis via conventional
leptogenesis, the out-of-equilibrium production of neutrinos from the cosmic
strings, or the Affleck-Dine mechanism. Cosmic string decays also boost the
relic density of dark matter above the low value normally obtained in AMSB

Hydrogeological typologies of the Indo-Gangetic basin alluvial aquifer, South Asia
The Indo-Gangetic aquifer is one of the world’s most important transboundary water resources, and the most heavily exploited aquifer in the world. To better understand the aquifer system, typologies have been characterized for the aquifer, which integrate existing datasets across the Indo-Gangetic catchment basin at a transboundary scale for the first time, and provide an alternative conceptualization of this aquifer system. Traditionally considered and mapped as a single homogenous aquifer of comparable aquifer properties and groundwater resource at a transboundary scale, the typologies illuminate significant spatial differences in recharge, permeability, storage, and groundwater chemistry across the aquifer system at this transboundary scale. These changes are shown to be systematic, concurrent with large-scale changes in sedimentology of the Pleistocene and Holocene alluvial aquifer, climate, and recent irrigation practices. Seven typologies of the aquifer are presented, each having a distinct set of challenges and opportunities for groundwater development and a different resilience to abstraction and climate change. The seven typologies are: (1) the piedmont margin, (2) the Upper Indus and Upper-Mid Ganges, (3) the Lower Ganges and Mid Brahmaputra, (4) the fluvially influenced deltaic area of the Bengal Basin, (5) the Middle Indus and Upper Ganges, (6) the Lower Indus, and (7) the marine-influenced deltaic areas
The dominant Anopheles vectors of human malaria in the Asia-Pacific region: occurrence data, distribution maps and bionomic précis
<p>Abstract</p> <p>Background</p> <p>The final article in a series of three publications examining the global distribution of 41 dominant vector species (DVS) of malaria is presented here. The first publication examined the DVS from the Americas, with the second covering those species present in Africa, Europe and the Middle East. Here we discuss the 19 DVS of the Asian-Pacific region. This region experiences a high diversity of vector species, many occurring sympatrically, which, combined with the occurrence of a high number of species complexes and suspected species complexes, and behavioural plasticity of many of these major vectors, adds a level of entomological complexity not comparable elsewhere globally. To try and untangle the intricacy of the vectors of this region and to increase the effectiveness of vector control interventions, an understanding of the contemporary distribution of each species, combined with a synthesis of the current knowledge of their behaviour and ecology is needed.</p> <p>Results</p> <p>Expert opinion (EO) range maps, created with the most up-to-date expert knowledge of each DVS distribution, were combined with a contemporary database of occurrence data and a suite of open access, environmental and climatic variables. Using the Boosted Regression Tree (BRT) modelling method, distribution maps of each DVS were produced. The occurrence data were abstracted from the formal, published literature, plus other relevant sources, resulting in the collation of DVS occurrence at 10116 locations across 31 countries, of which 8853 were successfully geo-referenced and 7430 were resolved to spatial areas that could be included in the BRT model. A detailed summary of the information on the bionomics of each species and species complex is also presented.</p> <p>Conclusions</p> <p>This article concludes a project aimed to establish the contemporary global distribution of the DVS of malaria. The three articles produced are intended as a detailed reference for scientists continuing research into the aspects of taxonomy, biology and ecology relevant to species-specific vector control. This research is particularly relevant to help unravel the complicated taxonomic status, ecology and epidemiology of the vectors of the Asia-Pacific region. All the occurrence data, predictive maps and EO-shape files generated during the production of these publications will be made available in the public domain. We hope that this will encourage data sharing to improve future iterations of the distribution maps.</p
Search for strongly interacting massive particles generating trackless jets in proton-proton collisions at s = 13 TeV
A search for dark matter in the form of strongly interacting massive particles (SIMPs) using the CMS detector at the LHC is presented. The SIMPs would be produced in pairs that manifest themselves as pairs of jets without tracks. The energy fraction of jets carried by charged particles is used as a key discriminator to suppress efficiently the large multijet background, and the remaining background is estimated directly from data. The search is performed using proton-proton collision data corresponding to an integrated luminosity of 16.1 fb - 1 , collected with the CMS detector in 2016. No significant excess of events is observed above the expected background. For the simplified dark matter model under consideration, SIMPs with masses up to 100 GeV are excluded and further sensitivity is explored towards higher masses
