This phenomenon, known as low doses hyperradiosensitivity (LDHRS), appears as an opportunity to decrease toxicity of radiotherapy and to enhance the effects of chemotherapy. However, the effect of low single doses IR on cell death is subtle and the mechanism underlying LDHRS has not been clearly explained, limiting the utility of LDHRS for clinical applications. To understand the mechanisms responsible for cell death induced by low-dose IR, LDHRS was evaluated in DLD-1 human colorectal cancer cells and the expression of 80 microRNAs (miRNAs) was assessed by qPCR array. Our results show that DLD-1 cells display an early DNA damage response and apoptotic cell death when exposed to 0.6 Gy. miRNA expression profiling identified 3 over-expressed (miR-205-3p, miR-1 and miR-133b) and 2 downregulated miRNAs (miR-122-5p, and miR-134-5p) upon exposure to 0.6 Gy. This miRNA profile differed from the one in cells exposed to high-dose IR (12 Gy), supporting a distinct low-dose radiation-induced cell death mechanism. Expression of a mimetic miR- 205-3p, the most overexpressed miRNA in cells exposed to 0.6 Gy, induced apoptotic cell death and, more importantly, increased LDHRS in DLD-1 cells. Thus, we propose miR-205-3p as a potential radiosensitizer to low-dose IR. © Andaur et al.http://www.oncotarget.com/index.php?journal=oncotarget&page=article&op=view&path[]=25405&path[]=7956

Effect of graphene substrate on the SERS Spectra of Aromatic bifunctional molecules on metal nanoparticles

    Get PDF
The design of molecular sensors plays a very important role within nanotechnology and especially in the development of different devices for biomedical applications. Biosensors can be classified according to various criteria such as the type of interaction established between the recognition element and the analyte or the type of signal detection from the analyte (transduction). When Raman spectroscopy is used as an optical transduction technique the variations in the Raman signal due to the physical or chemical interaction between the analyte and the recognition element has to be detected. Therefore any significant improvement in the amplification of the optical sensor signal represents a breakthrough in the design of molecular sensors. In this sense, Surface-Enhanced Raman Spectroscopy (SERS) involves an enormous enhancement of the Raman signal from a molecule in the vicinity of a metal surface. The main objective of this work is to evaluate the effect of a monolayer of graphene oxide (GO) on the distribution of metal nanoparticles (NPs) and on the global SERS enhancement of paminothiophenol (pATP) and 4-mercaptobenzoic acid (4MBA) adsorbed on this substrate. These aromatic bifunctional molecules are able to interact to metal NPs and also they offer the possibility to link with biomolecules. Additionally by decorating Au or Ag NPs on graphene sheets, a coupled EM effect caused by the aggregation of the NPs and strong electronic interactions between Au or Ag NPs and the graphene sheets are considered to be responsible for the significantly enhanced Raman signal of the analytes [1-2]. Since there are increasing needs for methods to conduct reproducible and sensitive Raman measurements, Grapheneenhanced Raman Scattering (GERS) is emerging as an important method [3].Universidad de Málaga. Velocity Statistics in the Two-Dimensional Granular Turbulence

We studied the macroscopic statistical properties on the freely evolving quasi-elastic hard disk (granular) system by performing a large-scale (up to a few million particles) event-driven molecular dynamics systematically and found that remarkably analogous to an enstrophy cascade process in the decaying two-dimensional fluid turbulence. There are four typical stages in the freely evolving inelastic hard disk system, which are homogeneous, shearing (vortex), clustering and final state. In the shearing stage, the self-organized macroscopic coherent vortices become dominant. In the clustering stage, the energy spectra are close to the expectation of Kraichnan-Batchelor theory and the squared two-particle separation strictly obeys Richardson law.Comment: 4 pages, 4 figures, to be published in PR

Early-Middle Miocene subtle compressional deformation in the Ebro foreland basin (northern Spain); insights from magnetic fabrics

    Get PDF
The results of anisotropy of magnetic susceptibility analyses of 19 sites carried out on magnetostrigraphically-dated, Lower to Middle Miocene (20.4 to 13.7 Ma) 'non-deformed' mudstones from the central part of the Ebro basin reveal the presence of a subtle tectonic overprint lasting at least until the Langhian (Middle Miocene) in the southern Pyrenean foreland. Magnetic ellipsoids show a sedimentary fabric in 42% of sites and a weak and well-defined magnetic lineation in 47% and 11% of sites. The magnetic lineation is roughly oriented around the east-west direction, compatible with a very weak deformation occurring there and related to the north-south compression linked to the convergence between Europe, Iberia and Africa during the Early-Middle Miocene. A slight variation of the magnetic parameters Km, Pj, and T exists through time, probably due to changes in the sedimentary conditions in the basin

Inoculation of the Leishmania infantum HSP70-II null mutant induces long-term protection against L. amazonensis infection in BALB/c mice

Leishmania amazonensis parasites are etiological agents of cutaneous leishmaniasis in the New World. BALB/c mice are highly susceptible to L. amazonensis challenge due to their inability to mount parasite-dependent IFN-γ-mediated responses. Here, we analyzed the capacity of a single administration of the LiΔHSP70-II genetically-modified attenuated L. infantum line in preventing cutaneous leishmaniasis in mice challenged with L. amazonensis virulent parasites. In previous studies, this live attenuated vaccine has demonstrated to induce long-protection against murine leishmaniasis due to Old World Leishmania species. Vaccinated mice showed a reduction in the disease evolution due to L. amazonensis challenge, namely reduction in cutaneous lesions and parasite burdens. In contrast to control animals, after the challenge, protected mice showed anti-Leishmania IgG2a circulating antibodies accompanied to the induction of Leishmania-driven specific IFN-γ systemic response. An analysis performed in the lymph node draining the site of infection revealed an increase of the parasite-specific IFN-ϒ production by CD4+ and CD8+ T cells and a decrease in the secretion of IL-10 against leishmanial antigens. Since the immunity caused by the inoculation of this live vaccine generates protection against different forms of murine leishmaniasis, we postulate LiΔHSP70-II as a candidate for the development of human vaccines. This research was funded by grants from Ministerio de Ciencia e Innovación FISPI11/00095 and FISPI14/00366 (FEDER FUNDING) to M.S. and RYC-2016-19463 and RTI2018-343 to S.I. Late Pleistocene to Holocene palaeoenvironmental variability in the north-west Spanish mountains: insights from a source-to-sink environmental magnetic study of Lake Sanabria

    Get PDF
International audienceWe present a source-to-sink environmental magnetic study of a sediment core from Lake Sanabria (north-west Iberian Peninsula) and rocks of its catchment. The results indicate the occurrence of magnetite, and probably also pyrrhotite, in sediments accumulated between ca. 26 and 13 cal ka BP in a proglacial lake environment. These minerals also appear to dominate the magnetic assemblage of Palaeozoic rocks from the lake catchment. This indicates that sedimentation was then driven by the erosion of glacial flour, which suffered minimal chemical transformation due to a rapid and short routing to the lake. A sharp change in magnetic properties observed in the lake sediments between 13 and 12.6 cal ka BP reflects the rapid retreat of glaciers from the lake catchment. Sediments from the upper half of the studied sequence, accumulated after 12.6 cal ka BP in a lacustrine environment with strong fluvial influence, contain magnetite and smaller amounts of maghemite and greigite. We suggest that greigite grew authigenically under anoxic conditions caused by enhanced accumulation of organic matter into the lake. The occurrence of maghemite in these sediments suggests pedogenic activity in the then deglaciated lake catchment before the erosion and transportation of detrital material into the lake

Scale insects are frequent invaders of new territories. The Bougainvillea mealybug, Phenacoccus peruvianus Granara de Willink, 2007, was recorded in Europe for the first time in 1999 in Spain (Almeria) and later in 2002 in Italy (Sicily). Initially, this unknown species was identified provisionally as Phenacoccus sp. Records of this species became frequent later when it was found in other localities in Spain (including the Balearic Islands), Great Britain, France (including Corsica), Monaco, and Portugal. The host plants of this mealybug were in most cases species of Bougainvillea

A model for the atomic-scale structure of a dense, nonequilibrium fluid: the homogeneous cooling state of granular fluids

It is shown that the equilibrium Generalized Mean Spherical Model of fluid structure may be extended to nonequilibrium states with equation of state information used in equilibrium replaced by an exact condition on the two-body distribution function. The model is applied to the homogeneous cooling state of granular fluids and upon comparison to molecular dynamics simulations is found to provide an accurate picture of the pair distribution function.Comment: 29 pages, 11 figures Revision corrects formatting of the figure
