    Impact of Dosing Intervals on Activity of Gentamicin and Ticarcillin Against Pseudomonas aeruginosa in Granulocytopenic Mice

    The influence of dosing intervals on the activity of gentamicin and ticarcillin against Pseudomonas aeruginosa was studied in vivo. Granulocytopenic mice infected with P. aeruginosa in the thigh muscle were treated with l-hr or 3-hr injections of gentamicin, ticarcillin, or gentamicin-ticarcillin. Plasma pharmacokinetics of the drugs were correlated with antibacterial activity. Gentamicin injected every 1 hr tended to be less active than gentamicin injected at longer intervals. In contrast, ticarcillin given every 1 hr was significantly more efficacious than equivalent total doses injected every 3 hr. The dosing schedule of gentamicin-ticarcillin was again important for ticarcillin but did not appreciably affect the antibacterial activity of gentamicin. Thus, antimicrobial chemotherapy of P. aeruginosa infections in the granulocytopenic host might be improved by administering ticarcillin rather than gentamicin as a constant infusio

    La pandemia provocada por la Covid-19 ha influido en los estilos de vida de las personas. El escenario de confinamiento domiciliario podría perjudicar la práctica de actividad física, generando nuevos desafíos para el mantenimiento de la salud durante la pandemia de la Covid-19. El objetivo de este estudio fue presentar el contexto actual de la pandemia de Covid-19, su impacto en la práctica de actividad física y las estrategias disponibles para mantenerse activo durante el confinamiento domiciliario según las recomendaciones internacionales. La revisión narrativa se realizó con base en estudios que analizaron temas relacionados con la actividad física y la Covid-19. Se realizaron búsquedas de artículos relevantes en las bases de datos Virtual Health Library (BVS), CINAHL, Cochrane, PsycINFO, PubMed, ScienceDirect, Scientific Electronic Library Online (SciELO), Scopus, SPORTDiscus y Web of Science. Aunque todavía es necesario un mayor número de estudios experimentales, las personas deberían dedicar más tiempo a la actividad física durante el aislamiento social. Las pautas fueron ajustadas por entidades internacionales para alentar a las personas a mantenerse activas, mediante la práctica de actividad física regular, utilizando estrategias alternativas como aplicaciones de programas de acondicionamiento físico, exergames, clases de ejercicios en línea e incluso tareas domésticas. La reducción del sedentarismo prolongado podría contribuir a mantener la salud y mejorar la calidad de vida durante la pandemia de Covid-19.  The pandemic caused by the COVID-19 has influenced peoples’ lifestyles. Home-confinement scenario might impair physical activity practice, resulting in new challenges for maintaining health during the pandemic of the COVID-19. The aim of this study was to present the current context of COVID-19 pandemic, its impact on the practice of physical activity, and the strategies available to remain active during home-confinement according to international recommendations. The narrative review was conducted based on studies that analyzed themes related to physical activity and COVID-19. Virtual Health Library (VHL), CINAHL, Cochrane, PsycINFO, PubMed, ScienceDirect, Scientific Electronic Library Online (SciELO), Scopus, SPORTDiscus, and Web of Science databases were searched for relevant papers. Although an increased number of experimental studies are still necessary, people should devote more time to physical activity during social isolation. Guidelines were adjusted by international entities in order to encourage people to remain active, through practice regular physical activity, using alternative strategies such as fitness program applications, exergames, online exercise classes, and even chores. Reduction of prolonged sedentary behavior could contribute to maintain health and improve quality of life during the COVID-19 pandemic

    100 Hz ROCS microscopy correlated with fluorescence reveals cellular dynamics on different spatiotemporal scales

    Fluorescence techniques dominate the field of live-cell microscopy, but bleaching and motion blur from too long integration times limit dynamic investigations of small objects. High contrast, label-free life-cell imaging of thousands of acquisitions at 160 nm resolution and 100 Hz is possible by Rotating Coherent Scattering (ROCS) microscopy, where intensity speckle patterns from all azimuthal illumination directions are added up within 10 ms. In combination with fluorescence, we demonstrate the performance of improved Total Internal Reflection (TIR)-ROCS with variable illumination including timescale decomposition and activity mapping at five different examples: millisecond reorganization of macrophage actin cortex structures, fast degranulation and pore opening in mast cells, nanotube dynamics between cardiomyocytes and fibroblasts, thermal noise driven binding behavior of virus-sized particles at cells, and, bacterial lectin dynamics at the cortex of lung cells.     Discovery and Follow-up Observations of the Young Type Ia Supernova 2016coj

    The Type~Ia supernova (SN~Ia) 2016coj in NGC 4125 (redshift z=0.004523z=0.004523) was discovered by the Lick Observatory Supernova Search 4.9 days after the fitted first-light time (FFLT; 11.1 days before BB-band maximum). Our first detection (pre-discovery) is merely 0.6±0.50.6\pm0.5 day after the FFLT, making SN 2016coj one of the earliest known detections of a SN Ia. A spectrum was taken only 3.7 hr after discovery (5.0 days after the FFLT) and classified as a normal SN Ia. We performed high-quality photometry, low- and high-resolution spectroscopy, and spectropolarimetry, finding that SN 2016coj is a spectroscopically normal SN Ia, but with a high velocity of \ion{Si}{2} λ\lambda6355 (12,600\sim 12,600\,\kms\ around peak brightness). The \ion{Si}{2} λ\lambda6355 velocity evolution can be well fit by a broken-power-law function for up to a month after the FFLT. SN 2016coj has a normal peak luminosity (MB18.9±0.2M_B \approx -18.9 \pm 0.2 mag), and it reaches a BB-band maximum \about16.0~d after the FFLT. We estimate there to be low host-galaxy extinction based on the absence of Na~I~D absorption lines in our low- and high-resolution spectra. The spectropolarimetric data exhibit weak polarization in the continuum, but the \ion{Si}{2} line polarization is quite strong (0.9%±0.1%\sim 0.9\% \pm 0.1\%) at peak brightness.Comment: Submitte

    Cryogenic time-domain multiplexer based on SQUID arrays and superconducting/normal conducting switches

It was manufactured using a fabrication process that is cross-compatible between VTT and IPHT-Jena. The multiplexer consists of twelve 12-SQUID series arrays, each shunted by a Zappe-style interferometer array acting as a flux-controlled superconducting/normal conducting switch. By keeping all switches but one in the superconducting state, it is possible to select one active readout channel at a time. A flux feedback coil common to all SQUID arrays allows realization of a flux-locked loop. We present characteristics of the multiplexer and measurement data from experiments with a 25-pixel X-ray calorimeter array operated at T <100 mK in a dilution refrigerator.Peer reviewe

    Human transmissible spongiform encephalopathies in eleven countries: diagnostic pattern across time, 1993–2002

    BACKGROUND: The objective of this study was to describe the diagnostic panorama of human transmissible spongiform encephalopathies across 11 countries. METHODS: From data collected for surveillance purposes, we describe annual proportions of deaths due to different human transmissible spongiform encephalopathies in eleven EUROCJD-consortium countries over the period 1993–2002, as well as variations in the use of diagnostic tests. Using logistic models we quantified international differences and changes across time. RESULTS: In general, pre-mortem use of diagnostic investigations increased with time. International differences in pathological confirmation of sporadic Creutzfeldt-Jakob disease, stable over time, were evident. Compared to their counterparts, some countries displayed remarkable patterns, such as: 1) the high proportion, increasing with time, of variant Creutzfeldt-Jakob disease in the United Kingdom, (OR 607.99 95%CI 84.72–4363.40), and France (OR 18.35, 95%CI 2.20–152.83); 2) high, decreasing proportions of iatrogenic Creutzfeldt-Jakob disease in France, (OR 5.81 95%CI 4.09–8.24), and the United Kingdom, (OR 1.54 95%CI 1.03–2.30); and, 3) high and stable ratios of genetic forms in Slovakia (OR 21.82 95%CI 12.42–38.33) and Italy (OR 2.12 95%CI 1.69–2.68). CONCLUSION: Considerable international variation in aetiological subtypes of human transmissible spongiform encephalopathies was evident over the observation period. With the exception of variant Creutzfeldt-Jakob disease and iatrogenic Creutzfeldt-Jakob disease in France and the United Kingdom, these differences persisted across time

    Biomarkers and diagnostic guidelines for sporadic Creutzfeldt-Jakob disease

    Sporadic Creutzfeldt-Jakob disease is a fatal neurodegenerative disease caused by misfolded prion proteins (PrP^{Sc}). Effective therapeutics are currently not available and accurate diagnosis can be challenging. Clinical diagnostic criteria use a combination of characteristic neuropsychiatric symptoms, CSF proteins 14-3-3, MRI, and EEG. Supportive biomarkers, such as high CSF total tau, could aid the diagnostic process. However, discordant studies have led to controversies about the clinical value of some established surrogate biomarkers. Development and clinical application of disease-specific protein aggregation and amplification assays, such as real-time quaking induced conversion (RT-QuIC), have constituted major breakthroughs for the confident pre-mortem diagnosis of sporadic Creutzfeldt-Jakob disease. Updated criteria for the diagnosis of sporadic Creutzfeldt-Jakob disease, including application of RT-QuIC, should improve early clinical confirmation, surveillance, assessment of PrP^{Sc} seeding activity in different tissues, and trial monitoring. Moreover, emerging blood-based, prognostic, and potentially pre-symptomatic biomarker candidates are under investigation

    Health professions and risk of sporadic Creutzfeldt- Jakob disease, 1965 to 2010

    In 2009, a pathologist with sporadic Creutzfeldt- Jakob Disease (sCJD) was reported to the Spanish registry. This case prompted a request for information on health-related occupation in sCJD cases from countries participating in the European Creutzfeldt Jakob Disease Surveillance network (EuroCJD). Responses from registries in 21 countries revealed that of 8,321 registered cases, 65 physicians or dentists, two of whom were pathologists, and another 137 healthcare workers had been identified with sCJD. Five countries reported 15 physicians and 68 other health professionals among 2,968 controls or non-cases, suggesting no relative excess of sCJD among healthcare professionals. A literature review revealed: (i) 12 case or small case-series reports of 66 health professionals with sCJD, and (ii) five analytical studies on health-related occupation and sCJD, where statistically significant findings were solely observed for persons working at physicians' offices (odds ratio: 4.6 (95 CI: 1.2-17.6)). We conclude that a wide spectrum of medical specialities and health professions are represented in sCJD cases and that the data analysed do not support any overall increased occupational risk for health professionals. Nevertheless, there may be a specific risk in some professions associated with direct contact with high human-infectivity tissue

    Technological processes for the fabrication of low- and high- Tc Josephson junctions, aimed for certain applications, are described. On the one hand, the integration of low- Tc superconductor digital electronics with superconducting sensor arrays enables input signal processing with quantum limited resolution at millikelvin temperatures. We describe this mixed signal superconductor technology for analogue sensor readout and signal multiplexing for operating temperatures down to 300 mK. On the other hand, by making use of modern high- Tc Josephson junction technology, sensitive magnetometers, which require a modest cooling power, can be developed. Examples of the application of the mentioned processes are shown
