Dosimetry in small x-ray fields
Treballs Finals de Grau de Física, Facultat de Física, Universitat de Barcelona, Curs: 2018, Tutor: José M. Fernández-VareaIn this work we study the behaviour of small x-ray fields in radiotherapy, whose dosimetry presents a number of problems. The study requires learning how to use the penEasy program. The analysis consists on measure the absorbed dose according to the field size. The study allows us to discuss some graphical representations of the data computed and compare them with experimental values taken with an Exradin W1 Scintillato
(HAM, A.
QUINTERO, J. BUTLER, S. SAINSBURY, B. NOBLE, J. ST.LOUIS, R.
CHRISTIANI, K. SYMINGTON, A. 2014).
(ESTEBAN TALAYA,
A. 2008).
actividad respectivamente. (MARÍ VIDAL, S; MATEOS RONCO, A; POLO
GARRIDO, F; SEGUÍ MAS, E. 2003).
Orobal Iscla, P. (2015). PLAN DE EMPRESA PARA UN COMPLEJO TURÍSTICO EN LA MANGA DEL MAR MENOR (MURCIA). http://hdl.handle.net/10251/57308.Archivo delegad
An agonist of the MscL channel affects multiple bacterial species and increases membrane permeability and potency of common antibiotics
The bacterial MscL channel normally functions as an emergency release valve discharging cytoplasmic solutes upon osmotic stress. The channel opens and passes molecules up to 30 Å and its pore is the largest of any gated channel. Opening the MscL pore inappropriately is detrimental to the bacterial cell, suggesting MscL as a potential novel drug target. A small‐molecule compound, 011A, has been shown to increase sensitivity of the Escherichia coli MscL channel, slow growth, and even decrease viability of quiescent cultures. The mscL gene is highly conserved and found in the vast majority of bacterial species, including pathogens. Here, we test the hypothesis that 011A can influence the growth and viability of other bacterial species, specifically Staphylococcus aureus and Mycobacterium smegmatis, in a MscL‐dependent manner. Furthermore, we demonstrate that the 011A compound can increase potency of other antibiotics, presumably by permeabilizing the membrane and allowing easier access of the antibiotic into the cytoplasm. Thus, MscL activators have potential as novel broad‐spectrum antibiotics or adjuvants that work with antibiotics to selectively allow passage across bacterial membranes
An improved open-channel structure of MscL determined from FRET confocal microscopy and simulation
Mechanosensitive channels act as molecular transducers of mechanical force exerted on the membrane of living cells by opening in response to membrane bilayer deformations occurring in physiological processes such as touch, hearing, blood pressure regulation, and osmoregulation. Here, we determine the likely structure of the open state of the mechanosensitive channel of large conductance using a combination of patch clamp, fluorescence resonance energy transfer (FRET) spectroscopy, data from previous electron paramagnetic resonance experiments, and molecular and Brownian dynamics simulations. We show that structural rearrangements of the protein can be measured in similar conditions as patch clamp recordings while controlling the state of the pore in its natural lipid environment by modifying the lateral pressure distribution via the lipid bilayer. Transition to the open state is less dramatic than previously proposed, while the N terminus remains anchored at the surface of the membrane where it can either guide the tilt of or directly translate membrane tension to the conformation of the pore-lining helix. Combining FRET data obtained in physiological conditions with simulations is likely to be of great value for studying conformational changes in a range of multimeric membrane proteins

Under the State of alarm aiming to halt the spread of COVID-19, personal and civil liberties have been cut through the national lockdown. Social control has been imposed by law and restrictions, but control has also operated from citizens to citizens. Based on the results of interviews and surveys we've done, we can point out how the population has accepted social control and how they have contributed to the constitution of this state. This research shows the diversity of reactions and behaviors in the assimilation of the new measures
S. aureus MscL Is a Pentamer In Vivo but of Variable Stoichiometries In Vitro: Implications for Detergent-Solubilized Membrane Proteins
Detergent-induced rearrangements of membrane-protein subunits explain why two MscL channel stoichiometries have been resolved by X-ray crystallography - but S. aureus MscL is truly a pentamer in vivo
A new antibiotic with potent activity targets MscL
The growing problem of antibiotic-resistant bacteria is a major threat to human health. Paradoxically, new antibiotic discovery is declining, with most of the recently approved antibiotics corresponding to new uses for old antibiotics or structurally similar derivatives of known antibiotics. We used an in silico approach to design a new class of nontoxic antimicrobials for the bacteria-specific mechanosensitive ion channel of large conductance, MscL. One antimicrobial of this class, compound 10, is effective against methicillin-resistant Staphylococcus aureus with no cytotoxicity in human cell lines at the therapeutic concentrations. As predicted from in silico modeling, we show that the mechanism of action of compound 10 is at least partly dependent on interactions with MscL. Moreover we show that compound 10 cured a methicillin-resistant S. aureus infection in the model nematode Caenorhabditis elegans. Our work shows that compound 10, and other drugs that target MscL, are potentially important therapeutics against antibiotic-resistant bacterial infections.Irene Iscla, Robin Wray, Paul Blount, Jonah Larkins-Ford, Annie L Conery, Frederick M Ausubel, Soumya Ramu, Angela Kavanagh, Johnny X Huang, Mark A Blaskovich, Matthew A Cooper, Andres Obregon-Henao, Ian Orme, Edwin S Tjandra, Uwe H Stroeher, Melissa H Brown, Cindy Macardle, Nick van Holst, Chee Ling Tong, Ashley D Slattery, Christopher T Gibson, Colin L Raston and Ramiz A Boulo
