    Activation of T cells by dendritic cells exposed to a reference sensitizer Towards a promising model to assess the allergenic potential of chemicals

    International audienceBackground - Low molecular weight chemicals constitute one of the major causes of occupational allergies. European legislation on chemicals recommends limiting the use of in vivo models for assessing the sensitizing potential of chemicals, and encourages the development of integrated alternative methods. An in vitro mouse model of bone marrow-derived dendritic cells (BMDCs) that showed good accuracy (75%) and sensitivity (69%) has previously been developed to assess the sensitizing potential of chemicals. Objective - To assess the ability of BMDCs to activate T cells (TCs) in vitro. Methods - BMDCs pre-exposed to the reference sensitizer ammonium hexachloroplatinate (AHCP) were co-cultured with different subpopulations of TCs. TC activation was assessed by surface marker expression, proliferation, and cytokine release. Results - The results showed significant activation of TCs co-cultured with dendritic cells pre-exposed to AHCP as evaluated by CD124 expression, proliferation, and cytokine secretion. Moreover, the response of TCs appeared to be Th2-oriented. Naive TCs were shown to be involved in this response, and the removal of regulatory TCs did not improve the cell response. Conclusions - The BMDCs used in this previously developed model appear to have the ability to activate TCs, confirming that the BMDC model represents a reliable assay for assessing the sensitizing potential of chemicals

    Radiatively Broadened Incandescent Sources

    We study the incandescence of a semiconductor system characterized by a radiatively broadened material excitation. We show that the shape of the emission spectrum and the peak emissivity value are determined by the ratio between radiative and nonradiative relaxation rates of the material mode. Our system is a heavily doped quantum well, exhibiting a collective bright electronic excitation in the mid-infrared. The spontaneous emission rate of this collective mode strongly depends on the emission direction and, uncommonly for an intersubband system, can dominate nonradiative scattering processes. Consequently the incandescence spectrum undergoes strong modifications when the detection angle is varied. Incandescence is modeled solving quantum Langevin equations, including a microscopic description of the collective excitations, decaying into electronic and photonic baths. We demonstrate that the emissivity reaches unity value for a well-defined direction and presents an angular radiative pattern that is very different from that of an oscillating dipole

    Fluctuating hearing loss in the only hearing ear: cochlear implantation in the contralateral deaf side

    Objective To investigate the hearing performance of adult patients presenting unilateral deafness with contralateral fluctuating hearing loss who received a cochlear implant on the deaf side. Study Design Case series with chart review. Setting University tertiary referral center. Subjects and Methods Preoperatively and at 6 and 12 months postoperatively, 23 patients underwent pure tone audiometry and speech audiometry with disyllabic and monosyllabic words in a quiet environment and sentences in quiet and noisy (signal-to-noise ratio +10 dB SPL) environments under best-aided conditions. The Abbreviated Profile of Hearing Aid Benefit (APHAB) inventory was evaluated preoperatively and at 6 and 12 months postoperatively. Results No difference was found between pre- and postoperative tests for disyllabic and monosyllabic words. For sentences in quiet and noisy environments, a difference between pre- and postoperative performance was present at 1 year ( P = .002 and P = .02, respectively). In a noisy environment, a difference was present at 6 and 12 months postoperatively as compared with the preoperative value (mean ± SD: 6 months: 42% ± 7.1% vs 61% ± 6.5%, P = .016). A significant improvement in the APHAB score was found at 6 and 12 months postimplantation (Friedman's 2-way analysis of variance by ranks, P < .001). The number of years of hearing deprivation of the deaf ear was not correlated with performance. Conclusion When incapacitating fluctuating hearing loss occurs in patients presenting a contralateral deaf ear, a cochlear implant is indicated in the latter ear, significantly improving performance in noisy conditions and allowing a better quality of communication to be achieved

    Olivine-phyric basalt in the Mesoproterozoic Gawler silicic large igneous province, South Australia: Examples at the Olympic Dam Iron Oxide Cu-U-Au-Ag deposit and other localities

    The felsic-dominant Gawler Range Volcanics and cogenetic Hiltaba Suite granitoids constitute the ca. 1590 Ma Gawler silicic large igneous province in the Gawler Craton, South Australia. The province includes minor occurrences of olivine-phyric basalt at Kokatha and Mount Gunson. In this study, we describe additional olivine-phyric basalts and dykes intersected by drill holes at the Olympic Dam Iron Oxide Cu–U–Au–Ag deposit and the Wirrda Well Cu–Au prospect. Laser Ablation Inductively Coupled Plasma Mass Spectrometry U–Pb dating results (un-anchored) of apatite in the basalts and dykes at Mount Gunson (1576 ± 33 Ma), Wirrda Well (1596 ± 17 Ma), and Olympic Dam (1621 ± 20 Ma) confirm their effective temporal correlation with the ca. 1590 Ma Gawler silicic large igneous province. Compositions of Cr-spinel inclusions enclosed in former olivine phenocrysts (forsterite number >80) in basalts at Mount Gunson, Wirrda Well and Olympic Dam imply derivation from a heterogeneous mantle source that may have been modified by subduction. Least-altered olivine-phyric basalts at Mount Gunson and Kokatha are characterized by negative Nb and Ta anomalies, which are typical of arc basalts but are also common in back-arc basin basalts, in accordance with the result suggested by the Cr-spinel source indicator. The mafic components of the Gawler Range Volcanics generally have higher Zr contents and Zr/TiO2 ratios than those of high-Mg basalts and picrites produced in variable tectonic settings worldwide, possibly reflecting continental crustal components involved in their mantle source.High abundances (typically ~20 vol.%) of former olivine phenocrysts in the basalt at Olympic Dam imply high-Mg whole-rock compositions and a high temperature of the primary magma. This result is consistent with previous models in which the heat flux from mantle magmas caused large-scale partial melting of crustal rocks that subsequently gave rise to silicic magmas erupted and intruded to form the Gawler silicic large igneous province
