    Impact of surface structure and feed gas composition on Bacillus subtilis endospore inactivation during direct plasma treatment

    This study investigated the inactivation efficiency of cold atmospheric pressure plasma treatment on Bacillus subtilis endospores dependent on the used feed gas composition and on the surface, the endospores were attached on. Glass petri-dishes, glass beads, and peppercorns were inoculated with the same endospore density and treated with a radio frequency plasma jet. Generated reactive species were detected using optical emission spectroscopy. A quantitative polymerase chain reaction (qPCR) based ratio detection system was established to monitor the DNA damage during the plasma treatment. Argon + 0.135% vol. oxygen + 0.2% vol. nitrogen as feed gas emitted the highest amounts of UV-C photons and considerable amount of reactive oxygen and nitrogen species. Plasma generated with argon + 0.135% vol. oxygen was characterized by the highest emission of reactive oxygen species (ROS), whereas the UV-C emission was negligible. The use of pure argon showed a negligible emission of UV photons and atomic oxygen, however, the emission of vacuum (V)UV photons was assumed. Similar maximum inactivation results were achieved for the three feed gas compositions. The surface structure had a significant impact on the inactivation efficiency of the plasma treatment. The maximum inactivation achieved was between 2.4 and 2.8 log10 on glass petri-dishes and 3.9 to 4.6 log10 on glass beads. The treatment of peppercorns resulted in an inactivation lower than 1.0 log10. qPCR results showed a significant DNA damage for all gas compositions. Pure argon showed the highest results for the DNA damage ratio values, followed by argon + 0.135% vol. oxygen + 0.2% vol. nitrogen. In case of argon + 0.135% vol. oxygen the inactivation seems to be dominated by the action of ROS.     Performance prediction and optimization for industrial sieves by simulation : a two-tier approach

    We present a numerical study of sieving behavior on industrial sieves, composed of several vibrating screens, a process widely used in diverse industries. The modeling approach is twofold: on the one hand, particle flow is modeled in some detail by means of a discrete element model (DEM). This allows studying the influence of various parameters on the behavior of individual particles, particularly transport velocities and collision rates. Computational complexity however forbids the simulation of an entire sieve as a DEM. Instead, the overall sieving behavior is modeled separately by means of a more phenomenological model, the so called thick layer model (TLM), which is based on mass-balance equations, that translate into an ordinary differential equation. The TLM obtains its most crucial input parameters as results of the DEM. Comparison of simulation results with measurements shows, that this combined approach is capable of accurately describing the sieving process at a reasonable computational cost

    The Epistemic Status of Processing Fluency as Source for Judgments of Truth

    This article combines findings from cognitive psychology on the role of processing fluency in truth judgments with epistemological theory on justification of belief. We first review evidence that repeated exposure to a statement increases the subjective ease with which that statement is processed. This increased processing fluency, in turn, increases the probability that the statement is judged to be true. The basic question discussed here is whether the use of processing fluency as a cue to truth is epistemically justified. In the present analysis, based on Bayes’ Theorem, we adopt the reliable-process account of justification presented by Goldman (1986) and show that fluency is a reliable cue to truth, under the assumption that the majority of statements one has been exposed to are true. In the final section, we broaden the scope of this analysis and discuss how processing fluency as a potentially universal cue to judged truth may contribute to cultural differences in commonsense beliefs

    Grey wolf genomic history reveals a dual ancestry of dogs

    The grey wolf (Canis lupus) was the first species to give rise to a domestic population, and they remained widespread throughout the last Ice Age when many other large mammal species went extinct. Little is known, however, about the history and possible extinction of past wolf populations or when and where the wolf progenitors of the present-day dog lineage (Canisfamiliaris) lived(1-8). Here we analysed 72 ancient wolf genomes spanning the last 100,000 years from Europe, Siberia and North America. We found that wolf populations were highly connected throughout the Late Pleistocene, with levels of differentiation an order of magnitude lower than they are today. This population connectivity allowed us to detect natural selection across the time series, including rapid fixation of mutations in the gene IFT8840,000-30,000 years ago. We show that dogs are overall more closely related to ancient wolves from eastern Eurasia than to those from western Eurasia, suggesting a domestication process in the east. However, we also found that dogs in the Near East and Africa derive up to half of their ancestry from a distinct population related to modern southwest Eurasian wolves, reflecting either an independent domestication process or admixture from local wolves. None of the analysed ancient wolf genomes is a direct match for either of these dog ancestries, meaning that the exact progenitor populations remain to be located.Peer reviewe

    Robust T cell immunity in convalescent individuals with asymptomatic or mild COVID-19

    SARS-CoV-2-specific memory T cells will likely prove critical for long-term immune protection against COVID-19. Here, we systematically mapped the functional and phenotypic landscape of SARS-CoV-2-specific T cell responses in unexposed individuals, exposed family members, and individuals with acute or convalescent COVID-19. Acute-phase SARS-CoV-2-specific T cells displayed a highly activated cytotoxic phenotype that correlated with various clinical markers of disease severity, whereas convalescent-phase SARS-CoV-2-specific T cells were polyfunctional and displayed a stem-like memory phenotype. Importantly, SARS-CoV-2-specific T cells were detectable in antibody-seronegative exposed family members and convalescent individuals with a history of asymptomatic and mild COVID-19. Our collective dataset shows that SARS-CoV-2 elicits broadly directed and functionally replete memory T cell responses, suggesting that natural exposure or infection may prevent recurrent episodes of severe COVID-19

Die KrankenhĂ€user und insbesondere das medizinische Personal sind bereits jetzt an ihren Grenzen und die GesundheitsĂ€mter ĂŒberlastet. Um die Kontrolle ĂŒber das Infektionsgeschehen zurĂŒckzuerlangen, empfiehlt die Nationale Akademie der Wissenschaften Leopoldina in der Ad-hoc-Stellungnahme „Coronavirus-Pandemie: Die Feiertage und den Jahreswechsel fĂŒr einen harten Lockdown nutzen“ ein zweistufiges Vorgehen. Die Rahmenbedingungen ‒ Weihnachtsferien in Bildungseinrichtungen und eingeschrĂ€nkter Betrieb in vielen Unternehmen und Behörden – bieten die Chance, in der EindĂ€mmung der Pandemie ein großes StĂŒck voranzukommen
