Design of a Software Tool Supporting Orientation in the Context of Digital Transformation
The digital transformation is a continuous and complex but indispensable endeavor. Because practice and academia regularly develop new digital technologies, companies face difficulties in selecting appropriate technologies to innovate their business model. Step-by-step instructions may guide companies to identify, evaluate, and select potential digitalization ideas. Software tools can support these activities in a structured way. Thus, the research objective of this paper is to design a software tool supporting the orientation phase of a company's digital transformation. To reach this objective, we conduct an action design research approach in cooperation with three companies. As an evaluation step, a focus group discussion with practitioners and end-users is carried out. The final tool design comprises 18 design principles that serve as the basis for a set of visual mockups and a prototype implementation of the intended tool

Child Sexual Abuse by Catholic Priests, Deacons, and Male Members of Religious Orders in the Authority of the German Bishops’ Conference 1946–2014
This study explores the extent of sexual abuse of minors by members of the Catholic Church in Germany. It is the first comprehensive study to examine this extent in a European country. The goals of this study are as follows: (a) to analyze whether the extent and characteristics of sexual abuse in a European country are comparable to those in the United States and Australia and (b) how discrepancies can be explained. The personnel files of 38,156 Catholic Priests, deacons, and male members of religious orders in the authority of the German Bishops’ Conference were analyzed. The study period lasted from 1946 to 2014. All 27 German dioceses took part in this study. A total of 4.4% of all clerics (N = 1,670) from 1946 to 2014 were alleged to have committed sexual abuse, and 3,677 children or adolescents were identified as victims. These results are similar to those from comparable studies in the United States. Sexual abuse of minors within the authority of the Catholic Church seems to be a worldwide phenomenon
Nuclear receptor NR2F6 inhibition potentiates responses to PD-L1/PD-1 cancer immune checkpoint blockade
Immune checkpoints blockade (ICB) is a viable anti-cancer strategy. Here the authors show that nuclear receptor NR2F6 acts as an immune checkpoint in T cells and, using mouse models and human T cells, they show NR2F6 inhibition might improve current ICB therapy or work as an alternative therapeutic strategy
Keeping health staff healthy: evaluation of a workplace initiative to reduce morbidity and mortality from HIV/AIDS in Malawi
ABSTRACT: BACKGROUND: In Malawi, the dramatic shortage of human resources for health is negatively impacted by HIV-related morbidity and mortality among health workers and their relatives. Many staff find it difficult to access HIV care through regular channels due to fear of stigma and discrimination. In 2006, two workplace initiatives were implemented in Thyolo District: a clinic at the district hospital dedicated to all district health staff and their first-degree relatives, providing medical services, including HIV care; and a support group for HIV-positive staff. METHODS: Using routine programme data, we evaluated the following outcomes up to the end of 2009: uptake and outcomes of HIV testing and counselling among health staff and their dependents; uptake and outcomes of antiretroviral therapy (ART) among health staff; and membership and activities of the support group. In addition, we included information from staff interviews and a job satisfaction survey to describe health workers' opinions of the initiatives. RESULTS: Almost two-thirds (91 of 144, 63%) of health workers and their dependents undergoing HIV testing and counselling at the staff clinic tested HIV positive. Sixty-four health workers had accessed ART through the staff clinic, approximately the number of health workers estimated to be in need of ART. Of these, 60 had joined the support group. Cumulative ART outcomes were satisfactory, with more than 90% alive on treatment as of June 2009 (the end of the study observation period). The availability, confidentiality and quality of care in the staff clinic were considered adequate by beneficiaries. CONCLUSIONS: Staff clinic and support group services successfully provided care and support to HIV-positive health workers. Similar initiatives should be considered in other settings with a high HIV prevalence
Reduction of Radiometric Miscalibration—Applications to Pushbroom Sensors
The analysis of hyperspectral images is an important task in Remote Sensing. Foregoing radiometric calibration results in the assignment of incident electromagnetic radiation to digital numbers and reduces the striping caused by slightly different responses of the pixel detectors. However, due to uncertainties in the calibration some striping remains. This publication presents a new reduction framework that efficiently reduces linear and nonlinear miscalibrations by an image-driven, radiometric recalibration and rescaling. The proposed framework—Reduction Of Miscalibration Effects (ROME)—considering spectral and spatial probability distributions, is constrained by specific minimisation and maximisation principles and incorporates image processing techniques such as Minkowski metrics and convolution. To objectively evaluate the performance of the new approach, the technique was applied to a variety of commonly used image examples and to one simulated and miscalibrated EnMAP (Environmental Mapping and Analysis Program) scene. Other examples consist of miscalibrated AISA/Eagle VNIR (Visible and Near Infrared) and Hawk SWIR (Short Wave Infrared) scenes of rural areas of the region Fichtwald in Germany and Hyperion scenes of the Jalal-Abad district in Southern Kyrgyzstan. Recovery rates of approximately 97% for linear and approximately 94% for nonlinear miscalibrated data were achieved, clearly demonstrating the benefits of the new approach and its potential for broad applicability to miscalibrated pushbroom sensor data
TREM2 expression in the brain and biological fluids in prion diseases
Triggering receptor expressed on myeloid cells 2 (TREM2) is an innate immune cell surface receptor that regulates microglial function and is involved in the pathophysiology of several neurodegenerative diseases. Its soluble form (sTREM2) results from shedding of the TREM2 ectodomain. The role of TREM2 in prion diseases, a group of rapidly progressive dementias remains to be elucidated. In the present study, we analysed the expression of TREM2 and its main sheddase ADAM10 in the brain of sporadic Creutzfeldt-Jakob disease (sCJD) patients and evaluated the role of CSF and plasma sTREM2 as a potential diagnostic marker of prion disease. Our data indicate that, compared to controls, TREM2 is increased in sCJD patient brains at the mRNA and protein levels in a regional and subtype dependent fashion, and expressed in a subpopulation of microglia. In contrast, ADAM10 is increased at the protein, but not the mRNA level, with a restricted neuronal expression. Elevated CSF sTREM2 is found in sCJD, genetic CJD with mutations E200K and V210I in the prion protein gene (PRNP), and iatrogenic CJD, as compared to healthy controls (HC) (AUC = 0.78-0.90) and neurological controls (AUC = 0.73-0.85), while CSF sTREM2 is unchanged in fatal familial insomnia. sTREM2 in the CSF of cases with Alzheimer's disease, and multiple sclerosis was not significantly altered in our series. CSF sTREM2 concentrations in sCJD are PRNP codon 129 and subtype-related, correlate with CSF 14-3-3 positivity, total-tau and YKL-40, and increase with disease progression. In plasma, sTREM2 is increased in sCJD compared with HC (AUC = 0.80), displaying positive correlations with plasma total-tau, neurofilament light, and YKL-40. We conclude that comparative study of TREM2 in brain and biological fluids of prion diseases reveals TREM2 to be altered in human prion diseases with a potential value in target engagement, patient stratification, and disease monitoring
