    From Linear Genes to Epigenetic Inheritance of Three-dimensional Epigenomes

    Fifty years ago Jacob and Monod reported their findings on the regulation of gene activity. Working on lambda bacteriophage lysogeny and the regulation of the production of an enzyme that cleaves lactose, they observed that its production was induced by the presence of lactose in the medium and came to general conclusions about gene expression that still hold true today. Thanks to decades of intense multidisciplinary research, these conclusions have been extended by several fundamental discoveries. In particular, gene regulatory circuits include the ability to propagate the memory of a specific gene regulatory state long after being established and even when the original inducer is no longer present. Furthermore, in addition to being regulated by binding of regulators such as RNAs or proteins in the vicinity of the site of transcription initiation, genes can be regulated by factor binding at incredible distances from their transcriptional start sites. Prominent among the regulatory components involved in these processes are Polycomb and Trithorax group proteins, pleiotropic gene regulators of critical importance in development, physiology and disease

    Discrete-time pilot model

    Pilot behavior is considered as a discrete-time process where the decision making has a sequential nature. This model differs from both the quasilinear model which follows from classical control theory and from the optimal control model which considers the human operator as a Kalman estimator-predictor. An additional factor considered is that the pilot's objective may not be adequately formulated as a quadratic cost functional to be minimized, but rather as a more fuzzy measure of the closeness with which the aircraft follows a reference trajectory. All model parameters, in the digital program simulating the pilot's behavior, were successfully compared in terms of standard-deviation and performance with those of professional pilots in IFR configuration. The first practical application of the model was in the study of its performance degradation when the aircraft model static margin decreases

    Discrete time modeling of heavy transport plane pilot behavior

    The desire to improve flight safety leads to a classification of various flight troubles in three groups: (1) troubles from sensitivity to flight disturbances, (2) maneuverability troubles (whenever a correction maneuver induces an unexpected deviation on another parameter), and (3) pilot troubles (pilot overload when required attention is excessive or underload entailing a loss of vigilance). Sensitivity to disturbances and maneuverability of a given aircraft may be evaluated from the early design stage. Evaluation of the pilot behavior, however, may be realized only in actual flight or with a flight simulator, that is quite late in the development period. For this reason, it is desirable to have available, at the design stage, a model of the pilot behavior to command the differential system describing the envisioned aircraft. This aim implies two major requirements. First, the program must be compatible with a wide range of possible aircraft designs; ideally, the program should be self-learning.     Harvesting systems for steep terrain in the Italian Alps: state of the art and future prospects

    Steep slope forest operations in Central Europe and in particular in the Alps are strongly related to the adoption of the cable-based harvesting system, even if innovative ground-based harvesting system, even if innovative ground-based harvesting systems have been proposed in the last years. In this context, the present works aim to acquire a thorough knowledge of yarding technologies used by the logging companies of the central Italian Alps, to evaluate their professionality in steep slope forest operations, and to predict the potential diffusion of innovative steep slope harvesting systems in the area. The results show a large number of logging companies (106) working with cable-based systems and in particular with four different standing skyline yarding technologies. The analysis of professionality in using cable cranes shows big differences between the companies. In particular, it identifies a consistent group of companies with a highly mechanized machinery fleet and high skills and experience in steep slope forest operations. These enterprises evidence a still limited potential diffusion of the innovative ground-based harvesting systems in the area, even if it is theoretically possible according to the GIS analysis of morphology and forest road networ

    Cellular memory is provided by two counteracting groups of chromatin proteins termed Trithorax group (TrxG) and Polycomb group (PcG) proteins. TrxG proteins activate transcription and are perhaps best known because of the involvement of the TrxG protein MLL in leukaemia. However, in terms of molecular analysis, they have lived in the shadow of their more famous counterparts, the PcG proteins. Recent advances have improved our understanding of TrxG protein function and demonstrated that the heterogeneous group of TrxG proteins is of critical importance in the epigenetic regulation of the cell cycle, senescence, DNA damage and stem cell biology
