    Studio Inside and Outside is a practice-led investigation into two educational studio sites in a UK University that has recently relocated its Art & Design Department to a newly renovated building. As the fine art and graphic design courses settle into their new studios it has become increasingly apparent how influential educational studios are. Each discipline demands a different kind of space. Each discipline uses space differently. Space and its models of occupation influence how students work, understand their discipline, how they learn, and how they engage. The studio ‘vernacular’ – the way it looks and sounds – not only encompasses the educational needs of the student, but is fused with visions of what professional life in that discipline might look like. The studio not only houses the daily activities that make up learning, but is a powerful part of student subjectivities and engagement

    Harnessing the power of BitTorrent for distributed denial-of-service attacks

    BitTorrent is a popular peer-to-peer (P2P) file-sharing protocol that utilizes a central server, known as a \u27tracker\u27, to coordinate connections between peers in a \u27swarm\u27, a term used to describe a Bit Torrent ad-hoc file sharing network. The tracker of a swarm is specified by the original file distributor and trusted unconditionally by peers in the swarm. This central point of control provides an opportunity for a file distributor to deploy a modified tracker to provide peers in a swarm with malicious coordination data, directing peer connection traffic toward an arbitrary target machine on an arbitrary service port. Although such an attack does not generate huge amount of attack traffic, it would set up many connections with the victim server successfully, which could cause serious denial-of-service by exhausting the victim server\u27s connection resource. In this paper, we present and demonstrate such an attack that is entirely tracker-based, requiring no modifications to Bit Torrent client software and could be deployed by an attacker right now. The results from both emulation and real-world experiments show the applicability of this attack. Due to the skyrocketing popularity of Bit Torrent and numerous large-scale swarms existed in the Internet, Bit Torrent swarms provide an intriguing platform for launching distributed denial-of-service (DDoS) attacks based on connection exhaustion.     This research explores the contemporary visibility of manufacturing processes, with a particular focus on the production of glass. The context for this study is the well documented sense of disconnection and estrangement said to result from our distance to and unfamiliarity with making processes. At the centre of the study is 'The Archive of Manufacture', an 'archival artwork' (Hal Foster) which has been especially collated for this research. The Archive responds to the question - how do we know how something is made - and gathers together 'points of visibility' - secondary sources where process is made visible, from industry, craft, popular culture, and press coverage. The study explores The Archive through three interrelated questions: Why, examines the social, political, and economic context in order to understand 'drivers' which affect the visibility of process, How, explores the formal and material aspects of the photograph, film, or object through which process is made visible, What is understood - investigates how this material contributes to an understanding of making process and how it shapes our understanding of objects. Initially, The Archive is explored in relation to a wide range of theoretical debates regarding our contemporary relationship with making process. This includes exploring the recent popular fascination with making (including Richard Sennett, Matthew Crawford), as well as descriptions of disconnection, alienation and invisibility (Karl Marx, James Heartfield, David Nye).Methods from art practice are employed as critical forms of looking to explore specific examples from The Archive. In particular, close reading as a 'meticulous visual analysis' (Shepard Steiner) is developed as a key method. The research expands this definition to include the written form of ekphrasis, and interrogative material and visual making processes.Through the production of the body of artworks, the research examines the contingency for the interpretation of the fragmentary or partial descriptions of process in The Archive, and the imagined or speculative understanding that results. The research identifies types of visibility and explores their limitations, and develops four key principles that describe how the visibility of process is effected and how it is subsequently understood. The key artworks offer a live experience, where the viewer's interpretation of the work mirrors the processes by which an understanding of process forms

    Strings as Solitons & Black Holes as Strings

    Supersymmetric closed string theories contain an infinite tower of BPS-saturated, oscillating, macroscopic strings in the perturbative spectrum. When these theories have dual formulations, this tower of states must exist nonperturbatively as solitons in the dual theories. We present a general class of exact solutions of low-energy supergravity that corresponds to all these states. After dimensional reduction they can be interpreted as supersymmetric black holes with a degeneracy related to the degeneracy of the string states. {}For example, in four dimensions we obtain a point-like solution which is asymptotic to a stationary, rotating, electrically-charged black hole with Regge-bounded angular momentum and with the usual ring-singularity replaced by a string source. This further supports the idea that the entropy of supersymmetric black holes can be understood in terms of counting of string states.     Initial Evaluation of the Effects of Aerosolized Florida Red Tide Toxins (Brevetoxins) in Persons with Asthma

    Florida red tides annually occur in the Gulf of Mexico, resulting from blooms of the marine dinoflagellate Karenia brevis. K. brevis produces highly potent natural polyether toxins, known as brevetoxins, that activate voltage-sensitive sodium channels. In experimental animals, brevetoxins cause significant bronchoconstriction. A study of persons who visited the beach recreationally found a significant increase in self-reported respiratory symptoms after exposure to aerosolized Florida red tides. Anecdotal reports indicate that persons with underlying respiratory diseases may be particularly susceptible to adverse health effects from these aerosolized toxins. Fifty-nine persons with physician-diagnosed asthma were evaluated for 1 hr before and after going to the beach on days with and without Florida red tide. Study participants were evaluated with a brief symptom questionnaire, nose and throat swabs, and spirometry approved by the National Institute for Occupational Safety and Health. Environmental monitoring, water and air sampling (i.e., K. brevis, brevetoxins, and particulate size distribution), and personal monitoring (for toxins) were performed. Brevetoxin concentrations were measured by liquid chromatography mass spectrometry, high-performance liquid chromatography, and a newly developed brevetoxin enzyme-linked immunosorbent assay. Participants were significantly more likely to report respiratory symptoms after Florida red tide exposure. Participants demonstrated small but statistically significant decreases in forced expiratory volume in 1 sec, forced expiratory flow between 25 and 75%, and peak expiratory flow after exposure, particularly those regularly using asthma medications. Similar evaluation during nonexposure periods did not significantly differ. This is the first study to show objectively measurable adverse health effects from exposure to aerosolized Florida red tide toxins in persons with asthma. Future studies will examine the possible chronic effects of these toxins among persons with asthma and other chronic respiratory impairment

    Type I insulin-like growth factor receptor over-expression induces proliferation and anti-apoptotic signaling in a three-dimensional culture model of breast epithelial cells

    INTRODUCTION: Activation of the type I insulin-like growth factor receptor (IGFIR) promotes proliferation and inhibits apoptosis in a variety of cell types. Transgenic mice expressing a constitutively active IGFIR or IGF-I develop mammary tumors and increased levels of IGFIR have been detected in primary breast cancers. However, the contribution of IGFIR activation in promoting breast cancer progression remains unknown. Mammary epithelial cell lines grown in three-dimensional cultures form acinar structures that mimic the round, polarized, hollow and growth-arrested features of mammary alveoli. We used this system to determine how proliferation and survival signaling by IGFIR activation affects breast epithelial cell biology and contributes to breast cancer progression. METHODS: Pooled, stable MCF-10A breast epithelial cells expressing wild-type IGFIR or kinase-dead IGFIR (K1003A) were generated using retroviral-mediated gene transfer. The effects of over-expression of wild-type or kinase-dead IGFIR on breast epithelial cell biology were analyzed by confocal microscopy of three-dimensional cultures. The contribution of signaling pathways downstream of IGFIR activation to proliferation and apoptosis were determined by pharmacological inhibition of phosphatidylinositol 3' kinase (PI3K) with LY294002, MAP kinase kinase (MEK) with UO126 and mammalian target of rapamycin (mTOR) with rapamycin. RESULTS: We found that MCF-10A cells over-expressing the IGFIR formed large, misshapen acinar structures with filled lumina and disrupted apico-basal polarization. This phenotype was ligand-dependent, occurring with IGF-I or supraphysiological doses of insulin, and did not occur in cells over-expressing the kinase-dead receptor. We observed increased proliferation, decreased apoptosis and increased phosphorylation of Ser(473 )of Akt and Ser(2448 )of mTOR throughout IGFIR structures. Inhibition of PI3K with LY294002 or MEK with UO126 prevented the development of acinar structures from IGFIR-expressing but not control cells. The mTOR inhibitor rapamycin failed to prevent IGFIR-induced hyperproliferation and survival signaling. CONCLUSION: Increased proliferation and survival signaling as well as loss of apico-basal polarity by IGFIR activation in mammary epithelial cells may promote early lesions of breast cancer. Three-dimensional cultures of MCF-10A cells over-expressing the IGFIR are a useful model with which to study the role of IGFIR signaling in breast cancer progression and for characterizing the effects of chemotherapeutics targeted to IGFIR signaling

    Mixed-species plantations of eucalyptus with nitrogen fixing trees: a review

    Mixed-species plantations of Eucalyptus with a nitrogen (N2) fixing species have the potential to increase productivity while maintaining soil fertility, compared to Eucalyptus monocultures. However, it is difficult to predict combinations of species and sites that will lead to these benefits. We review the processes and interactions occurring in mixed plantations, 5 and the influence of species or site attributes, to aid the selection of successful combinations of species and sites. Successful mixtures, where productivity is increased over that of monocultures, have often developed stratified canopies, such that the less shade-tolerant species overtops the more shadetolerant species. Successful mixtures also have significantly higher rates of N and P cycling than 10 Eucalyptus monocultures. It is therefore important to select N2-fixing species with readily decomposable litter and high rates of nutrient cycling, as well as high rates of N2-fixation. While the dynamics of N2-fixation in tree stands are not well understood, it appears as though eucalypts can benefit from fixed N as early as the first or second year following plantation establishment.     Functional antibody and T-cell immunity following SARS-CoV-2 infection, including by variants of concern, in patients with cancer: the CAPTURE study

    Patients with cancer have higher COVID-19 morbidity and mortality. Here we present the prospective CAPTURE study (NCT03226886) integrating longitudinal immune profiling with clinical annotation. Of 357 patients with cancer, 118 were SARS-CoV-2-positive, 94 were symptomatic and 2 patients died of COVID-19. In this cohort, 83% patients had S1-reactive antibodies, 82% had neutralizing antibodies against WT, whereas neutralizing antibody titers (NAbT) against the Alpha, Beta, and Delta variants were substantially reduced. Whereas S1-reactive antibody levels decreased in 13% of patients, NAbT remained stable up to 329 days. Patients also had detectable SARS-CoV-2-specific T cells and CD4+ responses correlating with S1-reactive antibody levels, although patients with hematological malignancies had impaired immune responses that were disease and treatment-specific, but presented compensatory cellular responses, further supported by clinical.
