    Knowledge and Attitudes of Population Towards HIV/Aids in Four States, Sudan

    Back ground: HIV/AIDS is a public health problem in the Sudan. The country is the most severely affected in North Africa and the Middle East with an estimated 500,000 people living with HIV/AIDS. The objectives of the study were to determine knowledge and attitude of the localpopulation in selected states about HIV/ AIDS and to determine the relation between knowledge and attitude towards HIV/AIDS and associated social factors.Methodology: Design was descriptive, cross-sectional, community- based study conducted in Gezira, Sinnar, River Nile and Northern states. The population was those found in their settings during the visits in April 2009. Both sexes were enrolled and children below the age of 15 years ofboth sexes were excluded from the study. Data were collected by a pre- coded and pre -tested questionnaire and were analyzed by the computer using SPSS, version 13, soft ware.Results: Knowledge of the population in the selected four states about HIV/AIDS was favorable (73%). Tolerant attitude towards HIV-infected people was low (40%). Results showed that males acquired better knowledge than females (76.2% versus 71.8%) but females were more tolerant towards HIV/AIDS than males (42% versus 37.4%). Results showed that HIV/AIDS Knowledge increased as the educational level increased. Level of knowledge was higher among university graduates (92.6%) followed by basic and secondary (76.7%). Illiterates acquired the least level of knowledge (42.4%). The study showed that tolerance of population towards PLWHA increased according to the level of education. The university graduated was more tolerant (55.1%) than thebasic/secondary (40.5%) and the illiterates (21.9%). It was shown that the single population was more tolerant towards PLWHA, followed by the married, the divorced and the widowed constituting 41.6%, 40.4%, 31.4% and 25% respectively.Conclusion: The study concluded that, level of knowledge about HIV/AIDS of the population in the selected states was good. Level of knowledge among males, the working force and the single population was higher than the females, those without jobs and the married. Level of HIV/AIDSknowledge increased according to the level of education, was higher among university graduates followed by basic / secondary and Illiterates. Attitude of the population in the selected states towards people living with HIV/AIDS “PLWHA” was low, only 40% had more tolerant attitude.Females, the highly educated and the working population were more tolerant towards HIV/AIDS than males, the less educated, and the nonworking population

    Neuroprotective properties of levosimendan in an in vitro model of traumatic brain injury

    <p>Abstract</p> <p>Background</p> <p>We investigated the neuroprotective properties of levosimendan, a novel inodilator, in an in vitro model of traumatic brain injury.</p> <p>Methods</p> <p>Organotypic hippocampal brain slices from mouse pups were subjected to a focal mechanical trauma. Slices were treated after the injury with three different concentrations of levosimendan (0.001, 0.01 and 0.1 μM) and compared to vehicle-treated slices. After 72 hrs, the trauma was quantified using propidium iodide to mark the injured cells.</p> <p>Results</p> <p>A significant dose-dependent reduction of both total and secondary tissue injury was observed in cells treated with either 0.01 or 0.1 μM levosimendan compared to vehicle-treated slices.</p> <p><b>Conclusion</b></p> <p>Levosimendan represents a promising new pharmacological tool for neuroprotection after brain injury and warrants further investigation in an in vivo model.</p

    Tension of knotted surgical sutures shows tissue specific rapid loss in a rodent model

    <p>Abstract</p> <p>Background</p> <p>Every surgical suture compresses the enclosed tissue with a tension that depends from the knotting force and the resistance of the tissue. The aim of this study was to identify the dynamic change of applied suture tension with regard to the tissue specific cutting reaction.</p> <p>Methods</p> <p>In rabbits we placed single polypropylene sutures (3/0) in skin, muscle, liver, stomach and small intestine. Six measurements for each single organ were determined by tension sensors for 60 minutes. We collected tissue specimens to analyse the connective tissue stability by measuring the collagen/protein content.</p> <p>Results</p> <p>We identified three phases in the process of suture loosening. The initial rapid loss of the first phase lasts only one minute. It can be regarded as cutting through damage of the tissue. The percentage of lost tension is closely related to the collagen content of the tissue (r = -0.424; p = 0.016). The second phase is characterized by a slower decrease of suture tension, reflecting a tissue specific plastic deformation. Phase 3 is characterized by a plateau representing the remaining structural stability of the tissue. The ratio of remaining tension to initial tension of phase 1 is closely related to the collagen content of the tissue (r = 0.392; p = 0.026).</p> <p>Conclusions</p> <p>Knotted non-elastic monofilament sutures rapidly loose tension. The initial phase of high tension may be narrowed by reduction of the surgeons' initial force of the sutures' elasticity to those of the tissue. Further studies have to confirm, whether reduced tissue compression and less local damage permits improved wound healing.</p

    TCRep 3D: An Automated In Silico Approach to Study the Structural Properties of TCR Repertoires

    TCRep 3D is an automated systematic approach for TCR-peptide-MHC class I structure prediction, based on homology and ab initio modeling. It has been considerably generalized from former studies to be applicable to large repertoires of TCR. First, the location of the complementary determining regions of the target sequences are automatically identified by a sequence alignment strategy against a database of TCR Vα and Vβ chains. A structure-based alignment ensures automated identification of CDR3 loops. The CDR are then modeled in the environment of the complex, in an ab initio approach based on a simulated annealing protocol. During this step, dihedral restraints are applied to drive the CDR1 and CDR2 loops towards their canonical conformations, described by Al-Lazikani et. al. We developed a new automated algorithm that determines additional restraints to iteratively converge towards TCR conformations making frequent hydrogen bonds with the pMHC. We demonstrated that our approach outperforms popular scoring methods (Anolea, Dope and Modeller) in predicting relevant CDR conformations. Finally, this modeling approach has been successfully applied to experimentally determined sequences of TCR that recognize the NY-ESO-1 cancer testis antigen. This analysis revealed a mechanism of selection of TCR through the presence of a single conserved amino acid in all CDR3β sequences. The important structural modifications predicted in silico and the associated dramatic loss of experimental binding affinity upon mutation of this amino acid show the good correspondence between the predicted structures and their biological activities. To our knowledge, this is the first systematic approach that was developed for large TCR repertoire structural modeling

    Mammary epithelial cell transformation: insights from cell culture and mouse models

    Normal human mammary epithelial cells (HMECs) have a finite life span and do not undergo spontaneous immortalization in culture. Critical to oncogenic transformation is the ability of cells to overcome the senescence checkpoints that define their replicative life span and to multiply indefinitely – a phenomenon referred to as immortalization. HMECs can be immortalized by exposing them to chemicals or radiation, or by causing them to overexpress certain cellular genes or viral oncogenes. However, the most efficient and reproducible model of HMEC immortalization remains expression of high-risk human papillomavirus (HPV) oncogenes E6 and E7. Cell culture models have defined the role of tumor suppressor proteins (pRb and p53), inhibitors of cyclin-dependent kinases (p16(INK4a), p21, p27 and p57), p14(ARF), telomerase, and small G proteins Rap, Rho and Ras in immortalization and transformation of HMECs. These cell culture models have also provided evidence that multiple epithelial cell subtypes with distinct patterns of susceptibility to oncogenesis exist in the normal mammary tissue. Coupled with information from distinct molecular portraits of primary breast cancers, these findings suggest that various subtypes of mammary cells may be precursors of different subtypes of breast cancers. Full oncogenic transformation of HMECs in culture requires the expression of multiple gene products, such as SV40 large T and small t, hTERT (catalytic subunit of human telomerase), Raf, phosphatidylinositol 3-kinase, and Ral-GEFs (Ral guanine nucleotide exchange factors). However, when implanted into nude mice these transformed cells typically produce poorly differentiated carcinomas and not adenocarcinomas. On the other hand, transgenic mouse models using ErbB2/neu, Ras, Myc, SV40 T or polyomavirus T develop adenocarcinomas, raising the possibility that the parental normal cell subtype may determine the pathological type of breast tumors. Availability of three-dimensional and mammosphere models has led to the identification of putative stem cells, but more studies are needed to define their biologic role and potential as precursor cells for distinct breast cancers. The combined use of transformation strategies in cell culture and mouse models together with molecular definition of human breast cancer subtypes should help to elucidate the nature of breast cancer diversity and to develop individualized therapies

    Resolving the strange behavior of extraterrestrial potassium in the upper atmosphere

    It has been known since the 1960s that the layers of Na and K atoms, which occur between 80 and 105 km in the Earth's atmosphere as a result of meteoric ablation, exhibit completely different seasonal behavior. In the extratropics Na varies annually, with a pronounced wintertime maximum and summertime minimum. However, K varies semiannually with a small summertime maximum and minima at the equinoxes. This contrasting behavior has never been satisfactorily explained. Here we use a combination of electronic structure and chemical kinetic rate theory to determine two key differences in the chemistries of K and Na. First, the neutralization of K+ ions is only favored at low temperatures during summer. Second, cycling between K and its major neutral reservoir KHCO3 is essentially temperature independent. A whole atmosphere model incorporating this new chemistry, together with a meteor input function, now correctly predicts the seasonal behavior of the K layer

    An evaluation of corporate social responsibility communication on the websites of telecommunication companies operating in Ghana: Impression management perspectives

Similarly, corporate websites have been used to manage stakeholders’ impressions about the organization. Meanwhile, CSR by firms have been criticized for been a manipulative tactics used by firms. The purpose of this paper therefore is to ascertain how telecommunication companies operating in Ghana communicate CSR on their corporate websites. Design/methodology/approach: This study used a qualitative content analysis technique. It also used Bolino et al.’s (2008) impression management framework as the conceptual framework. Data were extracted from the websites of four telecommunication companies operating in Ghana. Data were analyzed using thematic analysis techniques. Findings: The results show that the companies use impression management in communicating CSR on their corporate websites. Account, assertive impression management; blaring, boasting, defensive impression management; demonstrative impression management; exemplification, favor-rendering, illustrative impression management; ingratiation, other-enhancement, other-focused impression management; self-enhancement, self-focused impression management; and self-promotion tactics are tactics prevalent in the CSR communication of the companies. Actor-to-actor link impression management and other emotional appeal also emerged as some of the impression management tactics used by the companies. Originality/value: This provides insights into how organizations use impression management in their CSR communication on their corporate websites. Studies of this nature are limited. Again, studies specifically adapting Bolino et al.’s impression management to study CSR communication on corporate websites are rare

    Examining students’ satisfaction with online learning during the Covid-19 pandemic - an extended UTAUT2 approach

    Given that the educational sector was particularly hard hit in most countries around the world during the Covid-19 pandemic, the study employs an extended UTAUT2 to investigate student use and satisfaction with e-learning in a developing country setting. We employed partial least squares structural equation modelling (PLS-SEM) on 616 data responses gathered online from university students during the pandemic to evaluate our integrated model. This paper found that not all the predictors of the UTAUT2 model were statistically significant in predicting behavioural intention. Our results revealed that performance expectancy, effort expectancy, and facilitating conditions, as predictors, have no influence on various outcomes relating to the behavioural intention to use e-learning. The study also found that e-learning interaction quality predicts student satisfaction. Other theoretical and practical implications are also detailed in our study
