    µ-Particle tracking velocimetry and computational fluid dynamics study of cell seeding within a 3D porous scaffold

    Cell seeding of 3D scaffolds is a critical step in tissue engineering since the final tissue properties are related to the initial cell distribution and density within the scaffold unit. Perfusion systems can transport cells to the scaffold however; the fact that cells flow inside the scaffold pores does not guarantee cell deposition onto the scaffold substrate and cell attachment. The aim of this study was to investigate how fluid flow conditions modulate cell motion and deposition during perfusion. For such a purpose, a multiphase-based computational fluid dynamics (CFD) model was developed in conjunction with particle tracking velocimetry experiments (PTV) which for the first time were applied to observe cell seeding inside a 3D scaffold. CFD and PTV results showed the strong effect of gravity for lower flow rates leading to cell sedimentation and poor transport of cells to the scaffold. Higher flow rates help overcome the effect of gravity so more cells travelling inside the scaffold were found. Nonetheless, fluid flow drags cells along the fluid streamlines without intercepting the scaffold substrate. As a consequence, if cells do not deposit into the scaffold substrate, cell adhesion cannot occur. Therefore, cell-scaffold interception should be promoted and the present computational model which predicts the effect of gravity and fluid drag on cells trajectories could serve to optimise bioreactors and enhance cell seeding efficiency

    The Herschel Virgo Cluster Survey - XIII. Dust in early-type galaxies

    Aims. We study the dust content of a large optical input sample of 910 early-type galaxies (ETG) in the Virgo cluster, extending also to the dwarf ETG, and examine the results in relation with those on the other cold ISM components. Methods. We searched for far-infrared emission in all galaxies of the input sample using the 250 micron image of the Herschel Virgo Cluster Survey (HeViCS). This image covers a large fraction of the cluster. For the detected ETG we measured fluxes in 5 bands from 100 to 500 micron, and estimated the dust mass and temperature with modified black-body fits. Results. Dust is detected above the completeness limit of 25.4 mJy at 250 micron in 46 ETG, 43 of which are in the optically complete part of the input sample. In addition dust is present at fainter levels in another 6 ETG. We detect dust in the 4 ETG with synchrotron emission, including M 87. Dust appears to be much more concentrated than stars and more luminous ETG have higher dust temperatures. Dust detection rates down to the 25.4 mJy limit are 17% for ellipticals, about 40% for lenticulars (S0 + S0a) and around 3% for dwarf ETG. Dust mass does not correlate clearly with stellar mass and is often much more than that expected for a passive galaxy in a closed-box model. The dust-to-stars mass ratio anticorrelates with galaxy luminosity, and for some dwarf ETG reaches values as high as for dusty late-type galaxies. In the Virgo cluster slow rotators appear more likely to contain dust than fast ones. Comparing the dust results with those on HI from ALFALFA, there are only 8 ETG detected both in dust and in HI in the HeViCS area; 39 have dust but only an upper limit on HI, and 8 have HI but only an upper limit on dust. The locations of these galaxies in the cluster are different, with the dusty ETG concentrated in the densest regions, while the HI rich ETG are at the periphery.Comment: Accepted by Astronomy and Astrophysics; modified to reflect the on-line forthcoming version on the A&A web sit

    Physiological and transcriptomic evaluation of drought effect on own-rooted and grafted grapevine rootstock (1103P and 101-14MGt)

    Grapevines worldwide are grafted onto Vitis spp. rootstocks in order to improve their tolerance to biotic and abiotic stresses. Thus, the response of vines to drought is the result of the interaction between the scion variety and the rootstock genotype. In this work, the responses of genotypes to drought were evaluated on 1103P and 101-14MGt plants, own-rooted and grafted with Cabernet Sauvignon, in three different water deficit conditions (80, 50, and 20% soil water content, SWC). Gas exchange parameters, stem water potential, root and leaf ABA content, and root and leaf transcriptomic response were investigated. Under well-watered conditions, gas exchange and stem water potential were mainly affected by the grafting condition, whereas under sever water deficit they were affected by the rootstock genotype. Under severe stress conditions (20% SWC), 1103P showed an "avoidance" behavior. It reduced stomatal conductance, inhibited photosynthesis, increased ABA content in the roots, and closed the stomata. The 101-14MGt maintained a high photosynthetic rate, limiting the reduction of soil water potential. This behavior results in a "tolerance" strategy. An analysis of the transcriptome showed that most of the differentially expressed genes were detected at 20% SWC, and more significantly in roots than in leaves. A core set of genes has been highlighted on the roots as being related to the root response to drought that are not affected by genotype nor grafting. Genes specifically regulated by grafting and genes specifically regulated by genotype under drought conditions have been identified as well. The 1103P, more than the 101-14MGt, regulated a high number of genes in both own-rooted and grafted conditions. This different regulation revealed that 1103P rootstock readily perceived the water scarcity and rapidly faced the stress, in agreement with its avoidance strateg

    Artificial neural network analysis of bone quality DXA parameters response to teriparatide in fractured osteoporotic patients

    Teriparatide is a bone-forming therapy for osteoporosis that increases bone quantity and texture, with uncertain action on bone geometry. No data are available regarding its influence on bone strain. To investigate teriparatide action on parameters of bone quantity and quality and on Bone Strain Index (BSI), also derived from DXA lumbar scan, based on the mathematical model finite element method. Forty osteoporotic patients with fractures were studied before and after two years of daily subcutaneous 20 mcg of teriparatide with dual X-ray photon absorptiometry to assess bone mineral density (BMD), hip structural analysis (HSA), trabecular bone score (TBS), BSI. Spine deformity index (SDI) was calculated from spine X-ray. Shapiro-Wilks, Wilcoxon and Student's t test were used for classical statistical analysis. Auto Contractive Map was used for Artificial Neural Network Analysis (ANNs). In the entire population, the ameliorations after therapy regarded BSI (-13.9%), TBS (5.08%), BMD (8.36%). HSA parameters of femoral shaft showed a worsening. Dividing patients into responders (BMD increase >10%) and non-responders, the first presented TBS and BSI ameliorations (11.87% and -25.46%, respectively). Non-responders presented an amelioration of BSI only, but less than in the other subgroup (-6.57%). ANNs maps reflect the mentioned bone quality improvements. Teriparatide appears to ameliorate not only BMD and TBS, but also BSI, suggesting an increase of bone strength that may explain the known reduction in fracture risk, not simply justified by BMD increase. BSI appears to be a sensitive index of TPD effect. ANNs appears to be a valid tool to investigate complex clinical systems

    Nearby early-type galaxies with ionized gas. The UV emission from GALEX observations

    We present GALEX far-ultraviolet (FUV, λeff\lambda_{eff}=1538 \AA) and near-ultraviolet (NUV, λeff\lambda_{eff}=2316 \AA) surface photometry of 40 early-type galaxies (ETGs) selected from a wider sample of 65 nearby ETGs showing emission lines in their optical spectra. We derive FUV and NUV surface brightness profiles, (FUV-NUV) colour profiles and D25_{25} integrated magnitudes. We extend the photometric study to the optical {\it r} band from SDSS imaging for 14 of these ETGs. In general, the (FUV-NUV) radial colour profiles become redder with galactocentric distance in both rejuvenated (4\leq 4 Gyr) and old ETGs. Colour profiles of NGC 1533, NGC 2962, NGC 2974, NGC 3489, and IC 5063 show rings and/or arm-like structures, bluer than the body of the galaxy, suggesting the presence of recent star formation. Although seven of our ETGs show shell systems in their optical image, only NGC 7135 displays shells in the UV bands. We characterize the UV and optical surface brightness profiles, along the major axis, using a Sersic law. The Sersic law exponent, nn, varies from 1 to 16 in the UV bands. S0 galaxies tend to have lower values of nn (5\leq5). The Sersic law exponent n=4n=4 seems to be a watershed: ETGs with n>4n>4 tend to have [α\alpha/Fe] greater than 0.15, implying a short star-formation time scale. We find a significant correlation between the FUV-NUV colour and central velocity dispersions σ\sigma, with the UV colours getting bluer at larger σ\sigma. This trend is likely driven by a combined effect of `downsizing' and of the mass-metallicity relation.Comment: Accepted for publication in MNRAS, 33 pages, 7 figure

    The far-infrared view of M87 as seen by the Herschel Space Observatory

    The origin of the far-infrared emission from the nearby radio galaxy M87 remains a matter of debate. Some studies find evidence of a far-infrared excess due to thermal dust emission, whereas others propose that the far-infrared emission can be explained by synchrotron emission without the need for an additional dust emission component. We observed M87 with PACS and SPIRE as part of the Herschel Virgo Cluster Survey (HeViCS). We compare the new Herschel data with a synchrotron model based on infrared, submm and radio data to investigate the origin of the far-infrared emission. We find that both the integrated SED and the Herschel surface brightness maps are adequately explained by synchrotron emission.     Occurrence of mesothelial/monocytic incidental cardiac excrescences in material from open-heart procedures: case reports and literature review

    Mesothelial/monocytic incidental cardiac excrescences (MICE) are unusual findings during the histological analysis of material from the pericardium, mediastinum, or other tissues collected in open-heart surgery. Despite their somewhat worrisome histological appearance, they show a benign clinical course, and further treatment is virtually never necessary. Hence, the importance of recognizing the entity relays in its differential diagnosis, as an unaware medical pathologist may misinterpret it for a malignant neoplasm. Other mesothelial and histiocytic proliferative lesions, sharing very close histological morphology and immunohistochemistry features with MICE, have been described in sites other than the heart or the mediastinum. This similarity has led to the proposal of the common denomination “histiocytosis with raisinoid nuclei.” We report three cases from the pathology archives of the Heart Institute of São Paulo University (Incor/HC-FMUSP), diagnosed as “mesothelial/monocytic incidental cardiac excrescence,” with immunohistochemical documentation, and provide a literature review of this entity

    The Herschel Virgo Cluster Survey: I. Luminosity functions

    We describe the Herschel Virgo Cluster Survey (HeViCS) and the first data obtained as part of the Science Demonstration Phase (SDP). The data cover a central 4x4 sq deg region of the cluster. We use SPIRE and PACS photometry data to produce 100, 160, 250, 350 and 500 micron luminosity functions (LFs) for optically bright galaxies that are selected at 500 micron and detected in all bands. We compare these LFs with those previously derived using IRAS, BLAST and Herschel-ATLAS data. The Virgo Cluster LFs do not have the large numbers of faint galaxies or examples of very luminous galaxies seen previously in surveys covering less dense environments.Comment: Letter accepted for publication in A&A (Herschel special issue
