    This is the context in which this research paper is written though its goal is much more modest. We do not hope to settle the question of the intent of our founding fathers. But we want to try to determine whether the churches were at all involved in this movement toward religious freedom. Were they in favor of a separation between church and state, or did they only seek to end preference? Or were they not involved at all? At the same time, we also want to examine the various factors responsible for the first amendment and to see what they tell us about the intent of the fathers

    Can Beach Cleans Do More Than Clean-Up Litter? Comparing Beach Cleans to Other Coastal Activities

    Coastal visits not only provide psychological benefits but can also contribute to the accumulation of rubbish. Volunteer beach cleans help address this issue, but may only have limited, local impact. Consequently, it is important to study any broader benefits associated with beach cleans. This article examines the well-being and educational value of beach cleans, as well as their impacts on individuals’ behavioral intentions. We conducted an experimental study that allocated students (n = 90) to a beach cleaning, rock pooling, or walking activity. All three coastal activities were associated with positive mood and pro-environmental intentions. Beach cleaning and rock pooling were associated with higher marine awareness. The unique impacts of beach cleaning were that they were rated as most meaningful but linked to lower restorativeness ratings of the environment compared with the other activities. This research highlights the interplay between environment and activities, raising questions for future research on the complexities of person-environment interaction

    The aim of this study is to find information about what to do when riding arena footings are worn-out. Especially, the surface layer of riding arena footings is very interesting because of its importance of giving grip in every step for the horse, as well as shock ab-sorption. Generally, surface layers are containing several different parts in order to achieve desirable properties. These different parts can consist of organic material or in-organic material (mostly gravel). Gravel and many other materials in the top-layer are environmental friendly, and therefore, easy to get rid of. However, when mixing envi-ronmental unfriendly material with gravel it is not that easy to get rid of in a non-polluting way. To get information, interviews with individuals, companies and some municipalities were made. The literature about used riding arena footing materials is rare, contrary how to make a good footing. The municipalities use the Swedish envi-ronmental law regarding deposition of used material; consequently, some part of the environmental law had to be used in the study in order to compare the results giving from the municipalities. The answers from individuals and companies were mostly about why and how they thought when they bought used material. What they would to do when the riding arena footing material is worn-out; they did not know. Among other things, it is concluded that used but not worn-out footing materials has an aftermarket. Further, as a user, one has a responsibility to know what the footing mate-rial contains; if there are some materials that can be harmful to the environment. When getting rid of the material, talking with the municipality is recommended. The munici-palities always handle every case individually. If the material is totally friendly to the environment, you can take care of it on your own way at home

    Cada uno de nosotros en todo el mundo, en casi todas las actividades que realizamos a diario, generamos residuos que pueden ser o no ser reutilizables. Es casi inevitable generar residuos, por lo mismo es muy importante hacernos responsables de esta contaminación al medio ambiente, que por consecuencia también puede causar daños a nuestra salud. Teniendo en cuenta las consecuencias de la contaminación es que en el campamento lote 57 tiene implementado los tachos de diferentes colores en lugares específicos y a su vez el compromiso por las empresas que prestan servicios, también se cuenta con un almacén de residuos de la cual se encarga Megapack Trading S.A.C., donde se recepciona residuos tanto peligrosos como no peligrosos, estos residuos son generados en el campamento, lugares de exploración y plantas, por este mismo compromiso de mejorar la calidad y preservación del medio ambiente las empresas tienen conocimiento de llevar sus residuos en el color de bolsa correspondiente y debidamente rotulados para que sea más fácil la disposición o comercialización. Pero debido a nuevas empresas que han ganado licitaciones y a su cambio de personal, se ha comenzado a ver la mala segregación y falta de rotulación de los residuos que ingresan al almacén, las cuales trae consecuencias, como el aumento de residuos que van a relleno sanitario peligrosos y no peligrosos, y a la comercialización de residuos que se pueden reutilizar como cartón, papel, botellas de plástico, estos vienen combinados o manchados con diversos residuos los cuales ya no son aptos para nuestros clientes que compran estos, debido a este problema nace el análisis económico de cuanto se está perdiendo llevando los residuos a un relleno y cuanto se podría ahorrar realizando una buena segregación y recuperando un porcentaje de lo gastado siendo un costo de oportunidad para empresa.Trabajo de suficiencia profesiona

    Detection of TAR DNA-binding protein 43 (TDP-43) oligomers as initial intermediate species during aggregate formation

    Aggregates of the RNA-binding protein TDP-43 (TAR DNAbinding protein) are a hallmark of the overlapping neurodegenerative disorders amyotrophic lateral sclerosis (ALS) and frontotemporal dementia. The process of TDP-43 aggregation remains poorly understood, and whether it includes formation of intermediate complexes is unknown. Here, we analyzed aggregates derived from purified TDP-43 under semidenaturing conditions, identifying distinct oligomeric complexes at the initial time points before the formation of large aggregates. We found that this early oligomerization stage is primarily driven by TDP-43’s RNA-binding region. Specific binding to GU-rich RNA strongly inhibited both TDP-43 oligomerization and aggregation, suggesting that RNA interactions are critical for maintaining TDP-43 solubility. Moreover, we analyzed TDP-43 liquid–liquid phase separation and detected similar detergentresistant oligomers upon maturation of liquid droplets into solid-like fibrils. These results strongly suggest that the oligomers form during the early steps of TDP-43 misfolding. Importantly, the ALS-linked TDP-43 mutations A315T and M337V significantly accelerate aggregation, rapidly decreasing the monomeric population and shortening the oligomeric phase. We also show that aggregates generated from purified TDP-43 seed intracellular aggregation detected by established TDP-43 pathology markers. Remarkably, cytoplasmic aggregate seeding was detected earlier for the A315T and M337V variants and was 50% more widespread than forWTTDP-43 aggregates.We provide evidence for aninitial step of TDP-43 self-assembly into intermediate oligomeric complexes, whereby these complexes may provide a scaffold for aggregation. This process is altered by ALS-linked mutations, underscoring the role of perturbationsin TDP-43 homeostasisin protein aggregation and ALS-FTD pathogenesis

    El presente estudio se ha realizado con el propósito, determinar si “Un plan promocional basado en el diagnóstico situacional de mercado contribuirá a incrementar las ventas de Inversiones Coquito S.A.C del distrito de Trujillo”. Se utilizó el diseño lineal de resultados con observación antes y después, empleando la técnica de la encuesta, utilizando un solo cuestionario antes de la aplicación del plan promocional para la recolección de datos y el análisis de las ventas del mes de mayo del 2014 con relación al mes de mayo del 2015. Se consideró como muestra representativa al segmento “C” que frecuentan los establecimientos de cebicherias del distrito de Trujillo, conformada por un total de 284 personas a encuestar. Asimismo, se compararon las ventas de mayo del 2014 con relación al mes de mayo del 2015 para determinar si se incrementaron las ventas luego de aplicada el plan promocional. Los resultados de la investigación han permitido analizar el nivel de ventas antes y después de la aplicación del plan promocional, asimismo poder determinar los puntos débiles de la empresa para promocionar sus productos, con el fin de elaborar una propuesta para fortalecer el reconocimiento de la marca. De ésta manera se ha logrado determinar a través del diagnóstico situacional de mercado, que el nivel de promoción de la Cebichería Coquito es bajo, porque no hay mucho interés en invertir en marketing para fortalecer el reconocimiento de la cebichería Coquito. Esto genera bajas ventas o un mismo estándar de ventas, sin oportunidad a que estas se vayan incrementando y también un bajo posicionamiento en el mercado.This study was developed to determine whether "A promotional plan based on the market situation will help to increase sales of “Coquito Inversiones SAC” of Trujillo district." We used the linear design with observation results before and after using the survey technique, also we used an only questionnaire before of the application of the promotional plan for data collection and analysis of sales in May 2014 compared to May 2015. It was considered as representative sample to segment "C" who frequent to establishments of sea food at Trujillo district, comprising a total of 284 people surveyed. In addition, sales in May 2014 were compared in relation to May of 2015 to determine whether sales applied after increased of the promotional plan. The results of the research have allowed us to analyze the level of sales before and after the application of the promotional plan, also to determine the weak points of the company to promote their products, to develop a proposal to strengthen the brand recognition. In this way it has been determined through the market situation assessment that the level of promotion of sea food Coquito is low, because there is much interest in investing in marketing to strengthen the recognition of the Marine restaurant called “Coquito”. This generates lower sales or sales the same standard, without opportunity to increase sales and also a low position in the market.Tesi
