    On the refractive index for a nonmagnetic two-component medium: resolution of a controversy

    The refractive index of a dielectric medium comprising both passive and inverted components in its permittivity was determined using two methods: (i) in the time domain, a finite-difference algorithm to compute the frequency-domain reflectance from reflection data for a pulsed plane wave that is normally incident on a dielectric half-space, and (ii) in the frequency domain, the deflection of an obliquely incident Gaussian beam on transmission through a dielectric slab. The dielectric medium was found to be an active medium with a negative real part for its refractive index. Thereby, a recent controversy in the scientific literature was resolved.Comment: manuscript submitted to Optics Communication

    Two choices are possible for the refractive index of a linear, homogeneous, isotropic, active, dielectric material. Either of the choices is adequate for obtaining frequency-domain solutions for (i) scattering by slabs, spheres, and other objects of bounded extent; (ii) guided-wave propagation in homogeneously filled, cross-sectionally uniform, straight waveguide sections with perfectly conducting walls; and (iii) image formation due to flat lenses. The correct choice does matter for the half-space problem, but that problem is not realistic

    Modelling the impact of forest design plans on an endangered mammal species: The eurasian red squirrel

National measures to save the species include large conifer forest reserves where management encompasses measures to bolster the native species. However, forests are multipurpose environments and foresters have to balance different timber production, amenity and conservation objectives. We present a mathematical modelling framework that examines the impacts of potential felling and restocking plans for two reserves, Kidland and Uswayford forests, in northern England. In collaboration with forest managers, we employed an iterative process that used the model to assess four forest design plans (felling and restocking scenarios) with the aim of improving red squirrel population viability. Overall, the model predicted that extinction in both forests at the same time was rare, but high in Uswayford (84%) alone. Survival could be drastically increased (from 16% to 70%) by felling and restocking adjustments, and improving dispersal between the two adjacent forests. This study provides an exemplar of how modelling can have a direct input to land management to help managers objectively balance the differing pressures of multipurpose forestry.AW, MB and PL are supported in part by a NERC Innovations grant NE/M021319/1

    Characterization of the insulin sensitivity of ghrelin receptor KO mice using glycemic clamps

    <p>Abstract</p> <p>Background</p> <p>We and others have demonstrated previously that ghrelin receptor (<it>GhrR</it>) knock out (KO) mice fed a high fat diet (HFD) have increased insulin sensitivity and metabolic flexibility relative to WT littermates. A striking feature of the HFD-fed <it>GhrR </it>KO mouse is the dramatic decrease in hepatic steatosis. To characterize further the underlying mechanisms of glucose homeostasis in <it>GhrR </it>KO mice, we conducted both hyperglycemic (HG) and hyperinsulinemic-euglycemic (HI-E) clamps. Additionally, we investigated tissue glucose uptake and specifically examined liver insulin sensitivity.</p> <p>Results</p> <p>Consistent with glucose tolerance-test data, in HG clamp experiments, <it>GhrR </it>KO mice showed a reduction in glucose-stimulated insulin release relative to WT littermates. Nevertheless, a robust 1<sup>st </sup>phase insulin secretion was still achieved, indicating that a healthy β-cell response is maintained. Additionally, <it>GhrR </it>KO mice demonstrated both a significantly increased glucose infusion rate and significantly reduced insulin requirement for maintenance of the HG clamp, consistent with their relative insulin sensitivity. In HI-E clamps, both LFD-fed and HFD-fed <it>GhrR </it>KO mice showed higher peripheral insulin sensitivity relative to WT littermates as indicated by a significant increase in insulin-stimulated glucose disposal (Rd), and decreased hepatic glucose production (HGP). HFD-fed <it>GhrR </it>KO mice showed a marked increase in peripheral tissue glucose uptake in a variety of tissues, including skeletal muscle, brown adipose tissue and white adipose tissue. <it>GhrR </it>KO mice fed a HFD also showed a modest, but significant decrease in conversion of pyruvate to glucose, as would be anticipated if these mice displayed increased liver insulin sensitivity. Additionally, the levels of UCP2 and UCP1 were reduced in the liver and BAT, respectively, in <it>GhrR </it>KO mice relative to WT mice.</p> <p>Conclusions</p> <p>These results indicate that improved glucose homeostasis of <it>GhrR </it>KO mice is characterized by robust improvements of glucose disposal in both normal and metabolically challenged states, relative to WT controls. <it>GhrR </it>KO mice have an intact 1<sup>st </sup>phase insulin response but require significantly less insulin for glucose disposal. Our experiments reveal that the insulin sensitivity of <it>GhrR </it>KO mice is due to both BW independent and dependent factors. We also provide several lines of evidence that a key feature of the <it>GhrR </it>KO mouse is maintenance of hepatic insulin sensitivity during metabolic challenge.</p

    Hemodialysis in children: general practical guidelines

    Over the past 20 years children have benefited from major improvements in both technology and clinical management of dialysis. Morbidity during dialysis sessions has decreased with seizures being exceptional and hypotensive episodes rare. Pain and discomfort have been reduced with the use of chronic internal jugular venous catheters and anesthetic creams for fistula puncture. Non-invasive technologies to assess patient target dry weight and access flow can significantly reduce patient morbidity and health care costs. The development of urea kinetic modeling enables calculation of the dialysis dose delivery, Kt/V, and an indirect assessment of the intake. Nutritional assessment and support are of major importance for the growing child. Even if the validity of these “urea only” data is questioned, their analysis provides information useful for follow-up. Newer machines provide more precise control of ultrafiltration by volumetric assessment and continuous blood volume monitoring during dialysis sessions. Buffered bicarbonate solutions are now standard and more biocompatible synthetic membranes and specific small size material dialyzers and tubing have been developed for young infants. More recently, the concept of “ultrapure” dialysate, i.e. free from microbiological contamination and endotoxins, has developed. This will enable the use of hemodiafiltration, especially with the on-line option, which has many theoretical advantages and should be considered in the case of maximum/optimum dialysis need. Although the optimum dialysis dose requirement for children remains uncertain, reports of longer duration and/or daily dialysis show they are more effective for phosphate control than conventional hemodialysis and should be considered at least for some high-risk patients with cardiovascular impairment. In children hemodialysis has to be individualized and viewed as an “integrated therapy” considering their long-term exposure to chronic renal failure treatment. Dialysis is seen only as a temporary measure for children compared with renal transplantation because this enables the best chance of rehabilitation in terms of educational and psychosocial functioning. In long term chronic dialysis, however, the highest standards should be applied to these children to preserve their future “cardiovascular life” which might include more dialysis time and on-line hemodiafiltration with synthetic high flux membranes if we are able to improve on the rather restricted concept of small-solute urea dialysis clearance

    Bonsai Trees in Your Head: How the Pavlovian System Sculpts Goal-Directed Choices by Pruning Decision Trees

    When planning a series of actions, it is usually infeasible to consider all potential future sequences; instead, one must prune the decision tree. Provably optimal pruning is, however, still computationally ruinous and the specific approximations humans employ remain unknown. We designed a new sequential reinforcement-based task and showed that human subjects adopted a simple pruning strategy: during mental evaluation of a sequence of choices, they curtailed any further evaluation of a sequence as soon as they encountered a large loss. This pruning strategy was Pavlovian: it was reflexively evoked by large losses and persisted even when overwhelmingly counterproductive. It was also evident above and beyond loss aversion. We found that the tendency towards Pavlovian pruning was selectively predicted by the degree to which subjects exhibited sub-clinical mood disturbance, in accordance with theories that ascribe Pavlovian behavioural inhibition, via serotonin, a role in mood disorders. We conclude that Pavlovian behavioural inhibition shapes highly flexible, goal-directed choices in a manner that may be important for theories of decision-making in mood disorders
