    The timing and ecological consequences of Pleistocene megafaunal decline in the eastern Andes of Colombia (article)

We present the first multi-species record of spores of coprophilous fungi (SCF) from Monquentiva and the high-Andean forests of Colombia to reconstruct Late Pleistocene and Holocene megafaunal abundance. Fossilised pollen and charcoal are used to examine the consequences of megafaunal declines on the surrounding vegetation and fire activity. Our SCF record indicates the presence of Pleistocene megafauna at least since 30,290 BP, with two waves of megafaunal decline at ca. 22,900 BP and 10,990 BP. At Monquentiva, megafaunal decline in the Early Holocene resulted in transitional non-analogue vegetation, loss of some herbivore-dispersed plant taxa, an encroachment of palatable and woody flora, and a rise in fire activity. Differences with other published South-American records suggest that ecological consequences of megafaunal declines were habitat-specific. Overall, we show that ecosystems in the eastern Colombian Cordillera were highly sensitive to the decline of megafaunal populations.     Probing the close environment of young stellar objects with interferometry

    The study of Young Stellar Objects (YSOs) is one of the most exciting topics that can be undertaken by long baseline optical interferometry. The magnitudes of these objects are at the edge of capabilities of current optical interferometers, limiting the studies to a few dozen, but are well within the capability of coming large aperture interferometers like the VLT Interferometer, the Keck Interferometer, the Large Binocular Telescope or 'OHANA. The milli-arcsecond spatial resolution reached by interferometry probes the very close environment of young stars, down to a tenth of an astronomical unit. In this paper, I review the different aspects of star formation that can be tackled by interferometry: circumstellar disks, multiplicity, jets. I present recent observations performed with operational infrared interferometers, IOTA, PTI and ISI, and I show why in the next future one will extend these studies with large aperture interferometers.Comment: Review to be published in JENAM'2002 proceedings "The Very Large Telescope Interferometer Challenges for the future

    Randomized clinical trial to evaluate the effect of fecal microbiota transplant for initial Clostridium difficile infection in intestinal microbiome

    Objective The aim of this study was to evaluate the impact of fecal donor-unrelated donor mix (FMT-FURM) transplantation as first-line therapy for C. difficile infection (CDI) in intestinal microbiome. Methods We designed an open, two-arm pilot study with oral vancomycin (250mg every 6 h for 10–14 days) or FMT-FURM as treatments for the first CDI episode in hospitalized adult patients in Hospital Universitario “Dr. Jose Eleuterio Gonzalez”. Patients were randomized by a closed envelope method in a 1: 1 ratio to either oral vancomycin or FMT-FURM. CDI resolution was considered when there was a reduction on the Bristol scale of at least 2 points, a reduction of at least 50% in the number of bowel movements, absence of fever, and resolution of abdominal pain (at least two criteria). From each patient, a fecal sample was obtained at days 0, 3, and 7 after treatment. Specimens were cultured to isolate C. difficile, and isolates were characterized by PCR. Susceptibility testing of isolates was performed using the agar dilution method. Fecal samples and FMT-FURM were analyzed by 16S rRNA sequencing. Results We included 19 patients; 10 in the vancomycin arm and 9 in the FMT-FURM arm. However, one of the patients in the vancomycin arm and two patients in the FMT-FURM arm were eliminated. Symptoms resolved in 8/9 patients (88.9%) in the vancomycin group, while symptoms resolved in 4/7 patients (57.1%) after the first FMT-FURM dose (P = 0.26) and in 5/7 patients (71.4%) after the second dose (P = 0.55). During the study, no adverse effects attributable to FMT-FURM were observed in patients. Twelve isolates were recovered, most isolates carried tcdB, tcdA, cdtA, and cdtB, with an 18-bp deletion in tcdC. All isolates were resistant to ciprofloxacin and moxifloxacin but susceptible to metronidazole, linezolid, fidaxomicin, and tetracycline. In the FMT-FURM group, the bacterial composition was dominated by Firmicutes, Bacteroidetes, and Proteobacteria at all-time points and the microbiota were remarkably stable over time. The vancomycin group showed a very different pattern of the microbial composition when comparing to the FMT-FURM group over time. Conclusion The results of this preliminary study showed that FMT-FURM for initial CDI is associated with specific bacterial communities that do not resemble the donors’ sample.Peer reviewedFinal Published versio

    A Very Young Radio-loud Magnetar

    The magnetar Swift J1818.0–1607 was discovered in 2020 March when Swift detected a 9 ms hard X-ray burst and a long-lived outburst. Prompt X-ray observations revealed a spin period of 1.36 s, soon confirmed by the discovery of radio pulsations. We report here on the analysis of the Swift burst and follow-up X-ray and radio observations. The burst average luminosity was L burst ~ 2 × 1039 erg s−1 (at 4.8 kpc). Simultaneous observations with XMM-Newton and NuSTAR three days after the burst provided a source spectrum well fit by an absorbed blackbody (NH{N}_{{\rm{H}}} = (1.13 ± 0.03) × 1023 cm−2 and kT = 1.16 ± 0.03 keV) plus a power law (Γ = 0.0 ± 1.3) in the 1–20 keV band, with a luminosity of ~8 × 1034 erg s−1, dominated by the blackbody emission. From our timing analysis, we derive a dipolar magnetic field B ~ 7 × 1014 G, spin-down luminosity E˙rot1.4×1036{\dot{E}}_{\mathrm{rot}}\sim 1.4\times {10}^{36} erg s−1, and characteristic age of 240 yr, the shortest currently known. Archival observations led to an upper limit on the quiescent luminosity <5.5 × 1033 erg s−1, lower than the value expected from magnetar cooling models at the source characteristic age. A 1 hr radio observation with the Sardinia Radio Telescope taken about 1 week after the X-ray burst detected a number of strong and short radio pulses at 1.5 GHz, in addition to regular pulsed emission; they were emitted at an average rate 0.9 min−1 and accounted for ~50% of the total pulsed radio fluence. We conclude that Swift J1818.0–1607 is a peculiar magnetar belonging to the small, diverse group of young neutron stars with properties straddling those of rotationally and magnetically powered pulsars. Future observations will make a better estimation of the age possible by measuring the spin-down rate in quiescence

    Caspase-8 binding to cardiolipin in giant unilamellar vesicles provides a functional docking platform for bid

    Caspase-8 is involved in death receptor-mediated apoptosis in type II cells, the proapoptotic programme of which is triggered by truncated Bid. Indeed, caspase-8 and Bid are the known intermediates of this signalling pathway. Cardiolipin has been shown to provide an anchor and an essential activating platform for caspase-8 at the mitochondrial membrane surface. Destabilisation of this platform alters receptor-mediated apoptosis in diseases such as Barth Syndrome, which is characterised by the presence of immature cardiolipin which does not allow caspase-8 binding. We used a simplified in vitro system that mimics contact sites and/or cardiolipin-enriched microdomains at the outer mitochondrial surface in which the platform consisting of caspase-8, Bid and cardiolipin was reconstituted in giant unilamellar vesicles. We analysed these vesicles by flow cytometry and confirm previous results that demonstrate the requirement for intact mature cardiolipin for caspase-8 activation and Bid binding and cleavage. We also used confocal microscopy to visualise the rupture of the vesicles and their revesiculation at smaller sizes due to alteration of the curvature following caspase-8 and Bid binding. Biophysical approaches, including Laurdan fluorescence and rupture/tension measurements, were used to determine the ability of these three components (cardiolipin, caspase-8 and Bid) to fulfil the minimal requirements for the formation and function of the platform at the mitochondrial membrane. Our results shed light on the active functional role of cardiolipin, bridging the gap between death receptors and mitochondria

    Stellar encounters as the origin of distant solar system objects in highly eccentric orbits

    The discovery of Sedna places new constraints on the origin and evolution of our solar system. Here we investigate the possibility that a close encounter with another star produced the observed edge of the Kuiper belt, at roughly 50 AU, and the highly elliptical orbit of Sedna. We show that a passing star probably scattered Sedna from the Kuiper Belt into its observed orbit. The likelihood that a planet at 60-80 AU can be scattered into Sedna's orbit is roughly 50%; this estimate depends critically on the geometry of the flyby. Even more interesting, though, is the roughly 10% chance that Sedna was captured from the outer disk of the passing star. Most captures have very high inclination orbits; detection of these objects would confirm the presence of extrasolar planets in our own Solar System.Comment: 9 pages, 3 figure
