    Eml1 loss impairs apical progenitor spindle length and soma shape in the developing cerebral cortex

    The ventricular zone (VZ) of the developing cerebral cortex is a pseudostratified epithelium that contains progenitors undergoing precisely regulated divisions at its most apical side, the ventricular lining (VL). Mitotic perturbations can contribute to pathological mechanisms leading to cortical malformations. The HeCo mutant mouse exhibits subcortical band heterotopia (SBH), likely to be initiated by progenitor delamination from the VZ early during corticogenesis. The causes for this are however, currently unknown. Eml1, a microtubule (MT)-associated protein of the EMAP family, is impaired in these mice. We first show that MT dynamics are perturbed in mutant progenitor cells in vitro. These may influence interphase and mitotic MT mechanisms and indeed, centrosome and primary cilia were altered and spindles were found to be abnormally long in HeCo progenitors. Consistently, MT and spindle length regulators were identified in EML1 pulldowns from embryonic brain extracts. Finally, we found that mitotic cell shape is also abnormal in the mutant VZ. These previously unidentified VZ characteristics suggest altered cell constraints which may contribute to cell delamination

    Modal Ω-Logic: Automata, Neo-Logicism, and Set-Theoretic Realism

    This essay examines the philosophical significance of Ω\Omega-logic in Zermelo-Fraenkel set theory with choice (ZFC). The duality between coalgebra and algebra permits Boolean-valued algebraic models of ZFC to be interpreted as coalgebras. The modal profile of Ω\Omega-logical validity can then be countenanced within a coalgebraic logic, and Ω\Omega-logical validity can be defined via deterministic automata. I argue that the philosophical significance of the foregoing is two-fold. First, because the epistemic and modal profiles of Ω\Omega-logical validity correspond to those of second-order logical consequence, Ω\Omega-logical validity is genuinely logical, and thus vindicates a neo-logicist conception of mathematical truth in the set-theoretic multiverse. Second, the foregoing provides a modal-computational account of the interpretation of mathematical vocabulary, adducing in favor of a realist conception of the cumulative hierarchy of sets

    Investigation of late Pleistocene and early Holocene palaeoenvironmental change at El Mirón cave (Cantabria, Spain): insights from carbon and nitrogen isotope analysis of red deer

    Abstract: El Mirón Cavewas occupied by humans for over 40,000 years. Evidence of LateMousterian,Gravettian, Solutrean, Magdalenian, Azilian, Mesolithic, Neolithic, Chalcolithic, Bronze Age and Mediaeval occupations has been found in the cave. Understanding the local environmental conditions during the occupations is crucial for gaining an insight into the lifeways of El Mirón's inhabitants as they relied on the surrounding region and its natural resources for their subsistence. 170 bones of hunted red deer recovered from the cave were sampled for carbon and nitrogen stable isotope analyses with the aim of reconstructing the palaeoenvironment and palaeoclimate during the human occupation. The results show that the surrounding landscape underwent considerable environmental change during the Late Pleistocene and Early to Mid-Holocene. Shifts in d13C values between the Last Glacial Maximum, Heinrich stadial 1, Heinrich event 1, the Late-glacial interstadial and the onset of the Holocene are likely to reflect changes in water availability and temperature. Deer d15N generally increased over time indicating the regeneration of soil biological activity and nitrogen cycling, which was temporarily halted during the Younger Dryas. Comparison of the El Mirón results with those of 300 deer from other regions of Europe shows geographical variations in the timing and magnitude of the variation in d13C and d15N values. This variation tracks local climate (temperature andwater availability) and environmental (vegetation and forest development) change

    Quantitative Proteomics Reveals Myosin and Actin as Promising Saliva Biomarkers for Distinguishing Pre-Malignant and Malignant Oral Lesions

    Oral cancer survival rates increase significantly when it is detected and treated early. Unfortunately, clinicians now lack tests which easily and reliably distinguish pre-malignant oral lesions from those already transitioned to malignancy. A test for proteins, ones found in non-invasively-collected whole saliva and whose abundances distinguish these lesion types, would meet this critical need.To discover such proteins, in a first-of-its-kind study we used advanced mass spectrometry-based quantitative proteomics analysis of the pooled soluble fraction of whole saliva from four subjects with pre-malignant lesions and four with malignant lesions. We prioritized candidate biomarkers via bioinformatics and validated selected proteins by western blotting. Bioinformatic analysis of differentially abundant proteins and initial western blotting revealed increased abundance of myosin and actin in patients with malignant lesions. We validated those results by additional western blotting of individual whole saliva samples from twelve other subjects with pre-malignant oral lesions and twelve with malignant oral lesions. Sensitivity/specificity values for distinguishing between different lesion types were 100%/75% (p = 0.002) for actin, and 67%/83% (p<0.00001) for myosin in soluble saliva. Exfoliated epithelial cells from subjects' saliva also showed increased myosin and actin abundance in those with malignant lesions, linking our observations in soluble saliva to abundance differences between pre-malignant and malignant cells.Salivary actin and myosin abundances distinguish oral lesion types with sensitivity and specificity rivaling other non-invasive oral cancer tests. Our findings provide a promising starting point for the development of non-invasive and inexpensive salivary tests to reliably detect oral cancer early
