    Prebiotic xylo-oligosaccharides as high-value co-products on an integrated biorefinery approach from lignocellulosic feedstock

    The present work proposes the production of prebiotic xylo-oligosaccharides (XOS) as high-value co-products of the Lignocellulose Feedstock Biorefinery concept, foreseeing potential applications on food, feed and nutraceutical industries. Autohydrolysis was used to selectively solubilise the hemicellulosic fraction of several xylan-rich, widely available, agricultural, agro-industrial and forestry by-products: corn cobs, brewery’s spent grain and Eucalyptus wood chips. The soluble hemicellulose-rich and the solid cellulose- and lignin-rich fractions were separated, and the crude XOS-rich hydrolysates were further purified by gel filtration chromatography. Selected fractions of purified XOS within the desired ranges of polymerization degree were characterised and their prebiotic potential was investigated in in vitro fermentations by bifidobacteria, lactobacilli and intestinal inocula. Parameters such as bacterial growth and XOS consumption were evaluated and compared with commercially available xylo-oligosaccharides. The differences observed were considered of relevance for the formulation of symbiotic preparations and the future design of targeted, tailor-made prebiotic xylo-oligosaccharides

    Sustainable value methodology to compare the performance of conversion technologies for the production of electricity and heat, energy vectors and biofuels from waste biomass

    ABSTRACT: The Sustainable Value methodology was used to compare and rank eight combinations of waste biomass types and conversion technologies on a common assessment basis to produce energy, energy vectors and advanced biofuels. The studied combinations included agricultural and agro-industrial residues, slurries and effluents, pulp and paper mill sludge, piggery effluents and organic fractions of municipal solid waste, to produce biodiesel by (trans)esterification, biogas by anaerobic digestion, ethanol by fermentation, hydrogen by dark fermentation, electricity and heat by combustion, biogas and synthesis gas by gasification, and bio-oils by pyrolysis or hydrothermal liquefaction. The numerator “Functional Performance” of the Sustainable Value indicator was estimated according to 14 criteria of process technology, material and energy inputs and outputs, and acceptance by the stakeholders. The performance of the technologies was classified based on the values of relative importance (φ) and level of satisfaction (S) attributed to each criterion. The gasification of residues from the olive-oil industry reached the highest “Functional Performance”, followed by anaerobic digestion of chestnut processing residues and pig-rearing effluents. The Sustainable Value denominator “Costs” depended mainly on the degree of maturity of the technologies, which penalised pyrolysis, hydrothermal liquefaction and dark fermentation. The final ranking of the Sustainable Value indicator was gasification> combustion> anaerobic digestion> (trans)esterification> pyrolysis and fermentation to ethanol> hydrothermal liquefaction> dark fermentation, respectively for the most adequate waste biomass types under study.     Microsatellite Markers For Studies With The Carnivorous Plant Philcoxia Minensis (plantaginaceae).

    Microsatellite markers were developed for the critically endangered carnivorous species Philcoxia minensis (Plantaginaceae) for further population genetic studies aiming at its conservation. We identified 29 clones containing 40 microsatellites from a genomic enriched library. A total of 27 primer pairs were developed and evaluated in 30 individuals of a natural P. minensis population. Seventeen markers successfully presented amplification products within the expected size range, of which 12 were polymorphic. The expected and observed heterozygosities ranged from 0.03 to 0.65 and from 0.00 to 0.77, respectively. Positive transferability with the related species P. bahiensis was observed for the same 17 markers. The 12 polymorphic microsatellite markers are suitable for studies in genetic diversity and structure, mating system, and gene flow in P. minensis and also may be useful for similar issues regarding the related species P. bahiensis.

    Berberine reverts hepatic mitochondrial dysfunction in high-fat fed rats: A possible role for SirT3 activation

    Berberine is an isoquinoline alkaloid with anti-diabetic properties. Despite the central role of liver and thus hepaticmitochondria inwhole-bodymetabolism, berberine effects on hepaticmitochondrial function in an obesity model are still unknown. Here, we demonstrate that berberine treatment recovers mitochondrial efficiency when altered by a high-fat feeding.Mitochondria isolated from the liver of high-fat fed rats exhibited decreased capacity to accumulate calcium and impaired oxidative phosphorylation (OXPHOS) capacity, as shown by impaired mitochondrialmembrane potential, oxygen consumption and cellular ATP levels. Interestingly, the recovery of mitochondrial function by berberine was associated with an increased activity of the mitochondrial sirtuin 3 (SirT3).     Computer analysis of objects’ movement in image sequences: methods and applications

    Computer analysis of objects’ movement in image sequences is a very complex problem, considering that it usually involves tasks for automatic detection, matching, tracking, motion analysis and deformation estimation. In spite of its complexity, this computational analysis has a wide range of important applications; for instance, in surveillance systems, clinical analysis of human gait, objects recognition, pose estimation and deformation analysis. Due to the extent of the purposes, several difficulties arise, such as the simultaneous tracking of manifold objects, their possible temporary occlusion or definitive disappearance from the image scene, changes of the viewpoints considered in images acquisition or of the illumination conditions, or even nonrigid deformations that objects may suffer in image sequences. In this paper, we present an overview of several methods that may be considered to analyze objects’ movement; namely, for their segmentation, tracking and matching in images, and for estimation of the deformation involved between images.This paper was partially done in the scope of project “Segmentation, Tracking and Motion Analysis of Deformable (2D/3D) Objects using Physical Principles”, with reference POSC/EEA-SRI/55386/2004, financially supported by FCT -Fundação para a Ciência e a Tecnologia from Portugal. Strain-controlled low-cycle fatigue up to 6% was monitored by in situ electrical resistivity measurements. The experimental results show that a great motion of martensite fronts results in a significant accumulation of defects, as evidenced by transmission electron microscopy (TEM), before and after the tensile cycles. This gives rise to an overall increase of the resistivity values up to the maximum deformation. Therefore, the research suggests that shape memory alloy wire has great potential as a stress sensor inside bulk materials.publishersversionpublishe

    Genetic Variants Underlying Risk of Intracranial Aneurysms: Insights from a GWAS in Portugal

    Subarachnoid hemorrhage (SAH) is a life-threatening event that most frequently leads to severe disability and death. Its most frequent cause is the rupture of a saccular intracranial aneurysm (IA), which is a blood vessel dilation caused by disease or weakening of the vessel wall. Although the genetic contribution to IA is well established, to date no single gene has been unequivocally identified as responsible for IA formation or rupture. We aimed to identify IA susceptibility genes in the Portuguese population through a pool-based multistage genome-wide association study. Replicate pools were allelotyped in triplicate in a discovery dataset (100 IA cases and 92 gender-matched controls) using the Affymetrix Human SNP Array 6.0. Top SNPs (absolute value of the relative allele score difference between cases and controls |RASdiff|≥13.0%) were selected for technical validation by individual genotyping in the discovery dataset. From the 101 SNPs successfully genotyped, 99 SNPs were nominally associated with IA. Replication of technically validated SNPs was conducted in an independent replication dataset (100 Portuguese IA cases and 407 controls). rs4667622 (between UBR3 and MYO3B), rs6599001 (between SCN11A and WDR48), rs3932338 (214 kilobases downstream of PRDM9), and rs10943471 (96 kilobases upstream of HTR1B) were associated with IA (unadjusted allelic chi-square tests) in the datasets tested (discovery: 6.84E-04≤P≤1.92E-02, replication: 2.66E-04≤P≤2.28E-02, and combined datasets: 6.05E-05≤P≤5.50E-04). Additionally, we confirmed the known association with IA of rs1333040 at the 9p21.3 genomic region, thus validating our dataset.
