    Clarifying the absence of evidence regarding human health risks to microplastic particles in drinking-water: High quality robust data wanted

    In a recently published article, Leslie and Depledge (2020) raise concerns regarding statements on the risk that microplastic particles represent to human health and which have been attributed to reports published by both the Science Academies’ Group, Science Advice for Policy (SAPEA) (part of the European Commission’s Science Advice Mechanism) and the World Health Organization (WHO) (SAPEA. Science Advice for Policy by European Academies, 2019, WHO, 2019). Leslie and Depledge (2020), for instance, suggest that WHO (2019) conclude that there is ‘no evidence to indicate a human health concern.’ This statement, taken out of context from the WHO report (WHO, 2019), is then used to imply that the WHO conclude there is ‘no risk’ related to the exposure of microplastic particles (Leslie and Depledge, 2020). While, Leslie and Depledge (2020) highlight the importance of debate and systematic assessment of claims related to the assessment of risk, observations that we agree are important to highlight, there are a number of points raised in the article that require clarification

    Enhanced translation by Nucleolin via G-rich elements in coding and non-coding regions of target mRNAs

    RNA-binding proteins (RBPs) regulate gene expression at many post-transcriptional levels, including mRNA stability and translation. The RBP nucleolin, with four RNA-recognition motifs, has been implicated in cell proliferation, carcinogenesis and viral infection. However, the subset of nucleolin target mRNAs and the influence of nucleolin on their expression had not been studied at a transcriptome-wide level. Here, we globally identified nucleolin target transcripts, many of which encoded cell growth- and cancer-related proteins, and used them to find a signature motif on nucleolin target mRNAs. Surprisingly, this motif was very rich in G residues and was not only found in the 3â€Č-untranslated region (UTR), but also in the coding region (CR) and 5â€Č-UTR. Nucleolin enhanced the translation of mRNAs bearing the G-rich motif, since silencing nucleolin did not change target mRNA stability, but decreased the size of polysomes forming on target transcripts and lowered the abundance of the encoded proteins. In summary, nucleolin binds G-rich sequences in the CR and UTRs of target mRNAs, many of which encode cancer proteins, and enhances their translation

    Nucleolin Inhibits G4 Oligonucleotide Unwinding by Werner Helicase

    The Werner protein (WRNp), a member of the RecQ helicase family, is strongly associated with the nucleolus, as is nucleolin (NCL), an important nucleolar constituent protein. Both WRNp and NCL respond to the effects of DNA damaging agents. Therefore, we have investigated if these nuclear proteins interact and if this interaction has a possible functional significance in DNA damage repair.Here we report that WRNp interacts with the RNA-binding protein, NCL, based on immunoprecipitation, immunofluorescent co-localization in live and fixed cells, and direct binding of purified WRNp to nucleolin. We also map the binding region to the C-terminal domains of both proteins. Furthermore, treatment of U2OS cells with 15 ”M of the Topoisomerase I inhibitor, camptothecin, causes the dissociation of the nucleolin-Werner complex in the nucleolus, followed by partial re-association in the nucleoplasm. Other DNA damaging agents, such as hydroxyurea, Mitomycin C, and aphidicolin do not have these effects. Nucleolin or its C-terminal fragment affected the helicase, but not the exonuclease activity of WRNp, by inhibiting WRN unwinding of G4 tetraplex DNA structures, as seen in activity assays and electrophoretic mobility shift assays (EMSA).These data suggest that nucleolin may regulate G4 DNA unwinding by WRNp, possibly in response to certain DNA damaging agents. We postulate that the NCL-WRNp complex may contain an inactive form of WRNp, which is released from the nucleolus upon DNA damage. Then, when required, WRNp is released from inhibition and can participate in the DNA repair processes

    COVID-19 symptoms at hospital admission vary with age and sex: results from the ISARIC prospective multinational observational study

    Background: The ISARIC prospective multinational observational study is the largest cohort of hospitalized patients with COVID-19. We present relationships of age, sex, and nationality to presenting symptoms. Methods: International, prospective observational study of 60 109 hospitalized symptomatic patients with laboratory-confirmed COVID-19 recruited from 43 countries between 30 January and 3 August 2020. Logistic regression was performed to evaluate relationships of age and sex to published COVID-19 case definitions and the most commonly reported symptoms. Results: ‘Typical’ symptoms of fever (69%), cough (68%) and shortness of breath (66%) were the most commonly reported. 92% of patients experienced at least one of these. Prevalence of typical symptoms was greatest in 30- to 60-year-olds (respectively 80, 79, 69%; at least one 95%). They were reported less frequently in children (≀ 18 years: 69, 48, 23; 85%), older adults (≄ 70 years: 61, 62, 65; 90%), and women (66, 66, 64; 90%; vs. men 71, 70, 67; 93%, each P < 0.001). The most common atypical presentations under 60 years of age were nausea and vomiting and abdominal pain, and over 60 years was confusion. Regression models showed significant differences in symptoms with sex, age and country. Interpretation: This international collaboration has allowed us to report reliable symptom data from the largest cohort of patients admitted to hospital with COVID-19. Adults over 60 and children admitted to hospital with COVID-19 are less likely to present with typical symptoms. Nausea and vomiting are common atypical presentations under 30 years. Confusion is a frequent atypical presentation of COVID-19 in adults over 60 years. Women are less likely to experience typical symptoms than men

L'ouverture des marchĂ©s mondiaux, le dĂ©veloppement rapide des nouvelles technologies et le mouvement de rationalisation des dĂ©penses et des effectifs entrainent un climat d'instabilitĂ© non propice Ă  l'emploi et une augmentation du chĂŽmage surtout chez les jeunes mĂȘme s'ils sont diplĂŽmĂ©s. Il n'est pas si loin le temps ou la possession d'un diplĂŽme universitaire Ă©quivalait Ă  un emploi permanent, rĂ©munĂ©rateur et gratifiant. Les transformations rapides du marchĂ© du travail commandent l'acquisition de compĂ©tences spĂ©cifiques pour les personnes sortant de nos universitĂ©s. L'exploration de nouvelles voies s'impose pour favoriser leur insertion professionnelle et celle de l'entrepreneuriat constitue une alternative fort intĂ©ressante pour ceux et celles qui voudraient relever le dĂ©fi. Mais avant de se lancer dans cette aventure, une connaissance appropriĂ©e de ses qualitĂ©s entrepreneuriales correspond au premier palier de la rĂ©ussite pour l'entrepreneur et aussi pour celui qui vise un emploi dans une organisation. Actuellement, peu d'instruments permettent une autoĂ©valuation de ces qualitĂ©s pour cette clientĂšle. Nous nous proposons dans la prĂ©sente recherche d'adapter Ă  une clientĂšle universitaire le questionnaire dĂ©veloppĂ© par L'Heureux (1994) auprĂšs de jeunes de niveau secondaire et intitulĂ© «Évaluation de mes qualitĂ©s entrepreneuriales.» De plus, Ă  partir des rĂ©ponses fournies, nous pourrons Ă©valuer la perception qu'ont les sujets de leurs qualitĂ©s entrepreneuriales. Ainsi, nous contribuerons au dĂ©veloppement d'un outil adaptĂ© Ă  une clientĂšle universitaire et nous cumulerons une information prĂ©cieuse sur son profil entrepreneurial
