    Nanostructure-modulated planar high spectral resolution spectro-polarimeter

    We present a planar spectro-polarimeter based on Fabry-P{\'e}rot cavities with embedded polarization-sensitive high-index nanostructures. A 7 μ7~\mum-thick spectro-polarimetric system for 3 spectral bands and 2 linear polarization states is experimentally demonstrated. Furthermore, an optimal design is theoretically proposed, estimating that a system with a bandwidth of 127~nm and a spectral resolution of 1~nm is able to reconstruct the first three Stokes parameters \textcolor{black}{with a signal-to-noise ratio of -13.14~dB with respect to the the shot noise limited SNR}. The pixelated spectro-polarimetric system can be directly integrated on a sensor, thus enabling applicability in a variety of miniaturized optical devices, including but not limited to satellites for Earth observation

    Motif effects in Affymetrix GeneChips seriously affect probe intensities

    An Affymetrix GeneChip consists of an array of hundreds of thousands of probes (each a sequence of 25 bases) with the probe values being used to infer the extent to which genes are expressed in the biological material under investigation. In this article, we demonstrate that these probe values are also strongly influenced by their precise base sequence. We use data from >28 000 CEL files relating to 10 different Affymetrix GeneChip platforms and involving nearly 1000 experiments. Our results confirm known effects (those due to the T7-primer and the formation of G-quadruplexes) but reveal other effects. We show that there can be huge variations from one experiment to another, and that there may also be sizeable disparities between batches within an experiment and between CEL files within a batch. © 2012 The Author(s)

    Physico-chemical foundations underpinning microarray and next-generation sequencing experiments

    Hybridization of nucleic acids on solid surfaces is a key process involved in high-throughput technologies such as microarrays and, in some cases, next-generation sequencing (NGS). A physical understanding of the hybridization process helps to determine the accuracy of these technologies. The goal of a widespread research program is to develop reliable transformations between the raw signals reported by the technologies and individual molecular concentrations from an ensemble of nucleic acids. This research has inputs from many areas, from bioinformatics and biostatistics, to theoretical and experimental biochemistry and biophysics, to computer simulations. A group of leading researchers met in Ploen Germany in 2011 to discuss present knowledge and limitations of our physico-chemical understanding of high-throughput nucleic acid technologies. This meeting inspired us to write this summary, which provides an overview of the state-of-the-art approaches based on physico-chemical foundation to modeling of the nucleic acids hybridization process on solid surfaces. In addition, practical application of current knowledge is emphasized

    Employing geographical principles for sampling in state of the art dialectological projects

    The aims of this paper are twofold: First, we locate the most effective human geographical methods for sampling across space in large-scale dialectological projects. We propose two geographical concepts as a basis for sampling decisions: Geo-demographic classification, which is a multidimensional method used for the socio-economic grouping of areas. We also develop an updated version of functional regions that can be used in sociolinguistic research. We then report on the results of a pilot project that applies these models to collect data regarding the acceptability of vernacular morpho-syntactic forms in the North-East of England. Following the method of natural breaks advocated for dialectology by Horvath and Horvath (2002), we interpret breaks in the probabilistic patterns as areas of dialect transitions. This study contributes to the debate about the role and limitations of spatiality in linguistic analysis. It intends to broaden our knowledge about the interfaces between human geography and dialectology

    Of babies and bath water: Is there any place for Austin and Grice in interpersonal pragmatics?

    This paper discusses a particular strand of interpersonal pragmatics that may be known as ‘discursive’ pragmatics and attempts to delineate what is entailed in such an approach. Some scholars may characterise it as placing emphasis on participant evaluations, others may foreground the analysis of contextualised and sequential texts, while still others consider it to include both of these. In general, though, discursive pragmatics often seems to involve a reaction to, and a contrast with, so-called Gricean intention-based approaches. In this paper I argue that, far from discarding the insights of Grice, Austin and others, a discursive approach to interpersonal pragmatics should embrace those aspects of non-discursive pragmatics that provide us with a ‘tool-kit’ and a vocabulary for examining talk-in-interaction. At the same time, I will argue that the shortcomings of the speaker-based, intention- focused pragmatics can be compensated for, not by privileging hearer evaluations of meaning, but by taking an ethnographic and, to some extent, ethnomethodological approach to the analysis of naturally-occurring discourse data. By providing a critique of Locher and Watts’ (2005) paradigmatic example of a discursive approach to politeness and then a sample analysis of interactional data, I demonstrate how a combination of insights from Gricean pragmatics and from ethnomethodology allows the analyst to comment on the construction and negotiation of meaning in discourse, without having recourse to notions of either intention or evaluation

    The impact of team preferences on soccer offside judgments in laypersons

    The present study uses a quasi-experimental design to investigate the impact of team preferences on the accuracy of offside judgments. In Experiments 1 and 2, supporters of two German soccer clubs (i.e., Borussia Dortmund and FC Schalke 04) judged offsides in artificial scenes from a match between the clubs. We expected that supporters of both clubs would less frequently report the offside position of a forward from the preferred team. The results of Experiment 1 partly confirmed the predictions. Both groups reported the offside position of a yellow forward less frequently than that of a blue forward, and this effect was much larger for supporters of Borussia Dortmund than for supporters of Schalke 04. The difference between groups could be attributed to team preferences. The weaker effect of team preference in supporters of Schalke 04 was attributed to an unexpected perceptual effect that increased the accuracy of offside judgments for blue forwards in both groups. Experiments 2 and 3 showed the presumed effect of team preferences and the perceptual effect, respectively, in isolation. In summary, the results of our experiments provide evidence for (a) an effect of team preferences and (b) an effect of shirt–background contrast on offside judgments in soccer
