Dialogue and materiality/embodiment in science|arts creative pedagogy: their role and manifestation

Available on open access from Elsevier via the DOI in this record.This paper responds to recent calls to explore the nuances of the interaction between the sciences, the arts and their inherent creativity to better understand their potential within teaching and learning. Building on previous arguments that the science-arts-creativity relationship is dialogic and relational, this research focuses on the question: How are dialogue and material/embodied activity manifested within creative pedagogy? We begin with a fusion of Bakhtinian-inspired and New-Materialist understandings of dialogue drawing out the importance of embodiment in order to revitalize how we articulate dialogue within creative educational practice. We then take on the challenge of a materialist diffractive analysis to conduct research which complements the theoretical framing and offers our outcomes in a way that appropriately makes the phenomena tangible. We present the outcomes of the diffractive analysis including the constitution of matter as well as meaning in the dialogic space; and the emergence of new assemblages of embodied teachers, students, ideas, and objects within transdisciplinary educational practice. We conclude by arguing for the benefits of diffractive analysis: that we have fore-fronted the entangled relationality of trans-disciplinary creative pedagogy; avoided bracketing out aspects of education that are often side-lined; opened out the space of pedagogical
approaches that might be attempted; and begun to challenge what education is for. In so doing, the article aims to open up new ways for teachers, students and researchers to experience seeing, doing, feeling and researching science|arts creative pedagogy and provoke conversations about how this might develop in the future.European Commissio
Insights From Analysis of Human Antigen-Specific Memory B Cell Repertoires
Memory B cells that are generated during an infection or following vaccination act as sentinels to guard against future infections. Upon repeat antigen exposure memory B cells differentiate into new antibody-secreting plasma cells to provide rapid and sustained protection. Some pathogens evade or suppress the humoral immune system, or induce memory B cells with a diminished ability to differentiate into new plasma cells. This leaves the host vulnerable to chronic or recurrent infections. Single cell approaches coupled with next generation antibody gene sequencing facilitate a detailed analysis of the pathogen-specific memory B cell repertoire. Monoclonal antibodies that are generated from antibody gene sequences allow a functional analysis of the repertoire. This review discusses what has been learned thus far from analysis of diverse pathogen-specific memory B cell compartments and describes major differences in their repertoires. Such information may illuminate ways to advance the goal of improving vaccine and therapeutic antibody design
SDWFS-MT-1: A Self-Obscured Luminous Supernova at z~0.2
We report the discovery of a six-month-long mid-infrared transient,
SDWFS-MT-1 (aka SN 2007va), in the Spitzer Deep, Wide-Field Survey of the NOAO
Deep Wide-Field Survey Bootes field. The transient, located in a z=0.19 low
luminosity (M_[4.5]~-18.6 mag, L/L_MilkyWay~0.01) metal-poor (12+log(O/H)~7.8)
irregular galaxy, peaked at a mid-infrared absolute magnitude of M_[4.5]~-24.2
in the 4.5 micron Spitzer/IRAC band and emitted a total energy of at least
10^51 ergs. The optical emission was likely fainter than the mid-infrared,
although our constraints on the optical emission are poor because the transient
peaked when the source was "behind" the Sun. The Spitzer data are consistent
with emission by a modified black body with a temperature of ~1350 K. We rule
out a number of scenarios for the origin of the transient such as a Galactic
star, AGN activity, GRB, tidal disruption of a star by a black hole and
gravitational lensing. The most plausible scenario is a supernova exploding
inside a massive, optically thick circumstellar medium, composed of multiple
shells of previously ejected material. If the proposed scenario is correct,
then a significant fraction (~10%) of the most luminous supernova may be
self-enshrouded by dust not only before but also after the supernova occurs.
The spectral energy distribution of the progenitor of such a supernova would be

Observation of contemporaneous optical radiation from a gamma-ray burst
The origin of gamma-ray bursts (GRBs) has been enigmatic since their
discovery. The situation improved dramatically in 1997, when the rapid
availability of precise coordinates for the bursts allowed the detection of
faint optical and radio afterglows - optical spectra thus obtained have
demonstrated conclusively that the bursts occur at cosmological distances. But,
despite efforts by several groups, optical detection has not hitherto been
achieved during the brief duration of a burst. Here we report the detection of
bright optical emission from GRB990123 while the burst was still in progress.
Our observations begin 22 seconds after the onset of the burst and show an
increase in brightness by a factor of 14 during the first 25 seconds; the
brightness then declines by a factor of 100, at which point (700 seconds after
the burst onset) it falls below our detection threshold. The redshift of this
burst, approximately 1.6, implies a peak optical luminosity of 5 times 10^{49}
erg per second. Optical emission from gamma-ray bursts has been generally
thought to take place at the shock fronts generated by interaction of the
primary energy source with the surrounding medium, where the gamma-rays might
also be produced. The lack of a significant change in the gamma-ray light curve
when the optical emission develops suggests that the gamma-rays are not

Optical Lightcurve & Cooling Break of GRB 050502A
We present lightcurves of the afterglow of GRB050502A, including very early
data at t-t_{GRB} < 60s. The lightcurve is composed of unfiltered ROTSE-IIIb
optical observations from 44s to 6h post-burst, R-band MDM observations from
1.6 to 8.4h post-burst, and PAIRITEL J H K_s observations from 0.6 to 2.6h
post-burst. The optical lightcurve is fit by a broken power law, where
t^{alpha} steepens from alpha = -1.13 +- 0.02 to alpha = -1.44 +- 0.02 at
\~5700s. This steepening is consistent with the evolution expected for the
passage of the cooling frequency nu_c through the optical band. Even in our
earliest observation at 44s post-burst, there is no evidence that the optical
flux is brighter than a backward extrapolation of the later power law would
suggest. The observed decay indices and spectral index are consistent with
either an ISM or a Wind fireball model, but slightly favor the ISM
interpretation. The expected spectral index in the ISM interpretation is
consistent within 1 sigma with the observed spectral index beta = -0.8 +- 0.1;
the Wind interpretation would imply a slightly (~2 sigma) shallower spectral
index than observed. A small amount of dust extinction at the source redshift
could steepen an intrinsic spectrum sufficiently to account for the observed
value of beta. In this picture, the early optical decay, with the peak at or
below 4.7e14 Hz at 44s, requires very small electron and magnetic energy

Automating Genomic Data Mining via a Sequence-based Matrix Format and Associative Rule Set
There is an enormous amount of information encoded in each genome – enough to create living, responsive and adaptive organisms. Raw sequence data alone is not enough to understand function, mechanisms or interactions. Changes in a single base pair can lead to disease, such as sickle-cell anemia, while some large megabase deletions have no apparent phenotypic effect. Genomic features are varied in their data types and annotation of these features is spread across multiple databases. Herein, we develop a method to automate exploration of genomes by iteratively exploring sequence data for correlations and building upon them. First, to integrate and compare different annotation sources, a sequence matrix (SM) is developed to contain position-dependant information. Second, a classification tree is developed for matrix row types, specifying how each data type is to be treated with respect to other data types for analysis purposes. Third, correlative analyses are developed to analyze features of each matrix row in terms of the other rows, guided by the classification tree as to which analyses are appropriate. A prototype was developed and successful in detecting coinciding genomic features among genes, exons, repetitive elements and CpG islands

© 2017 American Speech-Language-Hearing Association. Purpose: The aim was to evaluate the effectiveness of computer-assisted input-based intervention for children with speech sound disorders (SSD). Method: The Sound Start Study was a cluster-randomized controlled trial. Seventy-nine early childhood centers were invited to participate, 45 were recruited, and 1,205 parents and educators of 4- and 5-year-old children returned questionnaires. Children whose parents and educators had concerns about speech were assessed (n =275); 132 children who were identified with phonological patternbased errors underwent additional assessment. Children with SSD and no difficulties with receptive language or hearing, typical nonverbal intelligence, and English as their primary language were eligible; 123 were randomized into two groups (intervention n = 65; control n = 58), and 3 withdrew. The intervention group involved Phoneme Factory Sound Sorter software (Wren & Roulstone, 2013) administered by educators over 9 weeks; the control group involved typical classroom practices. Participants were reassessed twice by a speech-language pathologist who was unaware of the initial assessment and intervention conditions. High-throughput processing and normalization of one-color microarrays for transcriptional meta-analyses
<p>Abstract</p> <p>Background</p> <p>Microarray experiments are becoming increasingly common in biomedical research, as is their deposition in publicly accessible repositories, such as Gene Expression Omnibus (GEO). As such, there has been a surge in interest to use this microarray data for meta-analytic approaches, whether to increase sample size for a more powerful analysis of a specific disease (e.g. lung cancer) or to re-examine experiments for reasons different than those examined in the initial, publishing study that generated them. For the average biomedical researcher, there are a number of practical barriers to conducting such meta-analyses such as manually aggregating, filtering and formatting the data. Methods to automatically process large repositories of microarray data into a standardized, directly comparable format will enable easier and more reliable access to microarray data to conduct meta-analyses.</p> <p>Methods</p> <p>We present a straightforward, simple but robust against potential outliers method for automatic quality control and pre-processing of tens of thousands of single-channel microarray data files. GEO GDS files are quality checked by comparing parametric distributions and quantile normalized to enable direct comparison of expression level for subsequent meta-analyses.</p> <p>Results</p> <p>13,000 human 1-color experiments were processed to create a single gene expression matrix that subsets can be extracted from to conduct meta-analyses. Interestingly, we found that when conducting a global meta-analysis of gene-gene co-expression patterns across all 13,000 experiments to predict gene function, normalization had minimal improvement over using the raw data.</p> <p>Conclusions</p> <p>Normalization of microarray data appears to be of minimal importance on analyses based on co-expression patterns when the sample size is on the order of thousands microarray datasets. Smaller subsets, however, are more prone to aberrations and artefacts, and effective means of automating normalization procedures not only empowers meta-analytic approaches, but aids in reproducibility by providing a standard way of approaching the problem.</p> <p>Data availability: matrix containing normalized expression of 20,813 genes across 13,000 experiments is available for download at . Extracellular DNA, cell surface proteins and c-di-GMP promote biofilm formation in Clostridioides difficile.
Clostridioides difficile is the leading cause of nosocomial antibiotic-associated diarrhoea worldwide, yet there is little insight into intestinal tract colonisation and relapse. In many bacterial species, the secondary messenger cyclic-di-GMP mediates switching between planktonic phase, sessile growth and biofilm formation. We demonstrate that c-di-GMP promotes early biofilm formation in C. difficile and that four cell surface proteins contribute to biofilm formation, including two c-di-GMP regulated; CD2831 and CD3246, and two c-di-GMP-independent; CD3392 and CD0183. We demonstrate that C. difficile biofilms are composed of extracellular DNA (eDNA), cell surface and intracellular proteins, which form a protective matrix around C. difficile vegetative cells and spores, as shown by a protective effect against the antibiotic vancomycin. We demonstrate a positive correlation between biofilm biomass, sporulation frequency and eDNA abundance in all five C. difficile lineages. Strains 630 (RT012), CD305 (RT023) and M120 (RT078) contain significantly more eDNA in their biofilm matrix than strains R20291 (RT027) and M68 (RT017). DNase has a profound effect on biofilm integrity, resulting in complete disassembly of the biofilm matrix, inhibition of biofilm formation and reduced spore germination. The addition of exogenous DNase could be exploited in treatment of C. difficile infection and relapse, to improve antibiotic efficacy
GRB 081008: from burst to afterglow and the transition phase in between
We present a multi-wavelength study of GRB 081008, at redshift 1.967, by
Swift, ROTSE-III and GROND. Compared to other Swift GRBs, GRB 081008 has a
typical gamma-ray isotropic equivalent energy output (10^53 erg) during the
prompt phase, and displayed two temporally separated clusters of pulses. The
early X-ray emission seen by the Swift/XRT was dominated by the softening tail
of the prompt emission, producing multiple flares during and after the
Swift/BAT detections. Optical observations that started shortly after the first
active phase of gamma-ray emission showed two consecutive peaks. We interpret
the first optical peak as the onset of the afterglow associated with the early
burst activities. A second optical peak, coincident with the later gamma-ray
pulses, imposes a small modification to the otherwise smooth lightcurve and
thus suggests a minimal contribution from a probable internal component. We
suggest the early optical variability may be from continuous energy injection
into the forward shock front by later shells producing the second epoch of
burst activities. These early observations thus provide a potential probe for
the transition from prompt to the afterglow phase. The later lightcurve of GRB
081008 displays a smooth steepening in all optical bands and X-ray. The
temporal break is consistent with being achromatic at the observed wavelengths.
Our broad energy coverage shortly after the break constrains a spectral break
within optical. However, the evolution of the break frequency is not observed.
