    Interlaminar shear stress effects on the postbuckling response of graphite-epoxy panels

    The influence of shear flexibility on overall postbuckling response was assessed, and transverse shear stress distributions in relation to panel failure were examined. Nonlinear postbuckling results are obtained for finite element models based on classical laminated plate theory and first-order shear deformation theory. Good correlation between test and analysis is obtained. The results presented analytically substantiate the experimentally observed failure mode

    Comparison of Requirements for Composite Structures for Aircraft and Space Applications

    In this report, the aircraft and space vehicle requirements for composite structures are compared. It is a valuable exercise to study composite structural design approaches used in the airframe industry and to adopt methodology that is applicable for space vehicles. The missions, environments, analysis methods, analysis validation approaches, testing programs, build quantities, inspection, and maintenance procedures used by the airframe industry, in general, are not transferable to spaceflight hardware. Therefore, while the application of composite design approaches from aircraft and other industries is appealing, many aspects cannot be directly utilized. Nevertheless, experiences and research for composite aircraft structures may be of use in unexpected arenas as space exploration technology develops, and so continued technology exchanges are encouraged

    Ectopic Expression of Vaccinia Virus E3 and K3 Cannot Rescue Ectromelia Virus Replication in Rabbit RK13 Cells

However, there is individual variability in the range of species able to be productively infected. In this study, we observed that ectromelia virus (ECTV) does not replicate efficiently in cultured rabbit RK13 cells. Conversely, vaccinia virus (VACV) replicates well in these cells. Upon infection of RK13 cells, the replication cycle of ECTV is abortive in nature, resulting in a greatly reduced ability to spread among cells in culture. We observed ample levels of early gene expression but reduced detection of virus factories and severely blunted production of enveloped virus at the cell surface. This work focused on two important host range genes, named E3L and K3L, in VACV. Both VACV and ECTV express a functional protein product from the E3L gene, but only VACV contains an intact K3L gene. To better understand the discrepancy in replication capacity of these viruses, we examined the ability of ECTV to replicate in wild-type RK13 cells compared to cells that constitutively express E3 and K3 from VACV. The role these proteins play in the ability of VACV to replicate in RK13 cells was also analyzed to determine their individual contribution to viral replication and PKR activation. Since E3L and K3L are two relevant host range genes, we hypothesized that expression of one or both of them may have a positive impact on the ability of ECTV to replicate in RK13 cells. Using various methods to assess virus growth, we did not detect any significant differences with respect to the replication of ECTV between wild-type RK13 compared to versions of this cell line that stably expressed VACV E3 alone or in combination with K3. Therefore, there remain unanswered questions related to the factors that limit the host range of ECTV

    Multiple Phosphatidylinositol 3-Kinases Regulate Vaccinia Virus Morphogenesis

    Poxvirus morphogenesis is a complex process that involves the successive wrapping of the virus in host cell membranes. We screened by plaque assay a focused library of kinase inhibitors for those that caused a reduction in viral growth and identified several compounds that selectively inhibit phosphatidylinositol 3-kinase (PI3K). Previous studies demonstrated that PI3Ks mediate poxviral entry. Using growth curves and electron microscopy in conjunction with inhibitors, we show that that PI3Ks additionally regulate morphogenesis at two distinct steps: immature to mature virion (IMV) transition, and IMV envelopment to form intracellular enveloped virions (IEV). Cells derived from animals lacking the p85 regulatory subunit of Type I PI3Ks (p85α−/−β−/−) presented phenotypes similar to those observed with PI3K inhibitors. In addition, VV appear to redundantly use PI3Ks, as PI3K inhibitors further reduce plaque size and number in p85α−/−β−/− cells. Together, these data provide evidence for a novel regulatory mechanism for virion morphogenesis involving phosphatidylinositol dynamics and may represent a new therapeutic target to contain poxviruses

    Preditores de não aderência ao seguimento preconizado para mulheres com lesão intraepitelial escamosa de alto grau (HSIL)

    O câncer de colo uterino é um tumor de natureza multifatorial que persiste como um importante problema de saúde pública. Aderência à linha de cuidado é fator associado ao controle desse câncer. O objetivo deste estudo foi avaliar fatores associados à "Não aderência" ao seguimento preconizado para mulheres com lesão intraepitelial de alto grau (HSIL), atendidas em um serviço de saúde da Baixada Fluminense, no Rio de Janeiro. Este estudo do tipo coorte retrospectivo incluiu mulheres rastreadas por citologia, matriculadas entre 01/01/2002 e 31/12/2005 e submetidas à colposcopia. A coleta de dados terminou em 31/12/07. Foram revisados 1496 prontuários e identificadas 641 mulheres elegíveis com diagnóstico de HSIL obtido por excisão da zona de transformação do colo uterino. Após a aplicação dos critérios de exclusão, a população de estudo foi constituída por 537 (84%) mulheres, classificadas em dois grupos: "não aderentes" (29,4%), que abandonaram o seguimento, e "aderentes" (70,6%), que permaneceram no seguimento até a alta. A análise estatística foi realizada pelo teste do qui-quadrado, teste t e regressão logística. O modelo final incluiu as variáveis: ser fumante (OR 1,72), dona de casa (OR 1,56), ter realizado o exame com o uso do vídeo-colposcópio (OR 1,80), idade (OR 0,97) e antecedente de três ou mais gestações (OR 0,49). O estudo revelou um perfil de vulnerabilidade apontando para determinantes de ordem individual e organizacional. Estratégias para melhorar a aderência ao seguimento devem contemplar medidas dirigidas a fatores modificáveis do estilo de vida, como o tabagismo, e estruturais característicos dos serviços de saúde da região.Cervical cancer is a tumor of multi-factorial nature that remains nowadays an a serious important public health problem. Adherence to health care procedures is a factor related to uterine cervical cancer control. The aim of this study was to evaluate factors associated with non-adherence to the prescribed follow-up in women with high-grade squamous intraepithelial lesions (HSIL), attending a public health care service in the "Baixada Fluminense", Rio de Janeiro, Brazil. This was a retrospective cohort study including women screened through cytology, entering the health care service between 01/01/2002 and 12/31/2005 and submitted to a colposcopy. Data collection ended on 12/31/07. Through the revision of 1496 medical records, 641 eligible women with a histopathological diagnosis of HSIL obtained after excision of the transformation zone were identified. After application of the exclusion criteria, the study population comprised 537 (84%) women, classified into two groups: "non-adherent" (29.4%), who abandoned the follow-up procedures and "adherent" (70.6%) who remained in follow-up along the required period. Statistical analysis was carried out though qui-square and t-tests and logistic regression. The final model contained the variables smoker (OR 1.72), not having a job outside the house (OR 1.56), having the examination carried out with a videocolposcopy (OR 1.80), age (OR 0.97) and history of three or more gestations (OR 0.49). The study disclosed a vulnerability profile pointing to individual and organizational-level determinants. Strategies to attain better follow-up must be aimed to modifiable life style factors as smoking and to structural characteristics of health care services in the studied area

    The Human Phenotype Ontology in 2017.

    Deep phenotyping has been defined as the precise and comprehensive analysis of phenotypic abnormalities in which the individual components of the phenotype are observed and described. The three components of the Human Phenotype Ontology (HPO; www.human-phenotype-ontology.org) project are the phenotype vocabulary, disease-phenotype annotations and the algorithms that operate on these. These components are being used for computational deep phenotyping and precision medicine as well as integration of clinical data into translational research. The HPO is being increasingly adopted as a standard for phenotypic abnormalities by diverse groups such as international rare disease organizations, registries, clinical labs, biomedical resources, and clinical software tools and will thereby contribute toward nascent efforts at global data exchange for identifying disease etiologies. This update article reviews the progress of the HPO project since the debut Nucleic Acids Research database article in 2014, including specific areas of expansion such as common (complex) disease, new algorithms for phenotype driven genomic discovery and diagnostics, integration of cross-species mapping efforts with the Mammalian Phenotype Ontology, an improved quality control pipeline, and the addition of patient-friendly terminology

    Strengthening the citizens' role in international organizations

    Today’s international organizations are characterized by a fundamental deficit in democracy. We therefore propose institutional measures to increase the direct participation possibilities of the citizens in international organizations. In order to reduce the number of citizens involved in decision-making to a manageable size, a representative sample of trustees is selected using a random mechanism. The trustees are given the right to launch initiatives and to vote in referendums on issues related to an international organization’s constitution. They can also recall executives when they are dissatisfied with their behavior. No specific changes to the organization of these entities, especially their executive function, are proposed. Rather, the executives of international organizations must obey the constitutional changes adopted by the trustees. The proposal gives international organizations democratic legitimacy. Moreover, the executives are subjected to the control of the citizens of the member states, which induces better responsiveness to the preferences of the people, as well as higher organizational efficiency

    Control of urea hydrolysis and nitrification in soil by chemicals - Prospects and problems

    A review is made of the recent work to assess the prospects of regulating urea hydrolysis and nitrification processes in soils by employing chemicals that can retard urea hydrolysis and nitrification. The possible benefits from control of nitrogen transformations in terms of conserving and enhancing fertilizer nitrogen efficiency for crop production and the problems associated with their use with regard to N metabolism of plants have also been discussed with examples. Prospects of using cheap and effective indigenous materials and chemicals for control of urea hydrolysis and nitrification under specific soil situations appear eminent in improving the fertilizer nitrogen efficiency. Urease inhibitors may be helpful in reducing problems associated with ammonia volatilization if this is not offset by leaching of urea. On the other hand retardation of nitrification appears useful in reducing losses that accompany nitrification due to leaching and denitrification, and with the plants that metabolize equally well with relatively higher amounts of NH4–N may be more effective in improving the utilization of fertilizer N under these situation
