Lake Characteristics Influencing Spawning Success of Muskellunge in Northern Wisconsin Lakes
We determined the physical, chemical, biological, and land use characteristics that distinguish northern Wisconsin lakes with selfâsustaining populations of muskellunge Esox masquinongy from lakes where stocking is required to maintain populations. Lakes that supported selfâsustaining muskellunge populations were characterized by fewer shoreline alterations and by spawning habitats with softer, organicânitrogenârich sediments. Lakes that required stocking had extensively developed shorelines. The direction of water level change during the spawning period, percentage of spawning area sediment covered by woody debris, number of deadfall trees per kilometer of shoreline, and percentage of shoreline that was totally developed were the most important variables for classifying the level of muskellunge reproduction a lake could support. A linear discriminant function correctly classified 83% of the lakes with selfâsustaining muskellunge populations and 89% of the lakes requiring stocking to sustain or enhance muskellunge populations. Lake managers wishing to use muskellunge stocking programs to reestablish selfâsustaining populations should critically review each candidate lake by considering our model and that of Dombeck et al. Multiple photosynthetic transitions, polyploidy, and lateral gene transfer in the grass subtribe Neurachninae
The Neurachninae is the only grass lineage known to contain C3, C4, and C3âC4 intermediate species, and as such has been suggested as a model system for studies of photosynthetic pathway evolution in the Poaceae; however, a lack of a robust phylogenetic framework has hindered this possibility. In this study, plastid and nuclear markers were used to reconstruct evolutionary relationships among Neurachninae species. In addition, photosynthetic types were determined with carbon isotope ratios, and genome sizes with flow cytometry. A high frequency of autopolyploidy was found in the Neurachninae, including in Neurachne munroi F.Muell. and Paraneurachne muelleri S.T.Blake, which independently evolved C4 photosynthesis. Phylogenetic analyses also showed that following their separate C4 origins, these two taxa exchanged a gene encoding the C4 form of phosphoenolpyruvate carboxylase. The C3âC4 intermediate Neurachne minor S.T.Blake is phylogenetically distinct from the two C4 lineages, indicating that intermediacy in this species evolved separately from transitional stages preceding C4 origins. The Neurachninae shows a substantial capacity to evolve new photosynthetic pathways repeatedly. Enablers of these transitions might include anatomical pre-conditions in the C3 ancestor, and frequent autopolyploidization. Transfer of key C4 genetic elements between independently evolved C4 taxa may have also facilitated a rapid adaptation of photosynthesis in these grasses that had to survive in the harsh climate appearing during the late Pliocene in Australia
Highly Variable Extinction and Accretion in the Jet-driving Class I Type Young Star PTF 10nvg (V2492 Cyg, IRAS 20496+4354)
We report extensive new photometry and spectroscopy of the highly variable
young stellar object PTF 10nvg including optical and near-infrared time series
data as well as mid-infrared and millimeter data. Following the previously
reported 2010 rise, during 2011 and 2012 the source underwent additional
episodes of brightening and dimming events including prolonged faint states.
The observed high-amplitude variations are largely consistent with extinction
changes having a 220 day quasi-periodic signal. Spectral evolution includes not
only changes in the spectral slope but correlated variation in the prominence
of TiO/VO/CO bands and atomic line emission, as well as anticorrelated
variation in forbidden line emission which, along with H_2, dominates optical
and infrared spectra at faint epochs. Neutral and singly-ionized atomic species
are likely formed in an accretion flow and/or impact while the origin of
zero-velocity atomic LiI 6707 in emission is unknown. Forbidden lines,
including several rare species, exhibit blueshifted emission profiles and
likely arise from an outflow/jet. Several of these lines are also seen
spatially offset from the continuum source position, presumably in a shocked
region of an extended jet. CARMA maps resolve on larger scales a spatially
extended outflow in mm-wavelength CO. We attribute the observed photometric and
spectroscopic behavior in terms of occultation of the central star as well as
the bright inner disk and the accretion/outflow zones that renders shocked gas
in the inner part of the jet amenable to observation at the faint epochs. We
discuss PTF 10nvg as a source exhibiting both accretion-driven (perhaps
analogous to V1647 Ori) and extinction-driven (perhaps analogous to UX Ori or
GM Cep) high-amplitude variability phenomena.Comment: accepted to AJ - in press (74 pages
HDAC-mediated control of ERK- and PI3K-dependent TGF-β-induced extracellular matrix-regulating genes
Histone deacetylases (HDACs) regulate the acetylation of histones in the control of gene expression. Many non-histone proteins are also targeted for acetylation, including TGF-Ă signalling pathway components such as Smad2, Smad3 and Smad7. Our studies in mouse C3H10T1/2 fibroblasts suggested that a number of TGF-Ă-induced genes that regulate matrix turnover are selectively regulated by HDACs. Blockade of HDAC activity with trichostatin A (TSA) abrogated the induction of a disintegrin and metalloproteinase 12 (Adam12) and tissue inhibitor of metalloproteinases-1 (Timp-1) genes by TGF-Ă, whereas plasminogen activator inhibitor-1 (Pai-1) expression was unaffected. Analysis of the activation of cell signalling pathways demonstrated that TGF-Ă induced robust ERK and PI3K activation with delayed kinetics compared to the phosphorylation of Smads. The TGF-Ă induction of Adam12 and Timp-1 was dependent on such non-Smad signalling pathways and, importantly, HDAC inhibitors completely blocked their activation without affecting Smad signalling. Analysis of TGF-Ă-induced Adam12 and Timp-1 expression and ERK/PI3K signalling in the presence of semi-selective HDAC inhibitors valproic acid, MS-275 and apicidin implicated a role for class I HDACs. Furthermore, depletion of HDAC3 by RNA interference significantly down-regulated TGF-Ă-induced Adam12 and Timp-1 expression without modulating Pai-1 expression. Correlating with the effect of HDAC inhibitors, depletion of HDAC3 also blocked the activation of ERK and PI3K by TGF-Ă. Collectively, these data confirm that HDACs, and in particular HDAC3, are required for activation of the ERK and PI3K signalling pathways by TGF-Ă and for the subsequent gene induction dependent on these signalling pathways
Macro-Climatic Distribution Limits Show Both Niche Expansion and Niche Specialization among C4 Panicoids
Grasses are ancestrally tropical understory species whose current dominance in warm open habitats is linked to the evolution of C4 photosynthesis. C4 grasses maintain high rates of photosynthesis in warm and water stressed environments, and the syndrome is considered to induce niche shifts into these habitats while adaptation to cold ones may be compromised. Global biogeographic analyses of C4 grasses have, however, concentrated on diversity patterns, while paying little attention to distributional limits. Using phylogenetic contrast analyses, we compared macro-climatic distribution limits among ~1300 grasses from the subfamily Panicoideae, which includes 4/5 of the known photosynthetic transitions in grasses. We explored whether evolution of C4 photosynthesis correlates with niche expansions, niche changes, or stasis at subfamily level and within the two tribes Paniceae and Paspaleae. We compared the climatic extremes of growing season temperatures, aridity, and mean temperatures of the coldest months. We found support for all the known biogeographic distribution patterns of C4 species, these patterns were, however, formed both by niche expansion and niche changes. The only ubiquitous response to a change in the photosynthetic pathway within Panicoideae was a niche expansion of the C4 species into regions with higher growing season temperatures, but without a withdrawal from the inherited climate niche. Other patterns varied among the tribes, as macro-climatic niche evolution in the American tribe Paspaleae differed from the pattern supported in the globally distributed tribe Paniceae and at family level.Fil: Aagesen, Lone. A Minimal Model of Metabolism Based Chemotaxis
Since the pioneering work by Julius Adler in the 1960's, bacterial chemotaxis has been predominantly studied as metabolism-independent. All available simulation models of bacterial chemotaxis endorse this assumption. Recent studies have shown, however, that many metabolism-dependent chemotactic patterns occur in bacteria. We hereby present the simplest artificial protocell model capable of performing metabolism-based chemotaxis. The model serves as a proof of concept to show how even the simplest metabolism can sustain chemotactic patterns of varying sophistication. It also reproduces a set of phenomena that have recently attracted attention on bacterial chemotaxis and provides insights about alternative mechanisms that could instantiate them. We conclude that relaxing the metabolism-independent assumption provides important theoretical advances, forces us to rethink some established pre-conceptions and may help us better understand unexplored and poorly understood aspects of bacterial chemotaxis

New Frontiers for Organismal Biology
Understanding how complex organisms function as integrated units that constantly interact with their environment is a long-standing challenge in biology. To address this challenge, organismal biology reveals general organizing principles of physiological systems and behaviorâin particular, in complex multicellular animals. Organismal biology also focuses on the role of individual variability in the evolutionary maintenance of diversity. To broadly advance these frontiers, cross-compatibility of experimental designs, methodological approaches, and data interpretation pipelines represents a key prerequisite. It is now possible to rapidly and systematically analyze complete genomes to elucidate genetic variation associated with traits and conditions that define individuals, populations, and species. However, genetic variation alone does not explain the varied individual physiology and behavior of complex organisms. We propose that such emergent properties of complex organisms can best be explained through a renewed emphasis on the context and life-history dependence of individual phenotypes to complement genetic data.Organismic and Evolutionary Biolog
