    A note on the antibiotic properties of Bacillus subtilis against Colletotrichum trifolii

    On a étudié l'influence et les mécanismes d'action in vitro et in vivoBacillus subtilissur le Colletotrichum trifolii, un champignon causant l'anthracnose de la luzerne (Medicago sativa). Lors d'essais en chambre de culture, l'application du filtrat du bouillon nutritif du B. subtilis a réduit significativement l'incidence et la gravité de la maladie de respectivement 56 à 16% et de 2,0 à 1,2. L'application de cellules lavées du B. subtilis n'a pas entraßné une diminution de l'incidence et de la gravité de la maladie. Sur les folioles de la luzerne, le filtrat a causé une réduction de la germination des conidies, la lyse des conidies et une formation réduite d'appressoriums. In vitro, il a réduit la germination des conidies, induit la lyse des conidies et la formation de tubes germinatifs enflés. Un antibiotique de la famille des iturines, l'iturine D, serait l'agent actif dans la suppression du C. trifolii par le B. subtilis.The influence and mechanisms of action of Bacillus subtilis on Colletotrichum trifolii, a causal agent of anthracnose of alfalfa (Medicago sativa), were studied in vivo and in vitro. In growth room conditions, a cell-free culture filtrate of B. subtilis significantly reduced disease incidence and severity on alfalfa seedlings from 56% to 16% and from 2.0 to 1.2, respectively. Treatment of seedlings with washed cell suspensions of B. subtilis had no influence on disease. Applications of crude filtrate on alfalfa leaflets inoculated with C. trifolii were associated with reduced germination of conidia, lysis of conidia, and reduced formation of appressoria. Under in vitro conditions, crude filtrate reduced germination of conidia, and induced lysis of conidia and the formation of inflated germ tubes on germinating conidia. An antibiotic of the iturin family, iturin D, was tentatively identified as the active compound responsible for the suppressive effect of B. subtilis on C. trifolii

    Transition between Two Regimes Describing Internal Fluctuation of DNA in a Nanochannel

    We measure the thermal fluctuation of the internal segments of a piece of DNA confined in a nanochannel about 50100 nm wide. This local thermodynamic property is key to accurate measurement of distances in genomic analysis. For DNA in 100 nm channels, we observe a critical length scale 10 m for the mean extension of internal segments, below which the de Gennes' theory describes the fluctuations with no fitting parameters, and above which the fluctuation data falls into Odijk's deflection theory regime. By analyzing the probability distributions of the extensions of the internal segments, we infer that folded structures of length 150250 nm, separated by 10 m exist in the confined DNA during the transition between the two regimes. For 50 nm channels we find that the fluctuation is significantly reduced since the Odijk regime appears earlier. This is critical for genomic analysis. We further propose a more detailed theory based on small fluctuations and incorporating the effects of confinement to explicitly calculate the statistical properties of the internal fluctuations. Our theory is applicable to polymers with heterogeneous mechanical properties confined in non-uniform channels. We show that existing theories for the end-to-end extension/fluctuation of polymers can be used to study the internal fluctuations only when the contour length of the polymer is many times larger than its persistence length. Finally, our results suggest that introducing nicks in the DNA will not change its fluctuation behavior when the nick density is below 1 nick per kbp DNA

    Transrenal Deployment of a Modular Stent Graft to Repair AAAs with Short Necks: Experiments in Dogs

    Severely angulated (> 60°) or short (< 15mm) proximal necks remain significant anatomical limitations for endovascular stent-graft repairs for abdominal aortic aneurysms. Ensuring proper proximal fixation of the stent-graft to the host artery without the short-or long-term risks of endoleak or migration represents a particular technical challenge for these anatomical circumstances. An innovative balloon expandable stent combined with a weft-knitted prosthesis was specifically designed for these situations by modelling the stent to the neck anatomy without overdistension or potential barotrauma allowing better incorporation of the device. The Latecba stent-graft consists of a 2 parts modular design. The first one, Module A, is deployed at the transrenal level and consists of a Palmaz type stent whose first half is bare and second half is sutured to a crimped weft-knitted polyester graft whose distal end holds a constriction. The second Module B is a non-crimped weft-knitted graft attached to 2 stainless steel stents. The first stent is entirely contained in the proximal textile tube, allowing fixation to module A. The second stent, which is left uncovered over the distal third, ensures proper fixation of the stent-graft distally. Following the creation of a prosthetic aneurysm in the infrarenal aorta in 32 dogs, 29 received the Latecba stent-graft for scheduled durations of 10 days, 1 month, 3 months and 6 months. Proper deployment of the stent-grafts was achieved without difficulty. All 29 animals survived and the devices were all patent at sacrifice. No device defects or migrations were observed and the stent-grafts proved to be efficient in this setting to exclude the aneurysm. Analyses of the explanted devices (gross observations, RX, CT scan, IVUS, angioscopy) confirmed the stability of this modular stent-graft. Further on-going clinical investigations are warranted to validate this concept before this stent-graft becomes commercially available without any restriction

    Coordination and resource-related difficulties encountered by Quebec's public health specialists and infectious diseases/medical microbiologists in the management of A (H1N1) - a mixed-method, exploratory survey

    <p>Abstract</p> <p>Background</p> <p>In Quebec, the influenza A (H1N1) pandemic was managed using a top-down style that left many involved players with critical views and frustrations. We aimed to describe physicians' perceptions - infectious diseases specialists/medical microbiologists (IDMM) and public health/preventive medicine specialists (PHPMS) - in regards to issues encountered with the pandemics management at the physician level and highlight suggested improvements for future healthcare emergencies.</p> <p>Methods</p> <p>In April 2010, Quebec IDMM and PHPMS physicians were invited to anonymously complete a web-based learning needs assessment. The survey included both open-ended and multiple-choice questions. Descriptive statistics were used to report on the frequency distribution of multiple choice responses whereas thematic content analysis was used to analyse qualitative data generated from the survey and help understand respondents' experience and perceptions with the pandemics.</p> <p>Results</p> <p>Of the 102 respondents, 85.3% reported difficulties or frustrations in their practice during the pandemic. The thematic analysis revealed two core themes describing the problems experienced in the pandemic management: coordination and resource-related difficulties. Coordination issues included communication, clinical practice guidelines, decision-making, roles and responsibilities, epidemiological investigation, and public health expert advisory committees. Resources issues included laboratory resources, patient management, and vaccination process.</p> <p>Conclusion</p> <p>Together, the quantitative and qualitative data suggest a need for improved coordination, a better definition of roles and responsibilities, increased use of information technologies, merged communications, and transparency in the decisional process. Increased flexibility and less contradiction in clinical practice guidelines from different sources and increased laboratory/clinical capacity were felt critical to the proper management of infectious disease emergencies.</p

    Helium trapping in apatite damage: insights from (U-Th-Sm)/He dating of different granitoid lithologies

    Apatite (U-Th-Sm)/He (AHe) thermochronometry is widely used to constrain thermal histories and rates of tectonic, exhumation, and erosion processes. However, data interpretation is often challenging, especially when the thermal history includes extended residence time in the He partial retention zone (HePRZ), with highly dispersed dates revealing the complexity of diffusion processes in natural systems. This study investigates chemical and physical factors that may have impacted He diffusion in apatite over long timescales in a context of protracted residence in the HePRZ. Nine samples from the Ploumanac'h pluton and North Tregor (Armorican Massif, France) were collected in granitoids, differing in petrography and chemisty. This area was chosen because these samples underwent a similar thermal history since ~300 Ma. We report new (U-Th Sm)/He dates, along with apatite fission-track (AFT) data, as well as lithological and chemical characterization. The results show dispersed (U-Th-Sm)/He dates, ranging from 87 ± 7 to 291 ± 23 Ma, whereas central AFT dates vary from 142 ± 6 to 199 ± 9 Ma. Current predictive models for He diffusion and fission-track annealing in apatite could not reproduce the two datasets together. However, this apparent discrepancy gives insight into the parameters influencing He diffusion at geological timescales. The data confirm that radiation damage enhances He trapping, as the AHe dates are positively correlated to effective uranium (eU) concentration. The He age dispersion for constant eU content cannot be explained just by variations in grain size or chemical composition. To explore the potential influence of recoil damage trapping behavior and annealing kinetics on AHe dates, we tested a new diffusion model from Gerin et al. (2017). Given the expected model of the thermal history provided by AFT inversion, we investigated the influence of the trapping energy on AHe dates. The AHe date variations can be explained only if the trapping energy evolves from one crystal to another, increasing with the amount of damage. For a given trapping energy, minor variations in the recoil-damage annealing rate can consistently explain most of the remaining dispersion of the AHe dates

    Liposarcoma cells with aldefluor and CD133 activity have a cancer stem cell potential

    Aldehyde dehydrogenase (ALDH) has recently been shown to be a marker of cancer stem-like cells (CSCs) across tumour types. The primary goals of this study were to investigate whether ALDH is expressed in liposarcomas, and whether CSCs can be identified in the ALDHhigh subpopulation. We have demonstrated that ALDH is indeed expressed in 10 out of 10 liposarcoma patient samples. Using a liposarcoma xenograft model, we have identified a small population of cells with an inducible stem cell potential, expressing both ALDH and CD133 following culturing in stem cell medium. This potential CSC population, which makes up for 0, 1-1, 7% of the cells, displayed increased self-renewing abilities and increased tumourigenicity, giving tumours in vivo from as few as 100 injected cells

    Compartmentalized Culture of Perivascular Stroma and Endothelial Cells in a Microfluidic Model of the Human Endometrium

    The endometrium is the inner lining of the uterus. Following specific cyclic hormonal stimulation, endometrial stromal fibroblasts (stroma) and vascular endothelial cells exhibit morphological and biochemical changes to support embryo implantation and regulate vascular function, respectively. Herein, we integrated a resin-based porous membrane in a dual chamber microfluidic device in polydimethylsiloxane that allows long term in vitro co-culture of human endometrial stromal and endothelial cells. This transparent, 2-m porous membrane separates the two chambers, allows for the diffusion of small molecules and enables high resolution bright field and fluorescent imaging. Within our primary human co-culture model of stromal and endothelial cells, we simulated the temporal hormone changes occurring during an idealized 28-day menstrual cycle. We observed the successful differentiation of stroma into functional decidual cells, determined by morphology as well as biochemically as measured by increased production of prolactin. By controlling the microfluidic properties of the device, we additionally found that shear stress forces promoted cytoskeleton alignment and tight junction formation in the endothelial layer. Finally, we demonstrated that the endometrial perivascular stroma model was sustainable for up to 4 weeks, remained sensitive to steroids and is suitable for quantitative biochemical analysis. Future utilization of this device will allow the direct evaluation of paracrine and endocrine crosstalk between these two cell types as well as studies of immunological events associated with normal versus disease-related endometrial microenvironments

    Mineralogical and geochemical features of sulfide chimneys from the 49°39â€ČE hydrothermal field on the Southwest Indian Ridge and their geological inferences

The definitive version was published in Chinese Science Bulletin 56 (2011): 2828-2838, doi:10.1007/s11434-011-4619-4.During January–May in 2007, the Chinese research cruise DY115-19 discovered an active hydrothermal field at 49°39â€ČE/37°47â€ČS on the ultraslow spreading Southwest Indian Ridge (SWIR). This was also the first active hydrothermal field found along an ultraslow-spreading ridge. We analyzed mineralogical, textural and geochemical compositions of the sulfide chimneys obtained from the 49°39â€ČE field. Chimney samples show a concentric mineral zone around the fluid channel. The mineral assemblages of the interiors consist mainly of chalcopyrite, with pyrite and sphalerite as minor constitunets. In the intermediate portion, pyrite becomes the dominant mineral, with chalcopyrite and sphalerite as minor constitunets. For the outer wall, the majority of minerals are pyrite and sphalerite, with few chalcopyrite. Towards the outer margin of the chimney wall, the mineral grains become small and irregular in shape gradually, while minerals within interstices are abundant. These features are similar to those chimney edifices found on the East Pacific Rise and Mid-Atlantic Ridge. The average contents of Cu, Fe and Zn in our chimney samples were 2.83 wt%, 45.6 wt% and 3.28 wt%, respectively. The average Au and Ag contents were up to 2.0 ppm and 70.2 ppm respectively, higher than the massive sulfides from most hydrothermal fields along mid-ocean ridge. The rare earth elements geochemistry of the sulfide chimneys show a pattern distinctive from the sulfides recovered from typical hydrothermal fields along sediment-starved mid-ocean ridge, with the enrichment of light rare earth elements but the weak, mostly negative, Eu anomaly.
