With the purpose to study the presence of swine leptospiral carrier,131 blood samples and the kidneys from the respective animals were taken during slaughter in abattoir house in Botucatu - SP region. By the Microscopic Agglutination Test (MAT), 48 serum samples were positive against one or more serovars, with a rate of the occurrence of 36.64% to anti-leptospiral antibodies and greater importance to the serovar icterohaemorrhagiae. The investigation of the agent, a total of 88 kidney samples were submitted to culture in EMJH media and analysed by PCR. From those 88 kidney samples, 48 were of serologically positive animals and 40 of negative animals, being isolated and detected the agent in one kidney sample, belonging to a seropositive animal. Although was not possible the statistical evaluation in terms of sensitivity and specificity of the two test to detection, the PCR showed faster and most practical to study leptospiral carriers. By the results of isolation it must be emphasized the importance of those animals as the transmitters of the disease to slaughter house workers and to meat inspectors.Com o intuito de se estudar a presença de suínos portadores renais de leptospiras foram colhidas 131 amostras sanguíneas e os respectivos rins de animais durante o abate em abatedouro da região de Botucatu-SP. Pela prova de Soroaglutinação Microscópica obteve-se 48 amostras sorológicas positivas para um ou mais sorovar de Leptospira spp., com uma taxa de ocorrência de anticorpos anti-leptospira de 36,64%, e maior importância para o sorovar icterohaemorrhagiae. Para a pesquisa do agente nos rins, das 88 amostras renais submetidas a cultura em meio de EMJH e analisadas pela prova de PCR, foi isolado e detectado o agente em uma única amostra renal, pertencente a um animal soropositivo. Embora não tenha sido possível a comparação estatística, em termos de sensibilidade e especificidade das duas provas de detecção do agente a partir de amostras renais, a PCR mostrou-se mais rápida e prática na pesquisa de portadores renais. Gene expression has been extensively studied in plant science research, mainly for the assessment of plant stress responses. Real-time-quantitative polymerase chain reaction (RT-qPCR) is an important tool for obtaining this information because it is a quick and easy technique to acquire a large amount of molecular data for both model and non-model plants. For a successful RT-qPCR analysis, gene expression should be carefully normalised. Genes involved in essential biological processes that exhibit constitutive expression are commonly selected as internal standards to normalise RT-qPCR experiments. In this study, the transcription profiles of 13 candidate reference genes for RT-qPCR were evaluated in three guarana cultivars (BRS-Amazonas, BRS-Maués and BRS-Luzéia) using different tissues (vegetative and fruit) in varying developmental stages. Two different algorithms, NormFinder and GeNorm, were utilised to assess gene stability. In general, the two algorithms did not select the same pairs of genes for all analysed conditions. For the largest group (the fruits of all cultivars), NormFinder selected the pair EF1A/UBQ, whereas GeNorm chose ACT/GAPDH as the best normalising genes. Thus, we recommend the use of at least four reference genes for the normalisation of gene expression in guarana plant studies. © 2015, Botanical Society of Sao Paulo

PRÓPOLIS NO CONTROLE DA MASTITE BOVINA

O consumidor em diferentes países exige cada vez mais alimentos naturais e de melhor qualidade. A mastite caracteriza-se pelo aumento da contagem de células somáticas, diminuição de caseína, lactose e gordura, gerando prejuízos para atividade. A própolis não acarreta prejuízos ao animal, sendo inócua quando adicionada à dieta humana. Dessa forma, objetivou-se verificar as atividades terapêuticas da própolis no controle de mastite bovina. Foram utilizadas 72 fêmeas Holandesas, provenientes de rebanho comercial, cuja mastite foi identificada pelo Teste de Tamis, Califórnia Mastite Teste, contagem de células somáticas e exame microbiológico do leite. Realizou-se quatro tratamentos, sendo Grupo EAP1: 10mL de extrato alcoólico de própolis a 30%, via oral, durante sete dias consecutivos; Grupo EAP2: além do procedimento descrito para Grupo EAP1, utilizou-se o EAP na imersão dos tetos antes e após a ordenha; Grupo CA: utilizou-se álcool de cereais na imersão dos tetos antes e após a ordenha; e Grupo CT: os animais controle-testemunha foram submetidos aos procedimentos rotineiramente empregados pela propriedade. Os resultados foram analisados por técnica de variância não paramétrica para modelo de medidas repetidas em grupos independentes complementada por comparações múltiplas. Observou-se diminuição na contagem de células somáticas em todos os grupos avaliados. As atividades biológicas da própolis propiciam grandes perspectivas, entretanto, para as condições avaliadas, não foi possível observar diferença entre os tratamentos. A alta diversidade em sua composição química, além da complexidade dos múltiplos mecanismos sinérgicos envolvidos em suas atividades biológicas, exige adicionais ensaios clínicos a fim de se determinar a terapêutica capaz de demonstrar seus benefícios “in vivo”

Isolation and characterization of ten microsatellite loci in\ud stingless bee Trigona spinipes (Apidae: Meliponini)

Stingless bees are the most abundant pollinators of Brazilian tropical flora. Trigona spinipes has some of the\ud largest colonies of any stingless bee species found in several types of environment. This work describes the\ud isolation and characterization of microsatellite loci for this species. A microsatellite-enriched genomic library\ud was constructed and ten primer pairs were designed for T. spinipes. The primers were tested in 20 unrelated\ud individuals. The mean number of alleles was 8.10 and mean observed and expected heterozygosity were\ud 0.655 and 0.680, respectively. Primers were also tested in cross-species amplification and five loci were\ud successfully amplified inTrigona chanchamayoensis,Trigona hyalinata,Tetragonisca angustula, Partamona\ud mulata and Frieseomelitta varia. Integrated monitoring of mola mola behaviour in space and time

Over the last decade, ocean sunfish movements have been monitored worldwide using various satellite tracking methods. This study reports the near-real time monitoring of finescale (< 10 m) behaviour of sunfish. The study was conducted in southern Portugal in May 2014 and involved satellite tags and underwater and surface robotic vehicles to measure both the movements and the contextual environment of the fish. A total of four individuals were tracked using custom-made GPS satellite tags providing geolocation estimates of fine-scale resolution. These accurate positions further informed sunfish areas of restricted search (ARS), which were directly correlated to steep thermal frontal zones. Simultaneously, and for two different occasions, an Autonomous Underwater Vehicle (AUV) videorecorded the path of the tracked fish and detected buoyant particles in the water column. Importantly, the densities of these particles were also directly correlated to steep thermal gradients. Thus, both sunfish foraging behaviour (ARS) and possibly prey densities, were found to be influenced by analogous environmental conditions. In addition, the dynamic structure of the water transited by the tracked individuals was described by a Lagrangian modelling approach. The model informed the distribution of zooplankton in the region, both horizontally and in the water column, and the resultant simulated densities positively correlated with sunfish ARS behaviour estimator (r(s) = 0.184, p < 0.001). The model also revealed that tracked fish opportunistically displace with respect to subsurface current flow. Thus, we show how physical forcing and current structure provide a rationale for a predator's finescale behaviour observed over a two weeks in May 2014
