    Atmospheric instability analysis and its relationship to precipitation patterns over the western Iberian Peninsula

    International audienceThermodynamic and dynamic atmospheric stability indices will be analysed in order to classify the rainfall types occurred in western Iberia and provide information about the associated meteorological phenomena and preferred regional instability sources. In this work, instability sources over the western Iberian Peninsula will be characterised by means of radiosonde station data and reanalysis data sets from the European Centre for Medium-Range Weather Forecasts (ECMWF), for the period 1971?2002. Dynamic stability indices such as the Q vector divergence (dQ) or the potential vorticity anomaly in the 330 K isentropic surface (PV) have been calculated with the aim of establishing different atmospheric stability scenarios. Furthermore, thermodynamic atmospheric stability contribution has been also evaluated with Total Totals index (TT) and moisture availability was inferred from circulation weather types (CWTs). The first results of this work assess the combined dynamic-thermodynamic preferred conditions, synoptic situations and humidity sources, leading to precipitation events over the western Iberian Peninsula and prove the essential role of CWT patterns on the rainfall genesis

    Linking data and BPMN processes to achieve executable models

    We describe a formally well founded approach to link data and processes conceptually, based on adopting UML class diagrams to represent data, and BPMN to represent the process. The UML class diagram together with a set of additional process variables, called Artifact, form the information model of the process. All activities of the BPMN process refer to such an information model by means of OCL operation contracts. We show that the resulting semantics while abstract is fully executable.     Time-varying effective EEG source connectivity: the optimization of model parameters*

    Adaptive estimation methods based on general Kalman filter are powerful tools to investigate brain networks dynamics given the non-stationary nature of neural signals. These methods rely on two parameters, the model order p and adaptation constant c, which determine the resolution and smoothness of the time-varying multivariate autoregressive estimates. A sub-optimal filtering may present consistent biases in the frequency domain and temporal distortions, leading to fallacious interpretations. Thus, the performance of these methods heavily depends on the accurate choice of these two parameters in the filter design. In this work, we sought to define an objective criterion for the optimal choice of these parameters. Since residual- and information-based criteria are not guaranteed to reach an absolute minimum, we propose to study the partial derivatives of these functions to guide the choice of p and c. To validate the performance of our method, we used a dataset of human visual evoked potentials during face perception where the generation and propagation of information in the brain is well understood and a set of simulated data where the ground truth is available

    CDK-dependent nuclear localization of B-Cyclin Clb1 promotes FEAR activation during meiosis I in budding yeast

    Cyclin-dependent kinases (CDK) are master regulators of the cell cycle in eukaryotes. CDK activity is regulated by the presence, post-translational modification and spatial localization of its regulatory subunit cyclin. In budding yeast, the B-cyclin Clb1 is phosphorylated and localizes to the nucleus during meiosis I. However the functional significance of Clb1's phosphorylation and nuclear localization and their mutual dependency is unknown. In this paper, we demonstrate that meiosis-specific phosphorylation of Clb1 requires its import to the nucleus but not vice versa. While Clb1 phosphorylation is dependent on activity of both CDK and polo-like kinase Cdc5, its nuclear localization requires CDK but not Cdc5 activity. Furthermore we show that increased nuclear localization of Clb1 during meiosis enhances activation of FEAR (Cdc Fourteen Early Anaphase Release) pathway. We discuss the significance of our results in relation to regulation of exit from meiosis I

    Nucleosome eviction from MHC class II promoters controls positioning of the transcription start site

    Nucleosome depletion at transcription start sites (TSS) has been documented genome-wide in multiple eukaryotic organisms. However, the mechanisms that mediate this nucleosome depletion and its functional impact on transcription remain largely unknown. We have studied these issues at human MHC class II (MHCII) genes. Activation-induced nucleosome free regions (NFR) encompassing the TSS were observed at all MHCII genes. Nucleosome depletion was exceptionally strong, attaining over 250-fold, at the promoter of the prototypical HLA-DRA gene. The NFR was induced primarily by the transcription factor complex that assembles on the conserved promoter-proximal enhancer situated upstream of the TSS. Functional analyses performed in the context of native chromatin demonstrated that displacing the NFR without altering the sequence of the core promoter induced a shift in the position of the TSS. The NFR thus appears to play a critical role in transcription initiation because it directs correct TSS positioning in vivo. Our results provide support for a novel mechanism in transcription initiation whereby the position of the TSS is controlled by nucleosome eviction rather than by promoter sequenc

    Synthesis of OCL Pre-conditions for Graph Transformation Rules

Málaga, Spain, 28 June-02 July, 2010Graph transformation (GT) is being increasingly used in Model Driven Engineering (MDE) to describe in-place transformations like animations and refactorings. For its practical use, rules are often complemented with OCL application conditions. The advancement of rule post-conditions into pre-conditions is a well-known problem in GT, but current techniques do not consider OCL. In this paper we provide an approach to advance post-conditions with arbitrary OCL expressions into pre-conditions.     Encapsulation of Phenolic Compounds from a Grape Cane Pilot-Plant Extract in Hydroxypropyl Beta-Cyclodextrin and Maltodextrin by Spray Drying

    Grape canes, the main byproducts of the viticulture industry, contain high-value bioactive phenolic compounds, whose application is limited by their instability and poorly solubility in water. Encapsulation in cyclodextrins allows these drawbacks to be overcome. In this work, a grape cane pilot-plant extract (GCPPE) was encapsulated in hydroxypropyl beta-cyclodextrin (HP-β-CD) by a spray-drying technique and the formation of an inclusion complex was confirmed by microscopy and infrared spectroscopy. The phenolic profile of the complex was analyzed by LC-ESI-LTQ-Orbitrap-MS and the encapsulation efficiency of the phenolic compounds was determined. A total of 42 compounds were identified, including stilbenes, flavonoids, and phenolic acids, and a complex of (epi)catechin with β-CD was detected, confirming the interaction between polyphenols and cyclodextrin. The encapsulation efficiency for the total extract was 80.5 ± 1.1%, with restrytisol showing the highest value (97.0 ± 0.6%) and (E)-resveratrol (32.7 ± 2.8%) the lowest value. The antioxidant capacity of the inclusion complex, determined by ORAC-FL, was 5300 ± 472 µmol TE/g DW, which was similar to the value obtained for the unencapsulated extract. This formulation might be used to improve the stability, solubility, and bioavailability of phenolic compounds of the GCPPE for water-soluble food and pharmaceutical applications

    Extensive remodeling of DC function by rapid maturation-induced transcriptional silencing

    The activation, or maturation, of dendritic cells (DCs) is crucial for the initiation of adaptive T-cell mediated immune responses. Research on the molecular mechanisms implicated in DC maturation has focused primarily on inducible gene-expression events promoting the acquisition of new functions, such as cytokine production and enhanced T-cell-stimulatory capacity. In contrast, mechanisms that modulate DC function by inducing widespread gene-silencing remain poorly understood. Yet the termination of key functions is known to be critical for the function of activated DCs. Genome-wide analysis of activation-induced histone deacetylation, combined with genome-wide quantification of activation-induced silencing of nascent transcription, led us to identify a novel inducible transcriptional-repression pathway that makes major contributions to the DC-maturation process. This silencing response is a rapid primary event distinct from repression mechanisms known to operate at later stages of DC maturation. The repressed genes function in pivotal processes—including antigen-presentation, extracellular signal detection, intracellular signal transduction and lipid-mediator biosynthesis—underscoring the central contribution of the silencing mechanism to rapid reshaping of DC function. Interestingly, promoters of the repressed genes exhibit a surprisingly high frequency of PU.1-occupied sites, suggesting a novel role for this lineage-specific transcription factor in marking genes poised for inducible repressio
