    Photo-controlled permeation of spiropyran modified gramicidin A ion channel

Gramicidin A (gA) is a well studied ionophore that can be easily modified at the C-terminus to be incorporated into phosphotidylcholine bilayer membrane systems. Potassium permeation of modified gA attached to spiropyran can be controlled with light. Upon ultra-violet irradiation spiropyran transforms to the more polar form merocyanine. The process back to spiropyran is completely reversible upon irradiation with 550 nm light or thermally. Free bilayer membrane vesicles are employed to describe the characteristics of modified ion channels. Characteristics of gA modified with spiropyran are described herein. A device has been created and characterized using electrochemical impedance spectroscopy to analyze potassium permeation through a tethered bilayer membrane system (tBMS) on a sheet of gold utilizing sulfur anchors. The device consists of a tethered phase and a mobile upper phase. The mobile lipid layer incorporates gA modified with spiropyran. The modification allows for control of potassium permeation across the tBMS. Impedance analysis shows good agreement with the ability to control potassium permeation to that of the free vesicle

    Twisted four-dimensional supersymmetric Yang-Mills theory famously gives a useful point of view on the Donaldson and Seiberg-Witten invariants of four-manifolds. In this paper we generalize the construction to include a path integral formulation of generalizations of Donaldson invariants for smooth families of four-manifolds. Mathematically these are equivariant cohomology classes for the action of the oriented diffeomorphism group on the space of metrics on the manifold. In principle these cohomology classes should contain nontrivial information about the topology of the diffeomorphism group of the four-manifold.     A Spitzer Transmission Spectrum for the Exoplanet GJ 436b, Evidence for Stellar Variability, and Constraints on Dayside Flux Variations

    In this paper we describe a uniform analysis of eight transits and eleven secondary eclipses of the extrasolar planet GJ 436b obtained in the 3.6, 4.5, and 8.0 micron bands using the IRAC instrument on the Spitzer Space Telescope between UT 2007 June 29 and UT 2009 Feb 4. We find that the best-fit transit depths for visits in the same bandpass can vary by as much as 8% of the total (4.7 sigma significance) from one epoch to the next. Although we cannot entirely rule out residual detector effects or a time-varying, high-altitude cloud layer in the planet's atmosphere as the cause of these variations, we consider the occultation of active regions on the star in a subset of the transit observations to be the most likely explanation. We reconcile the presence of magnetically active regions with the lack of significant visible or infrared flux variations from the star by proposing that the star's spin axis is tilted with respect to our line of sight, and that the planet's orbit is therefore likely to be misaligned. These observations serve to illustrate the challenges associated with transmission spectroscopy of planets orbiting late-type stars; we expect that other systems, such as GJ 1214, may display comparably variable transit depths. Our measured 8 micron secondary eclipse depths are consistent with a constant value, and we place a 1 sigma upper limit of 17% on changes in the planet's dayside flux in this band. Averaging over the eleven visits gives us an improved estimate of 0.0452% +/- 0.0027% for the secondary eclipse depth. We combine timing information from our observations with previously published data to produce a refined orbital ephemeris, and determine that the best-fit transit and eclipse times are consistent with a constant orbital period.     The AllWISE Motion Survey and the Quest for Cold Subdwarfs

    The AllWISE processing pipeline has measured motions for all objects detected on Wide-field Infrared Survey Explorer (WISE) images taken between 2010 January and 2011 February. In this paper, we discuss new capabilities made to the software pipeline in order to make motion measurements possible, and we characterize the resulting data products for use by future researchers. Using a stringent set of selection criteria, we find 22,445 objects that have significant AllWISE motions, of which 3525 have motions that can be independently confirmed from earlier Two Micron All Sky Survey (2MASS) images, yet lack any published motions in SIMBAD. Another 58 sources lack 2MASS counterparts and are presented as motion candidates only. Limited spectroscopic follow-up of this list has already revealed eight new L subdwarfs. These may provide the first hints of a "subdwarf gap" at mid-L types that would indicate the break between the stellar and substellar populations at low metallicities (i.e., old ages). Another object in the motion list—WISEA J154045.67–510139.3—is a bright (J ≈ 9 mag) object of type M6; both the spectrophotometric distance and a crude preliminary parallax place it ~6 pc from the Sun. We also compare our list of motion objects to the recently published list of 762 WISE motion objects from Luhman. While these first large motion studies with WISE data have been very successful in revealing previously overlooked nearby dwarfs, both studies missed objects that the other found, demonstrating that many other nearby objects likely await discovery in the AllWISE data products

    A Micro RNA Processing Defect in Rapidly Progressing Idiopathic Pulmonary Fibrosis

    BACKGROUND: Idiopathic pulmonary fibrosis exhibits differential progression from the time of diagnosis but the molecular basis for varying progression rates is poorly understood. The aim of the present study was to ascertain whether differential miRNA expression might provide one explanation for rapidly versus slowly progressing forms of IPF. METHODOLOGY AND PRINCIPAL FINDINGS: miRNA and mRNA were isolated from surgical lung biopsies from IPF patients with a clinically documented rapid or slow course of disease over the first year after diagnosis. A quantitative PCR miRNA array containing 88 of the most abundant miRNA in the human genome was used to profile lung biopsies from 9 patients with rapidly progressing IPF, 6 patients with slowly progressing IPF, and 10 normal lung biopsies. Using this approach, 11 miRNA were significantly increased and 36 were significantly decreased in rapid biopsies compared with normal biopsies. Slowly progressive biopsies exhibited 4 significantly increased miRNA and 36 significantly decreased miRNA compared with normal lung. Among the miRNA present in IPF with validated mRNA targets were those with regulatory effects on epithelial-mesenchymal transition (EMT). Five miRNA (miR-302c, miR-423-5p, miR-210, miR-376c, and miR-185) were significantly increased in rapid compared with slow IPF lung biopsies. Additional analyses of rapid biopsies and fibroblasts grown from the same biopsies revealed that the expression of AGO1 and AGO2 (essential components of the miRNA processing RISC complex) were lower compared with either slow or normal lung biopsies and fibroblasts. CONCLUSION: These findings suggest that the development and/or clinical progression of IPF might be the consequence of aberrant miRNA processing

    Large Scale Searches for Brown Dwarfs and Free-Floating Planets

    Searches of large scale surveys have resulted in the discovery of over 1000 brown dwarfs in the Solar neighbourhood.
