    Assessing the Consequences of Brood Parasitism and Nest Predation On Seasonal Fecundity in Passerine Birds

    Brood parasites and nest predators reduce the seasonal fecundity and, hence, the population growth rates of their victims. However, most field studies do not measure directly how parasites and predators decrease seasonal fecundity, but instead measure the impact of these organisms on individual nesting attempts. Because a female may renest after losing a nest to predation, abandoning a parasitized nest, or successfully fledging a brood, knowing how brood parasites and nest predators reduce the number of offspring fledged from individual nesting attempts is not equivalent to knowing their impact on seasonal fecundity. We address this problem by developing a mathematical model that: estimates several parameters describing the natural history of this system, including the brood-parasitism rate, nest-predation rate, and probability of nest abandonment in response to a parasitism event; and extrapolates to seasonal fecundity from these parameters and others describing the length of the breeding season, the timing of events in the nesting cycle, and the productivity of parasitized and unparasitized nests. We also show how different researchers using different observational methodologies to study exactly the same population likely would arrive at noticeably different conclusions regarding the intensity of brood parasitism, and we provide mathematical formulas for comparing among several of these measures of parasitism. Our procedures extend Mayfield's method for calculating nest-success rates from nest-history data in that we simultaneously estimate parameters describing nest predation and brood parasitism, predict seasonal fecundity from these parameters, and provide confidence intervals on all parameter estimates. The model should make the design and interpretation of logistically difficult empirical studies more efficient. It also can be specialized to species affected by nest predators but not brood parasites. We use the model to analyze prairie Warbler (Dendroica discolor) and Black-capped Vireo (Vireo atricapillus) nesting data. We estimate the model's parameters for these species and use the resulting estimates to predict seasonal fecundity.     Effective area calibration of the reflection grating spectrometers of XMM-Newton. II. X-ray spectroscopy of DA white dwarfs

    White dwarf spectra have been widely used as a calibration source for X-ray and EUV instruments. The in-flight effective area calibration of the reflection grating spectrometers (RGS) of XMM-Newton depend upon the availability of reliable calibration sources. We investigate how well these white dwarf spectra can be used as standard candles at the lowest X-ray energies in order to gauge the absolute effective area scale of X-ray instruments. We calculate a grid of model atmospheres for Sirius B and HZ 43A, and adjust the parameters using several constraints until the ratio of the spectra of both stars agrees with the ratio as observed by the low energy transmission grating spectrometer (LETGS) of Chandra. This ratio is independent of any errors in the effective area of the LETGS. We find that we can constrain the absolute X-ray spectrum of both stars with better than 5 % accuracy. The best-fit model for both stars is close to a pure hydrogen atmosphere, and we put tight limits to the amount of helium or the thickness of a hydrogen layer in both stars. Our upper limit to the helium abundance in Sirius B is 4 times below the previous detection based on EUVE data. We also find that our results are sensitive to the adopted cut-off in the Lyman pseudo-continuum opacity in Sirius B. We get best agreement with a long wavelength cut-off. White dwarf model atmospheres can be used to derive the effective area of X-ray spectrometers in the lowest energy band.     ALLN-177, oral enzyme therapy for hyperoxaluria

    PURPOSE: To evaluate the potential of ALLN-177, an orally administered, oxalate-specific enzyme therapy to reduce urine oxalate (UOx) excretion in patients with secondary hyperoxaluria. METHODS: Sixteen male and female subjects with both hyperoxaluria and a kidney stone history were enrolled in an open-label study. Subjects continued their usual diets and therapies. During a 3-day baseline period, two 24-h (24-h) urines were collected, followed by a 4-day treatment period with ALLN-177 (7,500 units/meal, 3 × day) when three 24-h urines were collected. The primary endpoint was the change in mean 24-h UOx from baseline. Safety assessments and 24-h dietary recalls were performed throughout. RESULTS: The study enrolled 5 subjects with enteric hyperoxaluria and 11 with idiopathic hyperoxaluria. ALLN-177 was well tolerated. Overall mean (SD) UOx decreased from 77.7 (55.9) at baseline to 63.7 (40.1) mg/24 h while on ALLN-177 therapy, with the mean reduction of 14 mg/24 h, (95% CI - 23.71, - 4.13). The calcium oxalate-relative urinary supersaturation ratio in the overall population decreased from a mean of 11.3 (5.7) to 8.8 (3.8) (- 2.8; 95% CI - 4.9, - 0.79). This difference was driven by oxalate reduction alone, but not any other urinary parameters. Mean daily dietary oxalate, calcium, and fluid intake recorded by frequent diet recall did not differ by study periods. CONCLUSION: ALLN-177 reduced 24-h UOx excretion, and was well tolerated.     Experimental Test of Connector Rotation during DNA Packaging into Bacteriophage ϕ29 Capsids

    The bacteriophage ϕ29 generates large forces to compact its double-stranded DNA genome into a protein capsid by means of a portal motor complex. Several mechanical models for the generation of these high forces by the motor complex predict coupling of DNA translocation to rotation of the head-tail connector dodecamer. Putative connector rotation is investigated here by combining the methods of single-molecule force spectroscopy with polarization-sensitive single-molecule fluorescence. In our experiment, we observe motor function in several packaging complexes in parallel using video microscopy of bead position in a magnetic trap. At the same time, we follow the orientation of single fluorophores attached to the portal motor connector. From our data, we can exclude connector rotation with greater than 99% probability and therefore answer a long-standing mechanistic question

    LSST: from Science Drivers to Reference Design and Anticipated Data Products

    (Abridged) We describe here the most ambitious survey currently planned in the optical, the Large Synoptic Survey Telescope (LSST). A vast array of science will be enabled by a single wide-deep-fast sky survey, and LSST will have unique survey capability in the faint time domain. The LSST design is driven by four main science themes: probing dark energy and dark matter, taking an inventory of the Solar System, exploring the transient optical sky, and mapping the Milky Way. LSST will be a wide-field ground-based system sited at Cerro Pach\'{o}n in northern Chile. The telescope will have an 8.4 m (6.5 m effective) primary mirror, a 9.6 deg2^2 field of view, and a 3.2 Gigapixel camera. The standard observing sequence will consist of pairs of 15-second exposures in a given field, with two such visits in each pointing in a given night. With these repeats, the LSST system is capable of imaging about 10,000 square degrees of sky in a single filter in three nights. The typical 5σ\sigma point-source depth in a single visit in rr will be ∌24.5\sim 24.5 (AB). The project is in the construction phase and will begin regular survey operations by 2022. The survey area will be contained within 30,000 deg2^2 with ÎŽ<+34.5∘\delta<+34.5^\circ, and will be imaged multiple times in six bands, ugrizyugrizy, covering the wavelength range 320--1050 nm. About 90\% of the observing time will be devoted to a deep-wide-fast survey mode which will uniformly observe a 18,000 deg2^2 region about 800 times (summed over all six bands) during the anticipated 10 years of operations, and yield a coadded map to r∌27.5r\sim27.5. The remaining 10\% of the observing time will be allocated to projects such as a Very Deep and Fast time domain survey.     Background: Cardiovascular disease is a major comorbidity of rheumatoid arthritis (RA) and a leading cause of death. Chronic systemic inflammation involving tumour necrosis factor alpha (TNF) could contribute to endothelial activation and atherogenesis. A number of anti-TNF therapies are in current use for the treatment of RA, including certolizumab pegol (CZP), (Cimzia Âź; UCB, Belgium). Anti-TNF therapy has been associated with reduced clinical cardiovascular disease risk and ameliorated vascular function in RA patients. However, the specific effects of TNF inhibitors on endothelial cell function are largely unknown. Our aim was to investigate the mechanisms underpinning CZP effects on TNF-activated human endothelial cells. Methods: Human aortic endothelial cells (HAoECs) were cultured in vitro and exposed to a) TNF alone, b) TNF plus CZP, or c) neither agent. Microarray analysis was used to examine the transcriptional profile of cells treated for 6 hrs and quantitative polymerase chain reaction (qPCR) analysed gene expression at 1, 3, 6 and 24 hrs. NF-ÎșB localization and IÎșB degradation were investigated using immunocytochemistry, high content analysis and western blotting. Flow cytometry was conducted to detect microparticle release from HAoECs. Results: Transcriptional profiling revealed that while TNF alone had strong effects on endothelial gene expression, TNF and CZP in combination produced a global gene expression pattern similar to untreated control. The two most highly up-regulated genes in response to TNF treatment were adhesion molecules E-selectin and VCAM-1 (q 0.2 compared to control; p > 0.05 compared to TNF alone). The NF-ÎșB pathway was confirmed as a downstream target of TNF-induced HAoEC activation, via nuclear translocation of NF-ÎșB and degradation of IÎșB, effects which were abolished by treatment with CZP. In addition, flow cytometry detected an increased production of endothelial microparticles in TNF-activated HAoECs, which was prevented by treatment with CZP. Conclusions: We have found at a cellular level that a clinically available TNF inhibitor, CZP reduces the expression of adhesion molecule expression, and prevents TNF-induced activation of the NF-ÎșB pathway. Furthermore, CZP prevents the production of microparticles by activated endothelial cells. This could be central to the prevention of inflammatory environments underlying these conditions and measurement of microparticles has potential as a novel prognostic marker for future cardiovascular events in this patient group.
