The Role of β7 Integrins in CD8 T Cell Trafficking During an Antiviral Immune Response
The requirement of β7 integrins for lymphocyte migration was examined during an ongoing immune response in vivo. Transgenic mice (OT-I) expressing an ovalbumin-specific major histocompatibility complex class Iârestricted T cell receptor for antigen were rendered deficient in expression of all β7 integrins or only the ÎąEβ7 integrin. To quantitate the relative use of β7 integrins in migration in vivo, equal numbers of OT-I and OT-I-β7â/â or OT-I-ÎąEâ/â lymph node (LN) cells were adoptively transferred to normal mice. Although OT-I-β7â/â LN cells migrated to mesenteric LN and peripheral LN as well as wild-type cells, β7 integrins were required for naive CD8 T cell and B cell migration to Peyer's patch. After infection with a recombinant virus (vesicular stomatitis virus) encoding ovalbumin, β7 integrins became critical for migration of activated CD8 T cells to the mesenteric LN and Peyer's patch. Naive CD8 T cells did not enter the lamina propria or the intestinal epithelium, and the majority of migration of activated CD8 T cells to the small and large intestinal mucosa, including the epithelium, was β7 integrinâmediated. The ÎąEβ7 integrin appeared to play no role in migration during a primary CD8 T cell immune response in vivo. Furthermore, despite dramatic upregulation of ÎąEβ7 by CD8 T cells after entry into the epithelium, long-term retention of intestinal intraepithelial lymphocytes was also ÎąEβ7 independent
Integrating spirituality as a key component of patient care
Patient care frequently focuses on physical aspects of disease management, with variable attention given to spiritual needs. And yet, patients indicate that spiritual suffering adds to distress associated with illness. Spirituality, broadly defined as that which gives meaning and purpose to a personâs life and connectedness to the significant or sacred, often becomes a central issue for patients. Growing evidence demonstrates that spirituality is important in patient care. Yet healthcare professionals (HCPs) do not always feel prepared to engage with patients about spiritual issues. In this project, HCPs attended an educational session focused on using the FICA Spiritual History Tool to integrate spirituality into patient care. Later, they incorporated the tool when caring for patients participating in the study. This research (1) explored the value of including spiritual history taking in clinical practice; (2) identified facilitators and barriers to incorporating spirituality into person-centred care; and (3) determined ways in which HCPs can effectively utilize spiritual history taking. Data were collected using focus groups and chart reviews. Findings indicate positive impacts at organizational, clinical/unit, professional/personal and patient levels when HCPs include spirituality in patient care. Recommendations are offered
Developmental Research in Space: Predicting Adult Neurobehavioral Phenotypes via Metabolomic Imaging
As human habitation and eventual colonization of space becomes an inevitable reality, there is a necessity to understand how organisms develop over the life span in the space environment. Microgravity, altered CO2, radiation and psychological stress are some of the key factors that could affect mammalian reproduction and development in space, however there is a paucity of information on this topic. Here we combine early (neonatal) in vivo spectroscopic imaging with an adult emotionality assay following a common obstetric complication (prenatal asphyxia) likely to occur during gestation in space. The neural metabolome is sensitive to alteration by degenerative changes and developmental disorders, thus we hypothesized that that early neonatal neurometabolite profiles can predict adult response to novelty. Late gestation fetal rats were exposed to moderate asphyxia by occluding the blood supply feeding one of the rats pair uterine horns for 15min. Blood supply to the opposite horn was not occluded (within-litter cesarean control). Further comparisons were made with vaginal (natural) birth controls. In one-week old neonates, we measured neurometabolites in three brain areas (i.e., striatum, prefrontal cortex, and hippocampus). Adult perinatally-asphyxiated offspring exhibited greater anxiety-like behavioral phenotypes (as measured the composite neurobehavioral assay involving open field activity, responses to novel object, quantification of fecal droppings, and resident-intruder tests of social behavior). Further, early neurometabolite profiles predicted adult responses. Non-invasive MRS screening of mammalian offspring is likely to advance ground-based space analogue studies informing mammalian reproduction in space, and achieving high-priority multigenerational research that will enable studies of the first truly space-developed mammals
Intonation and Compensation of Fretted String Instruments
In this paper we present mathematical and physical models to be used in the
analysis of the problem of intonation of musical instruments such as guitars,
mandolins and the like, i.e., we study how to improve the tuning on these
instruments. This analysis begins by designing the placement of frets on the
fingerboard according to mathematical rules and the assumption of an ideal
string, but becomes more complicated when one includes the effects of
deformation of the string and inharmonicity due to other string
characteristics. As a consequence of these factors, perfect intonation of all
the notes on the instrument can never be achieved, but complex compensation
procedures are introduced and studied to minimize the problem. To test the
validity of these compensation procedures, we have performed extensive
measurements using standard monochord sonometers and other basic acoustical
devices, confirming the correctness of our theoretical models. In particular,
these experimental activities can be easily integrated into standard acoustics
courses and labs, and can become a more advanced version of basic experiments

Biology and Impacts of Pacific Island Invasive Species: 8. Eleutherodactylus planirostris, the Greenhouse Frog (Anura: Eleutherodactylidae)
The greenhouse frog, Eleutherodactylus planirostris, is a direct-developing (i.e., no aquatic stage) frog native to Cuba and the Bahamas. It was introduced to Hawaii via nursery plants in the early 1990s and then subsequently from Hawaii to Guam in 2003. The greenhouse frog is now widespread on five Hawaiian Islands and Guam. Infestations are often overlooked due to the frogâs quiet calls, small size, and cryptic behavior, and this likely contributes to its spread. Because the greenhouse frog is an insectivore, introductions may reduce invertebrates. In Hawaii, the greenhouse frog primarily consumes ants, mites, and springtails, and obtains densities of up to 12,500 frogs ha-1. At this density, it is estimated that they can consume up to 129,000 invertebrates ha-1 night-1. They are a food source for the non-native brown tree snake in Guam and may be a food source for other non-native species. They may also compete with other insectivores for available prey. The greatest direct economic impacts of the invasions are to the nursery trade that must treat infested shipments. Although various control methods have been developed to control frogs in Hawaii, and citric acid, in particular, is effective in reducing greenhouse frogs, the frogâs inconspicuous nature often prevents populations from being identified and managed

However, because most biodiversity is invisible to EO, EO-based indicators could be misleading, which can reduce the effectiveness of nature conservation and even unintentionally decrease conservation effort. We describe an approach that combines automated recording devices, high-throughput DNA sequencing, and modern ecological modelling to extract much more of the information available in EO data. This approach is achievable now, 62 offering efficient and near-real time monitoring of management impacts on biodiversity and its functions and services
