    "Der Zweck als Seele der Handlung":Hegels Auffassung der Folgen der Handlung in den Grundlinien der Philosophie des Rechts (§§ 109 - 132)

    Die Arbeit untersucht die Hegelsche Auffassung der Handlung und der Handlungsfolgen, die im Kapitel "Moralität" der Grundlinien der Philosophie des Rechts dargestellt wird. Meine Hypothese lautet, dass die Handlungsfolgen in einem analogen Zusammenhang zum inneren Handlungszweck wie der Körper bzw. der Leib zur Seele stehen. In § 118 der Grundlinien definiert nämlich Hegel den Zweck als "Seele der Handlung". Die Analyse konzentriert sich auf die Hegelsche Auffassung des Handlungsprozesses, insbesondere auf die Aspekte der Handlungsrealisierung und die von der Handlung verursachten Folgen. Es wurde auf die Frage eingegangen, wie sich die Folgen vom subjektiven Willen des Handelnden unterscheiden und welche Faktoren hinzukommen, wenn der innere Zweck in die Objektivität übersetzt wird. Das erste Kapitel ist dem subjektiven Geist gewidmet, d.h. der Abhandlung des Begriffs der Seele im Systemteil "Anthropologie" der verschiedenen Ausgaben der Enzyklopädie der philosophischen Wissenschaften. Im zweiten und dritten Kapitel geht es um objektiven Geist. Im zweiten Kapitel wurde der Begriff des inneren Zwecks betrachtet, wie er in den Grundlinien von Hegel dargestellt ist, und das dritte Kapitel hat zum Thema die Handlungsfolgen. <br

    Efficacy in the field of two anticoccidial vaccines for broilers

    We compared two attenuated anticoccidial vaccines, administered to broilers by spray into the incubator (88,000 males and 210,100 females). Vaccine A container five species of Eimeria and vaccine B three. Zootechnical performance was similar in the two groups, with mean lesion scores no higher than 1; vaccine A caused only duodenal lesions, while vaccine B also caused typhlitis. Maximum oocyst count was 23,000/g feces at age 28 days with vaccine A and 38,000 at 21 days with vaccine B.     Seroconversion to viral haemorrhagic enteritis (HE) was studied in seven flocks of turkey breeders (17.974 birds in total), after 20 weeks of the onset of egg production. They showed no clinical signs, and mortality rate was normal. However, the infection caused a drop in egg production lasting about five weeks (-2.32 eggs laid during this period), but had no effect on hatching parameters

    The external shell of the CUORE cryostat is a large cryogen-free system designed to host the dilution refrigerator and the bolometers of the CUORE experiment in a low radioactivity environment. The three vessels that form the outer shell were produced and delivered to the Gran Sasso underground Laboratories in July 2012.     Stable Isotope Analysis of the Inca Mummy from Nevado de Chuscha (Salta, Argentina)

    This paper presents the carbon (13C/12C), nitrogen (15N/14N), oxygen (18O/16O), hydrogen (2H/1H) and sulfur (34S/32S) stable isotope values measured in the hair of a female individual from north-western Argentina. The analysis of segments of this tissue allows for the recording of the diet and migratory changes with a short time resolution. The sample is from a mummified young female individual discovered in Chuscha mount, Salta province. It was found at more than 5000 masl, in a mountain sanctuary of the Inca expansion (capacochas). The paper discusses the paleodiet and mobility patterns of this individual in the period before her death, focusing on the isotopic variations in a limited time scale. The results indicate that the individual moved from a different region to the place where she was sacrificed. Furthermore, in the last year the individual was alive, a shift in the isotopic composition of the food consumed is detected: a variation in the importance of C4 over C3 resources is evident. The results are compared with the isotopic estimations for other children and young people recovered in archaeological contexts associated with capacochas to infer variability in the geographical trajectories covered during their last months of life.Fil: Killian Galván, Violeta Anahí.     The CUORE cryostat: an infrastructure for rare event searches at millikelvin temperatures

    The CUORE experiment is the world's largest bolometric experiment. The detector consists of an array of 988 TeO2 crystals, for a total mass of 742 kg. CUORE is presently taking data at the Laboratori Nazionali del Gran Sasso, Italy, searching for the neutrinoless double beta decay of 130Te. A large custom cryogen-free cryostat allows reaching and maintaining a base temperature of about 10 mK, required for the optimal operation of the detector. This apparatus has been designed in order to achieve a low noise environment, with minimal contribution to the radioactive background for the experiment. In this paper, we present an overview of the CUORE cryostat, together with a description of all its sub-systems, focusing on the solutions identified to satisfy the stringent requirements. We briefly illustrate the various phases of the cryostat commissioning and highlight the relevant steps and milestones achieved each time. Finally, we describe the successful cooldown of CUORE

    E2F1-Mediated Upregulation of p19INK4d Determines Its Periodic Expression during Cell Cycle and Regulates Cellular Proliferation

    BACKGROUND: A central aspect of development and disease is the control of cell proliferation through regulation of the mitotic cycle. Cell cycle progression and directionality requires an appropriate balance of positive and negative regulators whose expression must fluctuate in a coordinated manner. p19INK4d, a member of the INK4 family of CDK inhibitors, has a unique feature that distinguishes it from the remaining INK4 and makes it a likely candidate for contributing to the directionality of the cell cycle. p19INK4d mRNA and protein levels accumulate periodically during the cell cycle under normal conditions, a feature reminiscent of cyclins. METHODOLOGY/PRINCIPAL FINDINGS: In this paper, we demonstrate that p19INK4d is transcriptionally regulated by E2F1 through two response elements present in the p19INK4d promoter. Ablation of this regulation reduced p19 levels and restricted its expression during the cell cycle, reflecting the contribution of a transcriptional effect of E2F1 on p19 periodicity. The induction of p19INK4d is delayed during the cell cycle compared to that of cyclin E, temporally separating the induction of these proliferative and antiproliferative target genes. Specific inhibition of the E2F1-p19INK4d pathway using triplex-forming oligonucleotides that block E2F1 binding on p19 promoter, stimulated cell proliferation and increased the fraction of cells in S phase. CONCLUSIONS/SIGNIFICANCE: The results described here support a model of normal cell cycle progression in which, following phosphorylation of pRb, free E2F induces cyclin E, among other target genes. Once cyclinE/CDK2 takes over as the cell cycle driving kinase activity, the induction of p19 mediated by E2F1 leads to inhibition of the CDK4,6-containing complexes, bringing the G1 phase to an end. This regulatory mechanism constitutes a new negative feedback loop that terminates the G1 phase proliferative signal, contributing to the proper coordination of the cell cycle and provides an additional mechanism to limit E2F activity

    SNX27, a protein involved in down syndrome, regulates GPR17 trafficking and oligodendrocyte differentiation

    The G protein-coupled receptor 17 (GPR17) plays crucial roles in myelination. It is highly expressed during transition of oligodendrocyte progenitor cells to immature oligodendrocytes, but, after this stage, it must be down-regulated to allow generation of mature myelinating cells. After endocytosis, GPR17 is sorted into lysosomes for degradation or recycled to the plasma membrane. Balance between degradation and recycling is important for modulation of receptor levels at the cell surface and thus for the silencing/activation of GPR17-signaling pathways that, in turn, affect oligodendrocyte differentiation. The molecular mechanisms at the basis of these processes are still partially unknown and their characterization will allow a better understanding of myelination and provide cues to interpret the consequences of GPR17 dysfunction in diseases. Here, we demonstrate that the endocytic trafficking of GPR17 is mediated by the interaction of a type I PDZ-binding motif located at the C-terminus of the receptor and SNX27, a recently identified protein of the endosome-associated retromer complex and whose functions in oligodendrocytes have never been studied. SNX27 knock-down significantly reduces GPR17 plasma membrane recycling in differentiating oligodendrocytes while accelerating cells' terminal maturation. Interestingly, trisomy-linked down-regulation of SNX27 expression in the brain of Ts65Dn mice, a model of Down syndrome, correlates with a decrease in GPR17+ cells and an increase in mature oligodendrocytes, which, however, fail in reaching full maturation, eventually leading to hypomyelination. Our data demonstrate that SNX27 modulates GPR17 plasma membrane recycling and stability, and that disruption of the SNX27/GPR17 interaction might contribute to pathological oligodendrocyte differentiation defects.
