    Chromatographic profiles of blood plasma free oligosaccharides in patients with cardiovascular disease

    Free oligosaccharides (FOS) are unbound structural analogs of glycans of glycoconjugates. There are several sources of them inside the cell: 1) multistep pathways of N-glycosylation, 2) the cell quality control and endoplastic reticulum-associated degradation of mis-glycosylated and/or misfolded glycoproteins, 3) lysosomal degradation of mature glycoconjugates. Some of these FOS are the earliest indicators of potential glycosylation alterations that would be revealed in the course of the cell quality control and the endoplastic reticulum-associated degradation. Ischemia and hypertension cause stress of intracellular organelles leading to disruption of their functions. The main objective of the work was the characterization of free oligosaccharides (FOS) in plasma obtained from patients with cardiovascular diseases compared to those from healthy subjects to evaluate the potential of these compounds for diagnostics. Chromatographic profiles of FOS composed of 4–12 monosaccharides were obtained and analyzed for quantitative and qualitative differences between the samples. After plasma deproteinization and FOS purification the oligosaccharides were labelled with anthranilic acid (2-AA), separated into the neutral and charged with QAE Sephadex (Q25-120) chromatography and analysed using high-performance liquid chromatography (HPLC). Glucose unit values were determined following comparison with a 2-AA-labelled glucose oligomer ladder derived from a partial hydrolysate of dextran as an external standard. The data were collected and processed using Empower software. The charged FOS were digested with the sialidase from Arthrobacter ureafaciens. 2-AA – labelled free oligosaccharides from transferrin were used as an external standard for the structure decoding. The profiles obtained were compared with intracellular free oligosaccharides of known structures and with the glycan structures and their descriptions in the databases GlycoBase and EUROCarbDB. These approaches allowed predicting a range of glycan structures for each of the main peaks of HPLC profiles of plasma free oligosaccharides and managing ways for their future experimental analysis. In the case of cardiovascular disorders, HPLC profiles of FOS revealed a changing pattern of heterogeneity, depending on the severity of the disease. Three main enlarged glycan species in the netral fraction and one peak in the charged fraction distinguished the FOS of the patients from those of the healthy volunteers. It has been revealed that the neutral marker peaks were represented by polimannose glycans with 5–7 mannose residues and 1–2 residues of N-acetylglucosamine, and one of the major peaks of the charged fraction – by two-antennary complex N-glycan with two sialic acid residues. The study of free oligosaccharides of blood plasma is a new field of glycobiology allowing an evaluation of an organism state at the level of the cell organelle functional status and openning up broad prospects for finding early diagnostic and prognostic markers of cardiac insufficiency

    The first WASP public data release

    The WASP (wide angle search for planets) project is an exoplanet transit survey that has been automatically taking wide field images since 2004. Two instruments, one in La Palma and the other in South Africa, continually monitor the night sky, building up light curves of millions of unique objects. These light curves are used to search for the characteristics of exoplanetary transits. This first public data release (DR1) of the WASP archive makes available all the light curve data and images from 2004 up to 2008 in both the Northern and Southern hemispheres. A web interface () to the data allows easy access over the Internet. The data set contains 3 631 972 raw images and 17 970 937 light curves. In total the light curves have 119 930 299 362 data points available between them

    The intriguing nature of the high energy gamma ray source XSSJ12270-4859

    The nature of the hard X-ray source XSSJ12270-4859 is still unclear though it was claimed to be a magnetic Cataclysmic Variable. We here present a broad-band X-ray and gamma ray study based on a recent XMM-Newton observation and archival INTEGRAL and RXTE data. From the Fermi/LAT 1-year point source catalogue, we tentatively associate XSSJ12270-4859 with 1FGLJ1227.9-4852, a source of high energy gamma rays with emission up to 10GeV. We complement the study with UV photometry from XMM-Newton and ground-based optical and near-IR photometry. The X-ray emission is highly variable showing flares and intensity dips. The X-ray flares consist of flare-dip pairs. Flares are also detected in the UV range but not the dips. Aperiodic dipping behaviour is also observed during X-ray quiescence but not in the UV. The 0.2-100keV spectrum is featureless and described by a power law model with Gamma=1.7. The 100MeV-10GeV spectrum is instead represented by a power law index of 2.45. The luminosity ratio between 0.1-100GeV and 0.2--100keV is ~0.8, hence the GeV emission is a significant component of the total energy output. Furthermore, the X-ray spectrum does not greatly change during flares, quiescence and the dips seen in quiescence but it hardens during the post-flare dips. Optical photometry reveals a period of 4.32hr likely related to the binary orbit. Near-IR, possibly ellipsoidal, variations are detected. Large amplitude variability on shorter (tens mins) timescales are found to be non-periodic. The observed variability at all wavelengths and the spectral characteristics strongly favour a low-mass atypical low-luminosity X-ray binary and are against a Cataclysmic Variable nature.     Increasing Dominance - the Role of Advertising, Pricing and Product Design

    Despite the empirical relevance of advertising strategies in concentrated markets, the economics literature is largely silent on the effect of persuasive advertising strategies on pricing, market structure and increasing (or decreasing) dominance. In a simple model of persuasive advertising and pricing with differentiated goods, we analyze the interdependencies between ex-ante asymmetries in consumer appeal, advertising and prices. Products with larger initial appeal to consumers will be advertised more heavily but priced at a higher level - that is, advertising and price discounts are strategic substitutes for products with asymmetric initial appeal. We find that the escalating effect of advertising dominates the moderating effect of pricing so that post-competition market shares are more asymmetric than pre-competition differences in consumer appeal. We further find that collusive advertising (but competitive pricing) generates the same market outcomes, and that network effects lead to even more extreme market outcomes, both directly and via the effect on advertising

    Measurement and Computation of Movement of Bromide Ions and Carbofuran in Ridged Humic-Sandy Soil

    Water flow and pesticide transport in the soil of fields with ridges and furrows may be more complex than in the soil of more level fields. Prior to crop emergence, the tracer bromide ion and the insecticide carbofuran were sprayed on the humic-sandy soil of a potato field with ridges and furrows. Rainfall was supplemented by sprinkler irrigation. The distribution of the substances in the soil profile of the ridges and furrows was measured on three dates in the potato growing season. Separate ridge and furrow systems were simulated by using the pesticide emission assessment at regional and local scales (PEARL) model for pesticide behavior in soil–plant systems. The substances travelled deeper in the furrow soil than in the ridge soil, because of runoff from the ridges to the furrows. At 19 days after application, the peak of the bromide distribution was measured to be in the 0.1–0.2 m layer of the ridges, while it was in the 0.3–0.5 m layer of the furrows. After 65 days, the peak of the carbofuran distribution in the ridge soil was still in the 0.1 m top layer, while the pesticide was rather evenly distributed in the top 0.6 m of the furrow soil. The wide ranges in concentration measured with depth showed that preferential water flow and substance transport occurred in the sandy soil. Part of the bromide ion distribution was measured to move faster in soil than the computed wave. The runoff of water and pesticide from the ridges to the furrows, and the thinner root zone in the furrows, are expected to increase the risk of leaching to groundwater in ridged fields, in comparison with more level fields

    Is there evidence for field restructuring or decay in accreting magnetic white dwarfs?

    The evolution of the magnetic field of an accreting magnetic white dwarf with an initially dipolar field at the surface has been studied for non-spherical accretion under simplifying assumptions. Accretion on to the polar regions tends to advect the field toward the stellar equator which is then buried. This tendency is countered by Ohmic diffusion and magneto-hydrodynamic instabilities. It is argued that if matter is accreted at a rate of M˙crit1016\dot{M}_{\rm crit} \sim 10^{16} g s1^{-1} and the total mass accreted exceeds a critical value \Delta M_{\rm crit} \sim 0.1-0.2\ms, the field may be expected to be restructured, and the polar field to be reduced} reaching a minimum value of 103\sim 10^3 G (the "bottom field") independently of the initial field strength. Below this critical accretion rate, the field diffuses faster than it can be advected, and accretion has little effect on field strength and structure.Comment: MNRAS 200

    A meta-analytic review of stand-alone interventions to improve body image

    Objective Numerous stand-alone interventions to improve body image have been developed. The present review used meta-analysis to estimate the effectiveness of such interventions, and to identify the specific change techniques that lead to improvement in body image. Methods The inclusion criteria were that (a) the intervention was stand-alone (i.e., solely focused on improving body image), (b) a control group was used, (c) participants were randomly assigned to conditions, and (d) at least one pretest and one posttest measure of body image was taken. Effect sizes were meta-analysed and moderator analyses were conducted. A taxonomy of 48 change techniques used in interventions targeted at body image was developed; all interventions were coded using this taxonomy. Results The literature search identified 62 tests of interventions (N = 3,846). Interventions produced a small-to-medium improvement in body image (d+ = 0.38), a small-to-medium reduction in beauty ideal internalisation (d+ = -0.37), and a large reduction in social comparison tendencies (d+ = -0.72). However, the effect size for body image was inflated by bias both within and across studies, and was reliable but of small magnitude once corrections for bias were applied. Effect sizes for the other outcomes were no longer reliable once corrections for bias were applied. Several features of the sample, intervention, and methodology moderated intervention effects. Twelve change techniques were associated with improvements in body image, and three techniques were contra-indicated. Conclusions The findings show that interventions engender only small improvements in body image, and underline the need for large-scale, high-quality trials in this area. The review identifies effective techniques that could be deployed in future interventions

    WASP-38b: a transiting exoplanet in an eccentric, 6.87d period orbit

    Aims: We report the discovery of WASP-38b, a long period transiting planet in an eccentric 6.871815 day orbit. The transit epoch is 2 455 335.92050 ± 0.00074 (HJD) and the transit duration is 4.663 h. Methods: WASP-38b's discovery was enabled due to an upgrade to the SuperWASP-North cameras. We performed a spectral analysis of the host star HD 146389/BD+10 2980 that yielded Teff = 6150 ± 80 K, log g = 4.3 ± 0.1, v sin i = 8.6 ± 0.4 km s-1, M* = 1.16 ± 0.04 Mʘand R* = 1.33 ± 0.03 Rʘ, consistent with a dwarf of spectral type F8. Assuming a main-sequence mass-radius relation for the star, we fitted simultaneously the radial velocity variations and the transit light curves to estimate the orbital and planetary parameters. Results: The planet has a mass of 2.69 ± 0.06 MJup and a radius of 1.09 ± 0.03 RJup giving a density, ρp = 2.1 ± 0.1 ρJ. The high precision of the eccentricity e = 0.0314 ± 0.0044 is due to the relative transit timing from the light curves and the RV shape. The planet equilibrium temperature is estimated at 1292 ± 33 K. WASP-38b is the longest period planet found by SuperWASP-North and with a bright host star (V = 9.4 mag), is a good candidate for followup atmospheric studies
