Mountain farmland protection and fire-smart management jointly reduce fire hazard and enhance biodiversity and carbon sequestration
The environmental and socio-economic impacts of wildfires are foreseen to increase across southern Europe over the next decades regardless of increasing resources allocated for fire suppression. This study aims to identify fire-smart management strategies that promote wildfire hazard reduction, climate regulation ecosystem service and biodiversity conservation. Here we simulate fire-landscape dynamics, carbon sequestration and species distribution (116 vertebrates) in the Transboundary Biosphere Reserve Gerês-Xurés (NW Iberia). We envisage 11 scenarios resulting from different management strategies following four storylines: Business-as-usual (BAU), expansion of High Nature Value farmlands (HNVf), Fire-Smart forest management, and HNVf plus Fire-Smart. Fire-landscape simulations reveal an increase of up to 25% of annual burned area. HNVf areas may counterbalance this increasing fire impact, especially when combined with fire-smart strategies (reductions of up to 50% between 2031 and 2050). The Fire-Smart and BAU scenarios attain the highest estimates for total carbon sequestered. A decrease in habitat suitability (around 18%) since 1990 is predicted for species of conservation concern under the BAU scenario, while HNVf would support the best outcomes in terms of conservation. Our study highlights the benefits of integrating fire hazard control, ecosystem service supply and biodiversity conservation to inform better decision-making in mountain landscapes of Southern Europe.This research work was funded by national funds through the FCT –

Using fire to enhance rewilding when agricultural policies fail
Rewilding has been proposed as an opportunity for biodiversity conservation in abandoned landscapes. However,
rewilding is challenged by the increasing fire risk associated with more flammable landscapes, and the
loss of open-habitat specialist species. Contrastingly, supporting High Nature Value farmlands (HNVf) has been
also highlighted as a valuable option, but the effective implementation of agricultural policies often fails leading
to uncertain scenarios wherein the effects of wildfire management remain largely unexplored. Herein, we simulated
fire-landscape dynamics to evaluate howfire suppression scenarios affect fire regime and biodiversity (102
species of vertebrates) under rewilding and HNVf policies in the future (2050), in a transnational biosphere reserve
(Gerês-Xurés Mountains, Portugal-Spain). Rewilding and HNVf scenarios were modulated by three different
levels of fire suppression effectiveness. Then, we quantified scenario effects on fire regime (burned and
suppressed areas) and biodiversity (habitat suitability change for 2050). Simulations confirm HNVf as a longterm
opportunity for fire suppression (up to 30,000 ha of additional suppressed areas between 2031 and 2050 in comparison to rewilding scenario) and for conservation (benefiting around 60% of species). Rewilding benefits
some species (20%), including critically endangered, vulnerable and endemic taxa, while several species (33%)
also profit from open habitats created by fire. Although HNVf remains the best scenario, rewilding reinforced
by low fire suppression management may provide a nature-based solution when societal support through

Predicting global habitat suitability for stony corals on seamounts
Aim Globally, species distribution patterns in the deep sea are poorly resolved, with spatial coverage being sparse for most taxa and true absence data missing. Increasing human impacts on deep-sea ecosystems mean that reaching a better understanding of such patterns is becoming more urgent. Cold-water stony corals (Order Scleractinia) form structurally complex habitats (dense thickets or reefs) that can support a diversity of other associated fauna. Despite their widely accepted ecological importance, records of scleractinian corals on seamounts are patchy and simply not available for most of the global ocean. The objective of this paper is to model the global distribution of suitable habitat for stony corals on seamounts. Location Seamounts worldwide. Methods We compiled a database containing all accessible records of scleractinian corals on seamounts. Two modelling approaches developed for presence-only data were used to predict global habitat suitability for seamount scleractinians: maximum entropy modelling (Maxent) and environmental niche factor analysis (ENFA). We generated habitat-suitability maps and used a cross-validation process with a threshold-independent metric to evaluate the performance of the models. Results Both models performed well in cross-validation, although the Maxent method consistently outperformed ENFA. Highly suitable habitat for seamount stony corals was predicted to occur at most modelled depths in the North Atlantic, and in a circumglobal strip in the Southern Hemisphere between 20° and 50° S and shallower than around 1500 m. Seamount summits in most other regions appeared much less likely to provide suitable habitat, except for small near-surface patches. The patterns of habitat suitability largely reflect current biogeographical knowledge. Environmental variables positively associated with high predicted habitat suitability included the aragonite saturation state, and oxygen saturation and concentration. By contrast, low levels of dissolved inorganic carbon, nitrate, phosphate and silicate were associated with high predicted suitability. High correlation among variables made assessing individual drivers difficult. Main conclusions Our models predict environmental conditions likely to play a role in determining large-scale scleractinian coral distributions on seamounts, and provide a baseline scenario on a global scale. These results present a first-order hypothesis that can be tested by further sampling. Given the high vulnerability of cold-water corals to human impacts, such predictions are crucial tools in developing worldwide conservation and management strategies for seamount ecosystems. © 2009 Blackwell Publishing Ltd

Extracellular Vesicles in the Fungi Kingdom
Extracellular vesicles (EVs) are membranous, rounded vesicles released by prokaryotic and eukaryotic cells in their normal and pathophysiological states. These vesicles form a network of intercellular communication as they can transfer cell- and function-specific information (lipids, proteins and nucleic acids) to different cells and thus alter their function. Fungi are not an exception; they also release EVs to the extracellular space. The vesicles can also be retained in the periplasm as periplasmic vesicles (PVs) and the cell wall. Such fungal vesicles play various specific roles in the lives of these organisms. They are involved in creating wall architecture and maintaining its integrity, supporting cell isolation and defence against the environment. In the case of pathogenic strains, they might take part in the interactions with the host and affect the infection outcomes. The economic importance of fungi in manufacturing high-quality nutritional and pharmaceutical products and in remediation is considerable. The analysis of fungal EVs opens new horizons for diagnosing fungal infections and developing vaccines against mycoses and novel applications of nanotherapy and sensors in industrial processes
Myosin Vb as a tumor suppressor gene in intestinal cancer
Colorectal cancer causes >900,000 deaths every year and a deeper understanding of the molecular mechanisms underlying this disease will contribute to improve its clinical management and survival. Myosin Vb (MYO5B) regulates intracellular vesicle trafficking, and inactivation of this myosin disrupts the polarization and differentiation of intestinal epithelial cells causing microvillous inclusion disease (MVID), a rare congenital disorder characterized by intractable life-threatening diarrhea. Here, we show that the loss Myosin Vb interfered with the differentiation/polarization of colorectal cancer cells. Although modulation of Myosin Vb expression did not affect the proliferation of colon cancer cells, MYO5B inactivation increased their migration, invasion, and metastatic potential. Moreover, Myo5b inactivation in an intestine-specific knockout mouse model caused a >15-fold increase in the number of azoxymethane-initiated small intestinal tumors. Consistently, reduced expression of Myosin Vb in a cohort of 155 primary colorectal tumors was associated with shorter patient survival. In conclusion, we show here that loss of Myosin Vb reduces polarization/differentiation of colon cancer cells while enhancing their metastatic potential, demonstrating a tumor suppressor function for this myosin. Moreover, reduced expression of Myosin Vb in primary tumors identifies a subset of poor prognosis colorectal cancer patients that could benefit from more aggressive therapeutic regimens
