    Water Defluoridation: Nanofiltration vs Membrane Distillation

    Nowadays, fluoride contamination of drinking water is a major problem for various countries, because high concentrations of fluoride pose a risk of dental and skeletal fluorosis. Over past years, membrane nanofiltration (NF) has been proposed as convenient defluoridation technology. However, NF cannot be applied to water systems with high fluoride concentration, and the disposal of the membrane concentrate remains an issue. In this work, we compared a commercial polyester NF membrane and a polypropylene hollow-fiber membrane distillation (MD) module for their ability to remove fluoride ions from water in the presence of hardness ions and organic fouling agents. The NF membrane can offer more than 10 times higher water productivity than MD, under realistic gradients of temperature and pressure, respectively. Despite that, after reaching a concentration factor of about 3, fouling and scaling caused the flux to drop to about 80% with respect to its initial value. Moreover, F- retention decreased from 90% to below 80%, thus providing a permeate of scarce quality. MD was operated in the direct-contact mode on a polypropylene hollow-fiber membrane, which was charged with a hot feed flow (average T = 58 °C) on one side and a cooled (20 °C) permeate flow of distilled water on the other side. The concentration of fluoride ions in the permeate was always below the detection limit of our electrode (0.2 ppm), regardless of the fluoride concentration in the feed. Moreover, the MD module showed higher resistance to fouling and scaling than NF, and CaF2 crystals were recovered from the MD concentrate after cooling. These results suggest that the synergic combination of the two techniques might be beneficial for the purification of fluoride-contaminated water systems: MD can be used to further concentrate the NF retentate, thus producing high-purity water and recovering CaF2 crystals.Fil: Moran Ayala, Lucia Isabel.     Next-Generation EU DataGrid Data Management Services

    We describe the architecture and initial implementation of the next-generation of Grid Data Management Middleware in the EU DataGrid (EDG) project. The new architecture stems out of our experience and the users requirements gathered during the two years of running our initial set of Grid Data Management Services. All of our new services are based on the Web Service technology paradigm, very much in line with the emerging Open Grid Services Architecture (OGSA). We have modularized our components and invested a great amount of effort towards a secure, extensible and robust service, starting from the design but also using a streamlined build and testing framework. Our service components are: Replica Location Service, Replica Metadata Service, Replica Optimization Service, Replica Subscription and high-level replica management.     Treatment of Marburg and Ebola hemorrhagic fevers: A strategy for testing new drugs and vaccines under outbreak conditions.

    The filoviruses, Marburg and Ebola, have the dubious distinction of being associated with some of the highest case-fatality rates of any known infectious disease-approaching 90% in many outbreaks. In recent years, laboratory research on the filoviruses has produced treatments and vaccines that are effective in laboratory animals and that could potentially drastically reduce case-fatality rates and curtail outbreaks in humans. However, there are significant challenges in clinical testing of these products and eventual delivery to populations in need. Most cases of filovirus infection are recognized only in the setting of large outbreaks, often in the most remote and resource-poor areas of sub-Saharan Africa, with little infrastructure and few personnel experienced in clinical research. Significant political, legal, and socio-cultural barriers also exist. Here, we review the present research priorities and environment for field study of the filovirus hemorrhagic fevers and outline a strategy for future prospective clinical research on treatment and vaccine prevention

    The landscape of expression and alternative splicing variation across human traits

    Understanding the consequences of individual transcriptome variation is fundamental to deciphering human biology and disease. We implement a statistical framework to quantify the contributions of 21 individual traits as drivers of gene expression and alternative splicing variation across 46 human tissues and 781 individuals from the Genotype-Tissue Expression project. We demonstrate that ancestry, sex, age, and BMI make additive and tissue-specific contributions to expression variability, whereas interactions are rare. Variation in splicing is dominated by ancestry and is under genetic control in most tissues, with ribosomal proteins showing a strong enrichment of tissue-shared splicing events. Our analyses reveal a systemic contribution of types 1 and 2 diabetes to tissue transcriptome variation with the strongest signal in the nerve, where histopathology image analysis identifies novel genes related to diabetic neuropathy. Our multi-tissue and multi-trait approach provides an extensive characterization of the main drivers of human transcriptome variation in health and disease.This study was funded by the HumTranscriptom project with reference PID2019-107937GA-I00.     Integrated management of ash from industrial and domestic combustion : a new sustainable approach for reducing greenhouse gas emissions from energy conversion

    This work supports, for the first time, the integrated management of waste materials arising from industrial processes (fly ash from municipal solid waste incineration and coal fly ash), agriculture (rice husk ash), and domestic activities (ash from wood biomass burning in domestic stoves). The main novelty of the paper is the reuse of wood pellet ash, an underestimated environmental problem, by the application of a new technology (COSMOS-RICE) that already involves the reuse of fly ashes from industrial and agricultural origins. The reaction mechanism involves carbonation: this occurs at room temperature and promotes permanent carbon dioxide sequestration. The obtained samples were characterized using XRD and TGA (coupled with mass spectroscopy). This allowed quantification of the mass loss attributed to different calcium carbonate phases. In particular, samples stabilized using wood pellet ash show a weight loss, attributed to the decomposition of carbonates greater than 20%. In view of these results, it is possible to conclude that there are several environmental benefits from wood pellet ash reuse in this way.     Agriculture and ritual in the 1000 BP human landscape of the Quebrada de Humahuaca headwaters: Casas Grandes site (Jujuy, Argentina)

    Este trabajo presenta los resultados de los estudios arqueológicos llevados a cabo en el sitio Casas Grandes, realizados dentro de un proyecto de investigación regional que abarca las nacientes de la Quebrada de Humahuaca en su intersección con la Puna, ecotono y umbral geo-ambiental que ocurre en este sector de la Provincia de Jujuy, Argentina. Se expone aquí una síntesis del marco general del proyecto, las escalas de análisis (espacial y temporal) así como los lineamientos teórico/metodológicos del mismo. Luego se presentan las características del sitio, su geomorfología, sus sectores y la evidencia arqueológica encontrada en cada uno de ellos. A su vez, se describen los distintos tipos hallazgos realizados y los resultados de sus respectivos análisis, focalizando en el sector bajo del sitio, donde la evidencia hallada aporta excepcional información sobre prácticas económicas y rituales realizadas en torno a los 1000 años antes del presente. En base a estos estudios y a los fechados radiocarbónicos obtenidos se presenta una discusión general sobre la articulación de este sitio con otros estudiados dentro del mismo proyecto, así como su importancia en el contexto de la arqueología de Quebrada y Puna jujeña, a partir de su comparación e integración con la evidencia proveniente de otros sitios con ocupaciones humanas contemporáneas localizados en estos dos geo-ambientes.This paper presents the archeological investigations at the Casas Grandes site, which were carried out as part of a regional research project that encompasses the headwaters of the Quebrada de Humahuaca, where it intersects the Puna. This is an ecotone and environmental threshold that occurs in this part of Jujuy Province, Argentina. It is presented here a synthesis of the project´s general framework, the scales of analysis (spatial and temporal) and the main points of the theoretical and methodological approaches employed. A discussion about the characteristics of the site, its geomorphology, the sectors into which it is divided, and the archaeological evidence recovered is also presented. The different types of findings and their analyses are discussed, focusing on the site´s lower sector, where the evidence recovered provides valuable information about economic and ritual practices from around 1000 BP. On the basis of these studies and the radiocarbon dates obtained, a general discussion about the articulation of this site and others studied within the same project is presented. The relevance of this site within the context of the archaeology of the Quebrada de Humahuaca and the Jujuy Puna is assessed by comparing and integrating the site´s evidence with that from contemporary sites in those two geo-environmental zones.Fil: Hernandez Llosas, Maria Isabel.     Inflammatory Transcriptome Profiling of Human Monocytes Exposed Acutely to Cigarette Smoke

    <div><h3>Background</h3><p>Cigarette smoking is responsible for 5 million deaths worldwide each year, and is a major risk factor for cardiovascular and lung diseases. Cigarette smoke contains a complex mixture of over 4000 chemicals containing 10<sup>15</sup> free radicals. Studies show smoke is perceived by cells as an inflammatory and xenobiotic stimulus, which activates an immune response. The specific cellular mechanisms driving cigarette smoke-induced inflammation and disease are not fully understood, although the innate immune system is involved in the pathology of smoking related diseases.</p> <h3>Methodology/Principle findings</h3><p>To address the impact of smoke as an inflammagen on the innate immune system, THP-1 cells and Human PBMCs were stimulated with 3 and 10% (v/v) cigarette smoke extract (CSE) for 8 and 24 hours. Total RNA was extracted and the transcriptome analysed using Illumina BeadChip arrays. In THP-1 cells, 10% CSE resulted in 80 genes being upregulated and 37 downregulated by ≥1.5 fold after 8 hours. In PBMCs stimulated with 10% CSE for 8 hours, 199 genes were upregulated and 206 genes downregulated by ≥1.5 fold. After 24 hours, the number of genes activated and repressed by ≥1.5 fold had risen to 311 and 306 respectively. The major pathways that were altered are associated with cell survival, such as inducible antioxidants, protein chaperone and folding proteins, and the ubiquitin/proteosome pathway.</p> <h3>Conclusions</h3><p>Our results suggest that cigarette smoke causes inflammation and has detrimental effects on the metabolism and function of innate immune cells. In addition, THP-1 cells provide a genetically stable alternative to primary cells for the study of the effects of cigarette smoke on human monocytes.</p> </div
