    Perturbations of embedded eigenvalues for the planar bilaplacian

    Operators on unbounded domains may acquire eigenvalues that are embedded in the essential spectrum. Determining the fate of these embedded eigenvalues under small perturbations of the underlying operator is a challenging task, and the persistence properties of such eigenvalues is linked intimately to the multiplicity of the essential spectrum. In this paper, we consider the planar bilaplacian with potential and show that the set of potentials for which an embedded eigenvalue persists is locally an infinite-dimensional manifold with infinite codimension in an appropriate space of potentials

    Domain Adapting Deep Reinforcement Learning for Real-world Speech Emotion Recognition

    Computers can understand and then engage with people in an emotionally intelligent way thanks to speech-emotion recognition (SER). However, the performance of SER in cross-corpus and real-world live data feed scenarios can be significantly improved. The inability to adapt an existing model to a new domain is one of the shortcomings of SER methods. To address this challenge, researchers have developed domain adaptation techniques that transfer knowledge learnt by a model across the domain. Although existing domain adaptation techniques have improved performances across domains, they can be improved to adapt to a real-world live data feed situation where a model can self-tune while deployed. In this paper, we present a deep reinforcement learning-based strategy (RL-DA) for adapting a pre-trained model to a real-world live data feed setting while interacting with the environment and collecting continual feedback. RL-DA is evaluated on SER tasks, including cross-corpus and cross-language domain adaption schema. Evaluation results show that in a live data feed setting, RL-DA outperforms a baseline strategy by 11% and 14% in cross-corpus and cross-language scenarios, respectively

    University Colleges can help smaller land surveying companies in introducing new data acquisition techniques

    The ever developing instruments, techniques and software are at the same time a blessing and a curse for the contemporary land surveyor. Laser scanning is one of these new techniques and although its versatility, speed and accuracy are praised in articles and on exhibitions, a lot of Belgian land surveyors are somewhat reluctant to introduce laser scanning in their own company. There is still a lot of uncertainty in regard to the efficiency of this data acquisition method and especially the processing of the data and the production of deliverables that meet the client's needs and expectations. University Colleges can contribute to stimulate the smaller land surveying companies to invest in innovation and thus maintain their competitiveness and consolidate their position in an open European market. This article describes how researchers at university colleges translate their knowledge and experience in such a way that they can support smaller companies, which do not have the resources to do their own research, in making an educated decision concerning the introduction of laser scanning in their company

    Elimination of the Warburg effect in Chinese hamster ovary (CHO) cells improves cell phenotype as a protein production platform

    Lactate is a common metabolite and is central to many important processes. One of its more prominent roles is in the Warburg effect, in which cancer cells exhibit high rates of glycolytic flux followed by secretion of lactate, even in the presence of oxygen. This fermentation of pyruvate to lactate via lactate dehydrogenase (Ldh) accompanies increased proliferation of cancer cells and several other types of rapidly proliferating cell types in immune cell activation and embryonic development. Aerobic glycolysis is also prominent in biotherapeutic protein production, where mammalian production cells often secrete high levels of lactate. The accumulation of lactate is deleterious for cell growth, viability, product formation, and quality, both directly via acidification of the media and indirectly through base addition to control culture pH. Despite a clear genetic target, efforts to eliminate lactate secretion via knockout of Ldh(s) in mammalian cells have been unsuccessful, pointing to the essentiality of Ldh mediated NAD regeneration. A wide variety of approaches have been utilized to limit lactate accumulation in culture, including knockdown or inhibition of Ldh, replacement of glucose with alternate sugars, controlled feeding strategies, and many others, however none have proven successful in eliminating the Warburg effect. We report the elimination of the Warburg effect in a CHO cell line by using CRISPR/Cas9-based engineering to simultaneously knockout enzymes responsible for lactate production and ancillary regulators. The resulting cell lines remain proliferative while consuming significantly less glucose and can be used to generate protein producing lines using standard industrial processes. In a pH-controlled fedbatch process, the Warburg null cells require minimal base addition to maintain a stable pH, allowing an extended growth phase. The knockout strategy was also successfully applied to a CHO cell line producing Rituximab, again resulting in a prolonged growth phase. Additionally, protein production was maintained, while product quality was improved with increased glycan galactosylation. Thus, CHO cells without the capacity of Warburg metabolism may be useful for engineering production cell lines with enhanced bioproduction traits

    Characterization of rare germline variants in familial multiple myeloma

    Multiple myeloma (MM) is a malignancy of plasma cells, characterized by the presence of monoclonal immunoglobulin, known as M protein1. MM is preceded by monoclonal gammopathy of undetermined significance (MGUS) which is also a precursor of immunoglobulin light chain (AL) amyloidosis. Previous studies have reported a 2- to 4-fold increased risk of MGUS or MM in first-degree relatives of MM or MGUS patients, suggesting the existence of inherited susceptibility. For many years, high-risk germline predisposing genes have been lacking for MM. However, recent sequencing efforts have proposed a few novel candidates, most notably loss-offunction (LoF) variants in the tumor suppressor gene DIS3 and in the histone demethylase gene KDM1A, and others as recently reviewed in detail in Pertesi et al. In addition to the suspected rare, high-penetrance variants, genome-wide association studies have identified over 20 common, low-penetrance variants associated with the risk of MM; these were estimated to account for about 15% of the familial MM risk

    European Respiratory Society guidelines for the management of adult bronchiectasis

    Bronchiectasis in adults is a chronic disorder associated with poor quality of life and frequent exacerbations in many patients. There have been no previous international guidelines.The European Respiratory Society guidelines for the management of adult bronchiectasis describe the appropriate investigation and treatment strategies determined by a systematic review of the literature.A multidisciplinary group representing respiratory medicine, microbiology, physiotherapy, thoracic surgery, primary care, methodology and patients considered the most relevant clinical questions (for both clinicians and patients) related to management of bronchiectasis. Nine key clinical questions were generated and a systematic review was conducted to identify published systematic reviews, randomised clinical trials and observational studies that answered these questions. We used the GRADE approach to define the quality of the evidence and the level of recommendations. The resulting guideline addresses the investigation of underlying causes of bronchiectasis, treatment of exacerbations, pathogen eradication, long term antibiotic treatment, anti-inflammatories, mucoactive drugs, bronchodilators, surgical treatment and respiratory physiotherapy.These recommendations can be used to benchmark quality of care for people with bronchiectasis across Europe and to improve outcomes

    The human secretome

    The proteins secreted by human cells (collectively referred to as the secretome) are important not only for the basic understanding of human biology but also for the identification of potential targets for future diagnostics and therapies. Here, we present a comprehensive analysis of proteins predicted to be secreted in human cells, which provides information about their final localization in the human body, including the proteins actively secreted to peripheral blood. The analysis suggests that a large number of the proteins of the secretome are not secreted out of the cell, but instead are retained intracellularly, whereas another large group of proteins were identified that are predicted to be retained locally at the tissue of expression and not secreted into the blood. Proteins detected in the human blood by mass spectrometry-based proteomics and antibody-based immuno-assays are also presented with estimates of their concentrations in the blood. The results are presented in an updated version 19 of the Human Protein Atlas in which each gene encoding a secretome protein is annotated to provide an open-access knowledge resource of the human secretome, including body-wide expression data, spatial localization data down to the single-cell and subcellular levels, and data about the presence of proteins that are detectable in the blood

Bryophytes respond to environmental changes at the species level, but because bryophyte species are relatively difficult to identify, they are often lumped into one functional group. Consequently, bryophyte function remains poorly resolved. Here, we explore how higher resolution of bryophyte functional diversity can be encouraged and implemented in tundra ecological studies. We briefly review previous bryophyte functional classifications and the roles of bryophytes in tundra ecosystems and their susceptibility to environmental change. Based on shoot morphology and colony organization, we then propose twelve easily distinguishable bryophyte functional groups. To illustrate how bryophyte functional groups can help elucidate variation in bryophyte effects and responses, we compiled existing data on water holding capacity, a key bryophyte trait. Although plant functional groups can mask potentially high interspecific and intraspecific variability, we found better separation of bryophyte functional group means compared with previous grouping systems regarding water holding capacity. This suggests that our bryophyte functional groups truly represent variation in the functional roles of bryophytes in tundra ecosystems. Lastly, we provide recommendations to improve the monitoring of bryophyte community changes in tundra study sites.Peer reviewe
