Le régime entre santé et esthétique ? Significations, parcours et mise en oeuvre du régime alimentaire
[paper in French] This paper analyses the meanings, trajectory and perception of nutritional recommendations concerning weight control and dieting, through 85 semi-directive interviews. The article underlines three oppositions concerning the meanings of “diet”: firstly, dieting is closely linked to health (hygiene of life) in the upper strata, whereas in the lower strata health does not often constitute an aim; secondly, dieting is perceived in a preventive perspective and in a broad meaning in the upper strata, whereas in the lower strata it is taken in a curative and narrow meaning (targeting treatment on a short time); thirdly, dieting may constitutes the basis of an experimented knowledge concerning overweight prevention, whereas dieting can remain an applied prescription, when the obsession of losing weight prevents obese people from constituting one’s personal knowledge. Sources of nutritional recommendations are numerous, coming from the medical field and dieting programs, in a context of a strong influence of media. The paper underlines also the importance of opinions leaders. In consequence, social belonging and intensity of social links constitute two main factors helping women to follow a diet, whereas loneliness and breaks in careers are often at the origin of a significant disinvestment in body care.food, diet, nutritional recommendations, slimness, health
Estudio de la comprensión del concepto “constructivismo”. El caso de la formación inicial de docentes de ciencias en la universidad
Some characteristics of the understanding that, on the concept of “constructivism”, obtains a set of students of the sciences teaching carriers of the University, in different stages from their training process appear. The performances were interpreted that show to the students when solving different types from activities, raised from the dimensions of the understanding. The work identifies, also, that the didactic strategy used –IDP- it turns out suitable to promote the understanding of concepts associated to the pedagogical-didactic field. The results allow to visualize differences in the profits of the students agreed to the advance in the Race at the reflection level that considers from didactic strategy IDP and of the possibility of recovering the previous knowledge in the analysis problems of the educational practice. As much the instruments designed specifically for this study like the associated theoretical indicators to such, give account of a comprehensive process on the subject.Se presentan algunas características de la comprensión alcanzada por alumnos del Profesorado de Ciencias de la Universidad, sobre el concepto de “constructivismo”, en diferentes años de la carrera. Se interpretaron los desempeños de los estudiantes al resolver diferentes actividades generadas a partir de la estrategia didáctica de Indagación Dialógica Problematizadora. Se analizan los desempeños logrados, resultando dicha estrategia adecuada para promover la comprensión de conceptos asociados al campo pedagógico-didáctico. También los resultados permiten visualizar diferencias en los logros de los estudiantes a medida que avanzan por la carrera. Tanto los instrumentos diseñados específicamente para este estudio como los indicadores teóricos asociados a los mismos, permiten dar cuenta de un proceso comprensivo sobre el tema “constructivismo” en la etapa de formación inicial del profesorado de Ciencias
Levothyroxine Replacement Improves Oxidative Status in Primary Hypothyroidism
Objective: Although hypothyroidism has been linked to oxidative stress, data regarding the relationship between thyroid hormone levels and oxidative stress is still inconsistent. This study was designed to evaluate the effect of levothyroxine replacement on oxidative stress in women with primary hypothyroidism.Design: A total of 25 female patients with primary hypothyroidism were included. Oxidative stress markers were measured before and after levothyroxine replacement treatment in all patients.Methods: Oxidative stress was evaluated through the measurement of oxidants (thiobarbituric acid reactive substances [TBARS] and nitrite/nitrate levels), and antioxidants (superoxide dismutase and catalase activity).Results: Antioxidant catalase activity (63.77 ± 23.8 vs. 50.12 ±12.75 atv/min; p = 0.03) was significantly increased and the levels of TBARS (3.02 ± 0.86 vs. 3.55 ± 0.87 μM; p = 0.03) were significantly decreased in the state of euthyroidism after levothyroxine replacement compared to the hypothyroidism before levothyroxine treatment. No significant change in neither nitrite/nitrate concentration (p = 0.18) nor in superoxide dismutase activity (p = 0.93) after L-T4 adjustment was found.Conclusions: Our data demonstrate that levothyroxine replacement improved oxidative status in patients with primary hypothyroidism, indexed by the significantly decreased levels of malonaldehyde (MDA) and increased catalase (CAT) activity
Triterpene Saponins from the Aerial Parts of Trifolium medium L. var. sarosiense
Seven previously unreported triterpene glycosides (1−7) were isolated from methanol extract of the aerial parts of
Trifolium medium var. sarosiense (zigzag clover). Their structures were established by the extensive use of 1D and 2D NMR experiments along with ESI-MS and HRMS analyses. Compounds 1−7 are oleanane derivatives characterized by the presence of a keto group at C-22 of an aglycone and a primary alcoholic function at C-24 and differing functions at C-30. Among these, compounds 1−3 and 6 showed a secondary alcoholic function at C-11, which is methoxylated in compounds 4 and 7. Compound 5 was shown to possess a known aglycone, wistariasapogenol A; however, it is described here for the first time as a saponin constituent of the Trifolium genus. Some aspects of taxonomic classification of zigzag clover are also discussed

We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer's disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer's disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer's disease
New insights into the genetic etiology of Alzheimer's disease and related dementias
Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele
Rare coding variants in PLCG2, ABI3, and TREM2 implicate microglial-mediated innate immunity in Alzheimer's disease
We identified rare coding variants associated with Alzheimer’s disease (AD) in a 3-stage case-control study of 85,133 subjects. In stage 1, 34,174 samples were genotyped using a whole-exome microarray. In stage 2, we tested associated variants (P<1×10-4) in 35,962 independent samples using de novo genotyping and imputed genotypes. In stage 3, an additional 14,997 samples were used to test the most significant stage 2 associations (P<5×10-8) using imputed genotypes. We observed 3 novel genome-wide significant (GWS) AD associated non-synonymous variants; a protective variant in PLCG2 (rs72824905/p.P522R, P=5.38×10-10, OR=0.68, MAFcases=0.0059, MAFcontrols=0.0093), a risk variant in ABI3 (rs616338/p.S209F, P=4.56×10-10, OR=1.43, MAFcases=0.011, MAFcontrols=0.008), and a novel GWS variant in TREM2 (rs143332484/p.R62H, P=1.55×10-14, OR=1.67, MAFcases=0.0143, MAFcontrols=0.0089), a known AD susceptibility gene. These protein-coding changes are in genes highly expressed in microglia and highlight an immune-related protein-protein interaction network enriched for previously identified AD risk genes. These genetic findings provide additional evidence that the microglia-mediated innate immune response contributes directly to AD development

Multiancestry analysis of the HLA locus in Alzheimer’s and Parkinson’s diseases uncovers a shared adaptive immune response mediated by HLA-DRB1*04 subtypes
Across multiancestry groups, we analyzed Human Leukocyte Antigen (HLA) associations in over 176,000 individuals with Parkinson’s disease (PD) and Alzheimer’s disease (AD) versus controls. We demonstrate that the two diseases share the same protective association at the HLA locus. HLA-specific fine-mapping showed that hierarchical protective effects of HLA-DRB1*04 subtypes best accounted for the association, strongest with HLA-DRB1*04:04 and HLA-DRB1*04:07, and intermediary with HLA-DRB1*04:01 and HLA-DRB1*04:03. The same signal was associated with decreased neurofibrillary tangles in postmortem brains and was associated with reduced tau levels in cerebrospinal fluid and to a lower extent with increased Aβ42. Protective HLA-DRB1*04 subtypes strongly bound the aggregation-prone tau PHF6 sequence, however only when acetylated at a lysine (K311), a common posttranslational modification central to tau aggregation. An HLA-DRB1*04-mediated adaptive immune response decreases PD and AD risks, potentially by acting against tau, offering the possibility of therapeutic avenues
