    Sputter-engineering a first-order magnetic phase transition in sub-15-nm-thick single-crystal FeRh films

    Equiatomic FeRh alloys undergo a fascinating first-order metamagnetic phase transition (FOMPT) just above room temperature, which has attracted reinvigorated interest for applications in spintronics. Until now, all attempts to grow nanothin FeRh alloy films have consistently shown that FeRh layers tend to grow in the Volmer-Weber growth mode. Here we show that sputter-grown sub-15-nm-thick FeRh alloy films deposited at low sputter-gas pressure, typically ∌0.1 Pa, onto (001)-oriented MgO substrates, grow in a peening-induced Frank-van der Merwe growth mode for FeRh film thicknesses above 5 nm, circumventing this major drawback. The bombardment of high-energy sputtered atoms, the atom-peening effect, induces a rebalancing between adsorbate-surface and adsorbate-adsorbate interactions, leading to the formation of a smooth continuous nanothin FeRh film. Chemical order in the films increases with the FeRh thickness, tFeRh, and varies monotonically from 0.75 up to 0.9. Specular x-ray diffraction scans around Bragg peaks show Pendellösung fringes for films with tFeRh≄5.2 nm, which reflects in smooth well-ordered densified single-crystal FeRh layers. The nanothin film's roughness varies from 0.6 down to about 0.1 nm as tFeRh increases, and scales linearly with the integral breadth of the rocking curve, proving its microstructured origin. Magnetometry shows that the FOMPT in the nanothin films is qualitatively similar to that of the bulk alloy, except for the thinnest film of 3.7 nm

    Emanuele Tesauro nos trópicos: caminhos de uma tradução e crítica teórico-histórica em Organização do Conhecimento

    After a series of historical-epistemological studies, focused on the Knowledge Organization (KO), which call attention to the thought of Emanuele Tesauro, a seventeenth-century theoretician, the urgency of bringing to discussion about the importance of translation, as a way of updating, his work. The present reflection tries to understand the Brazilian studies on Tesauro developed centrally in the last years, from the epistemological-historical perspective and the philosophy of the language, leading to the (necessary) interpretative experience of translation of the theoretical. The corpus of the proposal is focused on the introductory paragraph of the Categorical Index, a discourse that places Tesauro at the heart of the historical construction of the KO and, at the same time, at the forefront of the challenges of innovation in the contemporary domain. The Tesaurian Index is present in the third chapter, “Cagioni Instrumentali delle argutezze oratorie simboliche et lapidarie”, of his work Il Cannocchiale Aristotelico, published in 1670, an essential treatise for the modern understanding of metaphor as a possibility of understanding the world through language. One of the ways to enable this theoretical and epistemological deepening is the translation of his work, which can be considered a way of interpretation and updating, since it is a work of the seventeenth century. Starting from the point of view that the translation is an exercise of communication between different languages, epochs, histories, finally, cultures, that is to relate the meaning of a word with other words that do not belong to the same culture, the work was done involving four phases: definition of the scope of translation focusing on the relationship between Tesauro and KO, reaching the Categorical Index; first contact with the work with appropriation of the Introduction and the Categorical Index; translation proper, which corresponds to the creative part of the translation; evaluation of the translation in light of the KO lexicon. The Tesauro’s research practice and its work in KO seems immediately adherent, mainly by the (co) incidence of the name between subject and instrument (documentary language). The Tesauro’s metalinguistic character evokes not only a way of understanding Aristotelian rhetoric, but of establishing a writing practice, as well as, together with the presentation of the method of such a practice, filled with the interweaving of information, concepts, practices. This is, in short, Emanuele's twist: he is using logical (analytic-conceptual) Aristotle, but under a rhetorical (discursive) Aristotle's theory to indicate the dynamics of words in relation to things. With epistemological-historical lenses, focused on the constant instance of recontextualization, via the slow and necessary process of translation, Tesauro's thinking opens up, establishing itself not only as adherent, but as co-founder (among many margins of theoretical construction of the domain) of the OC. From the metaphorical plane, we can see that Emanuele Tesauro enters the heart of the most difficult and instigating dilemmas, current and historical, of the KO itself

    Superpulsed low-level laser therapy protects skeletal muscle of mdx mice against damage, inflammation and morphological changes delaying dystrophy progression.

    Aim: To evaluate the effects of preventive treatment with low-level laser therapy (LLLT) on progression of dystrophy in mdx mice. Methods: Ten animals were randomly divided into 2 experimental groups treated with superpulsed LLLT (904 nm, 15 mW, 700 Hz, 1 J) or placebo-LLLT at one point overlying the tibialis anterior muscle (bilaterally) 5 times per week for 14 weeks (from 6th to 20th week of age). Morphological changes, creatine kinase (CK) activity and mRNA gene expression were assessed in animals at 20th week of age. Results: Animals treated with LLLT showed very few morphological changes in skeletal muscle, with less atrophy and fibrosis than animals treated with placebo-LLLT. CK was significantly lower (p = 0.0203) in animals treated with LLLT (864.70 U.l−1, SEM 226.10) than placebo (1708.00 U.l−1, SEM 184.60). mRNA gene expression of inflammatory markers was significantly decreased by treatment with LLLT (p<0.05): TNF-α (placebo-control = 0.51 ”g/”l [SEM 0.12], - LLLT = 0.048 ”g/”l [SEM 0.01]), IL-1ÎČ (placebo-control = 2.292 ”g/”l [SEM 0.74], - LLLT = 0.12 ”g/”l [SEM 0.03]), IL-6 (placebo-control = 3.946 ”g/”l [SEM 0.98], - LLLT = 0.854 ”g/”l [SEM 0.33]), IL-10 (placebo-control = 1.116 ”g/”l [SEM 0.22], - LLLT = 0.352 ”g/”l [SEM 0.15]), and COX-2 (placebo-control = 4.984 ”g/”l [SEM 1.18], LLLT = 1.470 ”g/”l [SEM 0.73]). Conclusion: Irradiation of superpulsed LLLT on successive days five times per week for 14 weeks decreased morphological changes, skeletal muscle damage and inflammation in mdx mice. This indicates that LLLT has potential to decrease progression of Duchenne muscular dystrophy

    Anoxia- and hypoxia-induced expression of LDH-A* in the Amazon Oscar, Astronotus crassipinis

    Adaptation or acclimation to hypoxia occurs via the modulation of physiologically relevant genes, such as erythropoietin, transferrin, vascular endothelial growth factor, phosphofructokinase and lactate dehydrogenase A. In the present study, we have cloned, sequenced and examined the modulation of the LDH-A gene after an Amazonian fish species, Astronotus crassipinis (the Oscar), was exposed to hypoxia and anoxia. In earlier studies, we have discovered that adults of this species are extremely tolerant to hypoxia and anoxia, while the juveniles are less tolerant. Exposure of juveniles to acute hypoxia and anoxia resulted in increased LDH-A gene expression in skeletal and cardiac muscles. When exposed to graded hypoxia juveniles show decreased LDH-A expression. In adults, the levels of LDH-A mRNA did not increase in hypoxic or anoxic conditions. Our results demonstrate that, when given time for acclimation, fish at different life-stages are able to respond differently to survive hypoxic episodes

    Characterization of Shewanella oneidensis MtrC: a cell-surface decaheme cytochrome involved in respiratory electron transport to extracellular electron acceptors

    MtrC is a decaheme c-type cytochrome associated with the outer cell membrane of Fe(III)-respiring species of the Shewanella genus. It is proposed to play a role in anaerobic respiration by mediating electron transfer to extracellular mineral oxides that can serve as terminal electron acceptors. The present work presents the first spectropotentiometric and voltammetric characterization of MtrC, using protein purified from Shewanella oneidensis MR-1. Potentiometric titrations, monitored by UV–vis absorption and electron paramagnetic resonance (EPR) spectroscopy, reveal that the hemes within MtrC titrate over a broad potential range spanning between approximately +100 and approximately -500 mV (vs. the standard hydrogen electrode). Across this potential window the UV–vis absorption spectra are characteristic of low-spin c-type hemes and the EPR spectra reveal broad, complex features that suggest the presence of magnetically spin-coupled low-spin c-hemes. Non-catalytic protein film voltammetry of MtrC demonstrates reversible electrochemistry over a potential window similar to that disclosed spectroscopically. The voltammetry also allows definition of kinetic properties of MtrC in direct electron exchange with a solid electrode surface and during reduction of a model Fe(III) substrate. Taken together, the data provide quantitative information on the potential domain in which MtrC can operate

    A novel isolator-based system promotes viability of human embryos during laboratory processing

    In vitro fertilisation (IVF) and related technologies are arguably the most challenging of all cell culture applications. The starting material is a single cell from which one aims to produce an embryo capable of establishing a pregnancy eventually leading to a live birth. Laboratory processing during IVF treatment requires open manipulations of gametes and embryos, which typically involves exposure to ambient conditions. To reduce the risk of cellular stress, we have developed a totally enclosed system of interlinked isolator-based workstations designed to maintain oocytes and embryos in a physiological environment throughout the IVF process. Comparison of clinical and laboratory data before and after the introduction of the new system revealed that significantly more embryos developed to the blastocyst stage in the enclosed isolator-based system compared with conventional open-fronted laminar flow hoods. Moreover, blastocysts produced in the isolator-based system contained significantly more cells and their development was accelerated. Consistent with this, the introduction of the enclosed system was accompanied by a significant increase in the clinical pregnancy rate and in the proportion of embryos implanting following transfer to the uterus. The data indicate that protection from ambient conditions promotes improved development of human embryos. Importantly, we found that it was entirely feasible to conduct all IVF-related procedures in the isolator-based workstations

    Mood is a key determinant of cognitive performance in community-dwelling older adults: a cross-sectional analysis

    First Online: 06 October 2012Identification of predictors of cognitive trajectories through the establishment of composite or single-parameter dimensional categories of cognition and mood may facilitate development of strategies to improve quality of life in the elderly. Participants (n = 487, aged 50+ years) were representative of the Portuguese population in terms of age, gender, and educational status. Cognitive and mood profiles were established using a battery of neurocognitive and psychological tests. Data were subjected to principal component analysis to identify core dimensions of cognition and mood, encompassing multiple test variables. Dimensions were correlated with age and with respect to gender, education, and occupational status. Cluster analysis was applied to isolate distinct patterns of cognitive performance and binary logistic regression models to explore interrelationships between aging, cognition, mood, and socio-demographic characteristics. Four main dimensions were identified: memory, executive function, global cognitive status, and mood. Based on these, strong and weak cognitive performers were distinguishable. Cluster analysis revealed further distinction within these two main categories into very good, good, poor, and very poor performers. Mood was the principal factor contributing to the separation between very good and good, as well as poor and very poor, performers. Clustering was also influenced by gender and education, albeit to a lesser extent; notably, however, female gender × lower educational background predicted significantly poorer cognitive performance with increasing age. Mood has a significant impact on the rate of cognitive decline in the elderly.
