    The Development, Implementation, and Evaluation of a Preaching Discipleship Training Program for Church Members in the Knoxville First Seventh-day Adventist Church

    Problem The Knoxville First Seventh-day Adventist Church has members endowed with the spiritual gift of speaking (preaching). However, no training program was available for these gifted members to develop and practice their preaching skills. The senior pastor, associate pastor, and guest speakers handled the preaching responsibilities, while these members were not edifying the congregation through the exercise of their preaching gift. The absence of a preaching training program also revealed a neglect of the pastoral responsibility to disciple members in the area of preaching. This project was an effort to train gifted members in the preaching craft for a potential preaching ministry and fulfill a pastoral responsibility to disciple and equip members for ministry. Method A six-month preaching seminar was designed and delivered in the Knoxville First Seventh-day Adventist Church from March to August of 2018. Eleven members attended a two-month instructional phase and a four-month practicum phase on developing and delivering sermons. Seminar methodology included observation, instruction, application, and evaluation. Data collection involved reflective journaling, sermon reflection forms, video-recording lectures and sermons, and a seminar evaluation form. Results Ten regular participants and one alternate completed training. The instructor and a pastoral leader were also available as preaching mentors for trainees. Participants learned a method of sermon development. They also preached three practice sermons and were evaluated on content and delivery. Trainees developed preaching skills and increased in speaking confidence. Several participants were also interested in follow-up training, and expressed an interest in exploring future preaching opportunities. Conclusions Feedback from trainees revealed that the seminar was beneficial and successful. I also benefited from this training event. Lectures, preaching practice, and mentoring are essential components of preaching discipleship. To improve training, future seminars should offer (a) direct guidance from the instructor as trainees develop practice sermons, (b) shorter lectures, (c) a shorter training duration, (d) a smaller class size, and (e) sufficient practical exercises in exegesis and making appeals

    A True-Blue American Paradox: The Constitutional Tension between Religious Liberty and Sunday Legislation in the United States

    A fundamental legal tension exists between religious liberty and Sunday legislation in the United States. This tension is based upon the fact that the United States Supreme Court has ruled consistently that Sunday rest has developed into a secular principle, and no longer possesses the religious significance that it once had. As such, the Court has claimed that the individual states are within their constitutional authority to regulate the use of Sunday as a common day of rest for the welfare of their citizens. To contest this claim, this paper will present evidence to suggest that Sunday rest is still inherently religious, and that its legal enforcement as a common day of rest circumvents the First and Fourteenth Amendments. This evidence will also show that Sunday legislation poses a significant threat to religious liberty, and thus presents one of the great legal paradoxes of American constitutional history

    Investigation of laser-induced grating spectroscopy of O2 for accurate temperature measurements towards applications in harsh environments

    We present an in-depth investigation of laser-induced grating spectroscopy (LIGS) for temperature measurements in practical applications using a narrow-band dye laser with 760 nm wavelength and a pulse duration of 8 ns as the source for the pump beams creating the laser-induced grating. The pump laser wavelength was set to be either resonant with the (Formula presented.) transition from the (Formula presented.) band of O2 for generation of thermal LIGS or nonresonant for generation of purely electrostrictive LIGS. Signals were generated in ambient air as well as in high-pressure or high-temperature dry air mixtures. Pump laser irradiances up to 11 GW/cm2 were used, which resulted in strong electrostrictive contribution to the overall LIGS signals at atmospheric pressure, with a low thermal contribution due to the weak absorption by the singlet O2 (Formula presented.). The advantage and disadvantage of thermal or electrostrictive LIGS for temperature measurements are discussed, as well as potential applications in high-pressure environments. Furthermore, the precision of the temperature measurement is discussed by comparing different analysis methods

    Gain mechanism of femtosecond two-photon-excited lasing effect in atomic hydrogen

    By aiming to establish single-ended standoff combustion diagnostics, bidirectional lasing emissions of atomic hydrogen at 656 nm wavelength have been generated via two-photon resonant excitation by focusing 205 nm femtosecond laser pulses into a premixed CH4/O2 flame. The forward lasing strength is approximately one order of magnitude stronger than that of the backward one, due to the geometry of traveling wave excitation over a 2-mm-long pencil-shaped gain volume and the short gain lifetime of 3.5 ps. The gain coefficient of hydrogen lasing was determined to approximate 52/cm. As for the underlying physics of hydrogen lasing, amplified spontaneous emission (ASE) occurs simultaneously with four-wave mixing (FWM), and ASE dominates in the forward direction, whereas the backward lasing is virtually only ASE

    Low-noise mid-IR upconversion detector for improved IR-degenerate four-wave mixing gas sensing

    We compare a nonlinear upconversion detector with a conventional cryogenic InSb detector for the detection of coherent infrared light showing near-shot-noise-limited performance in the upconversion system. The InSb detector is limited by dark noise, which results in a 500 times lower signal-to-noise ratio. The two detectors are compared for the detection of a coherent degenerate four-wave mixing (DFWM) signal in the mid-infrared, and applied to measure trace-level acetylene in a gas flow at atmospheric pressure, probing its fundamental rovibrational transitions. In addition to lower noise, the upconversion system provides image information of the signal, thus adding new functionality compared to standard point detection methods. We further show that the upconversion detector system can be implemented as a simple replacement of the cryogenic detector. (C) 2014 Optical Society of Americ

    Multi-species PLIF study of the structures of turbulent premixed methane/air jet flames in the flamelet and thin-reaction zones regimes

    Simultaneously planar laser-induced fluorescence (PLIF) measurements of OH, CH, CH2O and toluene are carried out to investigate the structures of turbulent premixed methane/air jet flames in the flamelet regime and the thin-reaction zones regime. A premixed flame jet burner of an inner diameter of 1.5 mm is employed. Stoichiometric methane/air mixtures introduced as a jet are ignited and stabilized in a hot co-flow generated by a coaxial porous plug pilot flame surrounding the jet. The Reynolds number for the studied jet ranges from 960 to 11,500 with the characteristic Karlovitz number ranging from 1 to 60. The focus of this study is on the characterization of the structures and turbulent burning velocity of premixed flames in the flamelet and the thin-reaction zones regimes. The preheat zone is analyzed using the CH2O and toluene PLIF fields, whereas the reaction zone is analyzed using the CH and OH PLIF fields. Laser Doppler Anemometer (LDA) measurements are performed to characterize the turbulence field and it is noted that when the Reynolds/Karlovitz number increases a successive thickening of the preheat zone is observed, whereas the reaction zone, characterized by the CH layer maintains nearly the same thickness. The heat release zone, characterized by the combination of the OH and CH2O PLIF fields, is shown to nearly maintain the same thickness under the present experimental conditions. The flame surface wrinkle ratio is shown to be Reynolds number and Karlovitz number independent when the Reynolds number is high enough such that the smallest wrinkle scales reach to the length scales of the thin reaction layers. The global fuel consumption speed of the jet flame is analyzed using the toluene PLIF field and the OH PLIF field. A discrepancy in the two consumption velocities is found as the Karlovitz number increases. This is found to be a result of the broadening of the oxidation zone. These findings provide experimental support to the flamelet and thin-reaction zone regime hypotheses of turbulent premixed combustion

    Rare Copy Number Variants in \u3cem\u3eNRXN1\u3c/em\u3e and \u3cem\u3eCNTN6\u3c/em\u3e Increase Risk for Tourette Syndrome

    Tourette syndrome (TS) is a model neuropsychiatric disorder thought to arise from abnormal development and/or maintenance of cortico-striato-thalamo-cortical circuits. TS is highly heritable, but its underlying genetic causes are still elusive, and no genome-wide significant loci have been discovered to date. We analyzed a European ancestry sample of 2,434 TS cases and 4,093 ancestry-matched controls for rare (\u3c 1% frequency) copy-number variants (CNVs) using SNP microarray data. We observed an enrichment of global CNV burden that was prominent for large (\u3e 1 Mb), singleton events (OR = 2.28, 95% CI [1.39–3.79], p = 1.2 × 10−3) and known, pathogenic CNVs (OR = 3.03 [1.85–5.07], p = 1.5 × 10−5). We also identified two individual, genome-wide significant loci, each conferring a substantial increase in TS risk (NRXN1 deletions, OR = 20.3, 95% CI [2.6–156.2]; CNTN6 duplications, OR = 10.1, 95% CI [2.3–45.4]). Approximately 1% of TS cases carry one of these CNVs, indicating that rare structural variation contributes significantly to the genetic architecture of TS

    B-cell Zone Reticular Cell Microenvironments Shape CXCL13 Gradient Formation

    Through the formation of concentration gradients, morphogens drive graded responses to extracellular signals, thereby fine-tuning cell behaviors in complex tissues. Here we show that the chemokine CXCL13 forms both soluble and immobilized gradients. Specifically, CXCL13+ follicular reticular cells form a small-world network of guidance structures, with computer simulations and optimization analysis predicting that immobilized gradients created by this network promote B-cell trafficking. Consistent with this prediction, imaging analysis show that CXCL13 binds to extracellular matrix components in situ, constraining its diffusion. CXCL13 solubilization requires the protease cathepsin B that cleaves CXCL13 into a stable product. Mice lacking cathepsin B display aberrant follicular architecture, a phenotype associated with effective B cell homing to but not within lymph nodes. Our data thus suggest that reticular cells of the B cell zone generate microenvironments that shape both immobilized and soluble CXCL13 gradient

    Menstrual function among women exposed to polybrominated biphenyls: A follow-up prevalence study

    BACKGROUND: Alteration in menstrual cycle function is suggested among rhesus monkeys and humans exposed to polybrominated biphenyls (PBBs) and structurally similar polychlorinated biphenyls (PCBs). The feedback system for menstrual cycle function potentially allows multiple pathways for disruption directly through the hypothalamic-pituitary-ovarian axis and indirectly through alternative neuroendocrine axes. METHODS: The Michigan Female Health Study was conducted during 1997–1998 among women in a cohort exposed to PBBs in 1973. This study included 337 women with self-reported menstrual cycles of 20–35 days (age range: 24–56 years). Current PBB levels were estimated by exponential decay modeling of serum PBB levels collected from 1976–1987 during enrollment in the Michigan PBB cohort. Linear regression models for menstrual cycle length and the logarithm of bleed length used estimated current PBB exposure or enrollment PBB exposure categorized in tertiles, and for the upper decile. All models were adjusted for serum PCB levels, age, body mass index, history of at least 10% weight loss in the past year, physical activity, smoking, education, and household income. RESULTS: Higher levels of physical activity were associated with shorter bleed length, and increasing age was associated with shorter cycle length. Although no overall association was found between PBB exposure and menstrual cycle characteristics, a significant interaction between PBB exposures with past year weight loss was found. Longer bleed length and shorter cycle length were associated with higher PBB exposure among women with past year weight loss. CONCLUSION: This study suggests that PBB exposure may impact ovarian function as indicated by menstrual cycle length and bleed length. However, these associations were found among the small number of women with recent weight loss suggesting either a chance finding or that mobilization of PBBs from lipid stores may be important. These results should be replicated with larger numbers of women exposed to similar lipophilic compounds

    Genomic Relationships, Novel Loci, and Pleiotropic Mechanisms across Eight Psychiatric Disorders

    Genetic influences on psychiatric disorders transcend diagnostic boundaries, suggesting substantial pleiotropy of contributing loci. However, the nature and mechanisms of these pleiotropic effects remain unclear. We performed analyses of 232,964 cases and 494,162 controls from genome-wide studies of anorexia nervosa, attention-deficit/hyper-activity disorder, autism spectrum disorder, bipolar disorder, major depression, obsessive-compulsive disorder, schizophrenia, and Tourette syndrome. Genetic correlation analyses revealed a meaningful structure within the eight disorders, identifying three groups of inter-related disorders. Meta-analysis across these eight disorders detected 109 loci associated with at least two psychiatric disorders, including 23 loci with pleiotropic effects on four or more disorders and 11 loci with antagonistic effects on multiple disorders. The pleiotropic loci are located within genes that show heightened expression in the brain throughout the lifespan, beginning prenatally in the second trimester, and play prominent roles in neurodevelopmental processes. These findings have important implications for psychiatric nosology, drug development, and risk prediction.Peer reviewe
