Morphologic, viability and ultrastructural analysis of vitrified sheep preantral follicles enclosed in ovarian tissue
The main objective was to compare the efficiency of vitrification techniques and solutions on the preservation of morphology, ultrastructure and viability of sheep preantral follicles enclosed in ovarian tissue fragments. The fragments were cryopreserved by using macrotube vitrification (MTV), solid-surface vitrification (SSV) or conventional vitrification (CV). These techniques were combined with one of the six solutions containing 6 M ethylene glycol (EG) and with or without sucrose (SUC) (0.25 or 0.50 M) and with or without fetal calf serum (FCS) (10%). After one week, samples were warmed and histological analysis was performed, showing that the percentage of normal follicles after CV (66.20 ± 8.87%) using a solution containing 6 M EG, 0.25 M SUC and 10% FCS (vitrification solution 4 â VS4) was similar to fresh control (79.40 ± 7.83%), MTV (53.40 ± 10.60%) and SSV (56.75 ± 15.33%), all of them with the same vitrification solution (P 0.05). As the decrease of viability in non-vitrified follicles at day 2 was similar to the decrease of MTV/VS4 in the same time, follicle viability at day 2 is not affected by MTV/VS4. In conclusion, using the experimental conditions of the present study, an efficient solution (VS4: 6 M EG, 0.25 M SUC and 10% FCS) and technique (MTV) were successfully used to vitrify ovine ovarian tissue

Efeito da suplementação com castanha do Brasil sobre o ganho de peso e conversão alimentar de aves caipiras durante a fase inicial de desenvolvimento
O uso de alimentos alternativos regionais como o farelo de Castanha do Brasil na criação de aves Ă© uma realidade em todo o Brasil, diminuindo o custo na alimentação e o custo de produção. Foi suplementado na ração de aves caipiras na fase inicial o farelo da Castanha do Brasil, nos grupos (G5, G10 e G15), respectivamente. Durante todo o perĂodo avaliado os animais tiveram um incremento no ganho de peso, mas esse aumento nĂŁo diferiu entres os grupos. Foi observado que o grupo (G15) teve uma tendĂȘncia a ter um melhor Ăndice de conversĂŁo e eficiĂȘncia alimentar, contudo essa suplementação de farelo de Castanha do Brasil, nĂŁo resultou em alteração significativa nos Ăndices de conversĂŁo alimentar e ganho de peso das aves.

Vida de prateleira de ricota temperada
O estudo da vida de prateleira da ricota Ă© importante tendo em vista as caracterĂsticas que o referido alimento apresenta a qualidade da matĂ©ria-prima, as condiçÔes higiĂȘnico-sanitĂĄrias do processamento, o armazenamento, bem como, a qualidade dos ingredientes acrescidos ao produto. A ricota cremosa foi processada no Setor de LaticĂnios do NUEPPA/CCA/UFPI. Foram considerados trĂȘs tratamentos: a ricota sem tempero (GC), ricota com um tempero base acrescida de orĂ©gano (RO) ou acrescida de molho de pimenta (RP). Antes de ser adicionado Ă ricota os temperos sofreram tratamento tĂ©rmico (fervura) por +/- 2 minutos. Assim, o objetivo deste trabalho foi avaliar a qualidade microbiolĂłgica e a vida de prateleira da ricota temperada.  As ricotas apresentaram qualidade higiĂȘnico-sanitĂĄria satisfatĂłria por 30 dias de estocagem em refrigeração e a adição de tempero tratado termicamente nĂŁo alterou a qualidade higiĂȘnico-sanitĂĄria do produto
Optimization of capture protocols across species targeting up to 32000 genes and their extension to pooled DNA
Abstract Premise In-solution based capture is becoming a method of choice for sequencing targeted sequence. Methods and results We assessed and optimized a capture protocol in 20 different species from 6 different plant genus using kits from 20,000 to 200,000 baits targeting from 300 to 32,000 genes. We evaluated both the effectiveness of the capture protocol and the fold enrichment in targeted sequences. We proposed a protocol with multiplexing up to 96 samples in a single hybridization and showed it was an efficient and cost-effective strategy. We also extended the use of capture to pools of 100 samples and proved the efficiency of the method to assess allele frequency. Using a set of various organisms with different genome sizes, we demonstrated a correlation between the percentage of on-target reads vs . the relative size of the targeted sequences. Conclusion Altogether, we proposed methods, strategies, cost-efficient protocols and statistics to better evaluate and more effectively use hybridization capture
