    Consumo de álcool e morte súbita em epilepsia: uma abordagem experimental

    Using the pilocarpine model of epilepsy, we investigated the effects of alcohol consumption on the frequency of seizures in animals with epilepsy as well the underlying a possible association between alcohol intake and sudden unexpected death in epilepsy (SUDEP) occurrence. Rats were divided randomly into two groups: (A) rats with epilepsy and (B) rats with epilepsy that received a daily dose of ethanol solution (350 mg kg-1, i.p.) for 30 days. The basal frequency of seizures observed in the A and B groups during the first 30 days were 3.4±1.5 and 3.2±1.9 seizures per week per animal, respectively. In B group, it was observed a significant seizure increase (11.6±5.3) during the first 2 weeks of alcohol administration and quite interesting, one rat died suddenly after a generalized tonic-clonic seizure during this period. We concluded in our experimental study that exist a possible association between alcohol abuse and SUDEP occurrence.Utilizando o modelo de epilepsia induzido pela pilocarpina, investigamos os efeitos do consumo de álcool sobre a frequência de crises epilépticas em animais com epilepsia, como também uma possível associação entre a ingestão de álcool e ocorrência de morte súbita e inesperada nas epilepsias (SUDEP). Os animais foram randomicamente divididos em dois grupos: (A) ratos com epilepsia e (B) ratos com epilepsia que receberam uma dose diária de etanol (350 mg kg-1, i.p.) por 30 dias consecutivos. A frequência basal de crises epilépticas observadas nos grupos A e B durante os primeiros 30 dias foram de 3,4±1,5 e 3,2±1,9 crises por semana/animal, respectivamente. No grupo B, ocorreu aumento significativo na frequência de crises (11,6±5,3) durante as duas primeiras semanas de administração do álcool e de forma interessante, um animal morreu subitamente após uma crise generalizada tônico-clonica durante esse período. Concluímos em nossa abordagem experimental que existe uma possível associação entre o consumo de álcool e a ocorrência de SUDEP

    Essential oil from eucalyptus benthamii Maiden et Cambage reduces nitric oxide production in lipopolysaccharide-induced murine peritoneal macrophages

    Few studies are concerned about the essential oil extracted from leaves of Eucalyptus benthamii Maiden et Cambage that shows high content of α-pinene. The goal of this paper was to investigate the in vitro effect of the essential oil of E. benthamii and α-pinene on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in mouse peritoneal macrophages. Macrophages were harvested by washing with phosphate buffered saline and cultured with 10 μg/mL LPS. Three concentrations (5, 10, and 20 μg/mL) of the essential oil of E. benthamii and α-pinene were investigated. Nitrite levels were measured based on the Griess reaction, an indirect assay for NO production. The essential oil of E. benthamii significantly suppressed NO production in murine peritoneal macrophages at 10 and 20 μg/mL.     PET-Saúde Interprofissionalidade: experiência de interconsultas

    Introdução: O Programa de Educação pelo Trabalho para a Saúde (PET-Saúde) visa fortalecer a parceria entre o ensino e o serviço. Na sua última edição o programa traz como tema central a Interprofissionalidade, atendendo ao chamado da Organização Mundial de Saúde para implementar a educação interprofissional (EIP) como ferramenta essencial para o desenvolvimento e fortalecimento da força de trabalho na saúde, contribuindo para a redução de muitos desafios enfrentados pelos sistemas de saúde no mundo. Nesse sentido, o PET-Saúde constitui-se uma estratégia de indução de mudanças no processo de formação profissional, possibilitando a identificação de necessidades frequentes vivenciadas no serviço de saúde. A EIP ocorre quando duas ou mais profissões aprendem sobre os outros, com os outros e entre si para uma colaboração mútua e melhoria dos resultados em saúde1. Objetivo: O objetivo desse trabalho é relatar a experiência vivenciada por um grupo de acadêmicos da área da saúde, participantes do PET–Saúde Interprofissionaldiade, sobre a simulação da realização de interconsultas. Metodologia: Trata-se de um relato de experiência sobre a preparação para a realização de interconsultas, realizado por acadêmicos de enfermagem, fisioterapia, medicina, nutrição, psicologia e farmácia de variados semestres, juntamente com seus preceptores e tutores, durante o mês de agosto de 2019. Inicialmente, foram desenvolvidos os materiais físicos que iriam compor o prontuário interprofissional e subsidiar as consultas. Para o atendimento na atenção primária o grupo julgou necessário a elaboração de uma ficha de caracterização do sujeito, genograma, ecomapa e a classificação internacional de funcionalidade (CIF). Posteriormente os membros forma divididos em dois grupos (1 e 2). Resultados: O grupo 1 realizou a vivência de uma interconsulta através de simulação com paciente standartizado, enquanto o grupo 2 observou a condução do grupo 1. Após o atendimento todos os membros de ambos os grupos discutiram os aspectos positivos da interconsulta e os aspectos a serem melhorados sob a ótica das práticas colaborativas. O exercício dessa atividade possibilitou aos acadêmicos uma reflexão da importância e desafios do trabalho em equipe, desde a construção do prontuário interprofissional até a realização do atendimento. A dificuldade da realização da interconsulta está relacionada à complexidade dos saberes das diversas profissões em saúde, principalmente por não haver conhecimento sobre as práticas comuns e colaborativas entre os profissionais. Considerações Finais: Dessa forma, a partir da experiência do treino da interconsulta, evidencia-se a importância dos profissionais de saúde terem contato com profissões colegas e aprenderam a trabalhar em equipe, desde a graduação, para prestar um atendimento mais eficaz e de qualidade aos pacientes, principalmente no Sistema Único de Saúde.     A study of 82 extended HLA haplotypes in HFE-C282Y homozygous hemochromatosis subjects: relationship to the genetic control of CD8+ T-lymphocyte numbers and severity of iron overload

    BACKGROUND: It has been recently demonstrated that CD8+ T-lymphocyte numbers are genetically transmitted in association with the MHC class I region. The present study was designed with the objective of narrowing the region associated with the setting of CD8+ T-lymphocyte numbers in a population of C282Y homozygous hemochromatosis subjects, in whom a high prevalence of abnormally low CD8+ T-lymphocyte counts has been described. METHODS: The study includes 43 C282Y homozygous subjects fully characterized both phenotypically and genotypically. Clinical characterization includes measurements of iron parameters at diagnosis (transferrin saturation and serum ferritin), total body iron stores and T-cell immunophenotyping determined by flow cytometry. Genetic characterization includes HLA class I alleles (A, B and C) and four additional microsatellite markers (D6S265, D6S2222, D6S105 and D6S2239) spanning 5 Megabases in the 6p21.3 region. RESULTS: Eighty-two extended C282Y carrying haplotypes were defined. Single-locus analysis revealed that the HLA-A region was associated with CD8+ T-cell numbers. Multivariate analysis showed that the combinations of the most common HLA-A alleles (HLA-A*03, -A*02 and -A*01) were associated with significantly lower numbers of CD8+ T-lymphocytes (0.30 ± 0.14 × 10(6)/ml), in comparison with subjects carrying only one copy of those alleles (0.46 ± 0.19 × 10(6)/ml) and subjects without any copy of those alleles (0.79 ± 0.15 × 10(6)/ml;p = 0.0001). No differences were observed in CD8+ T-cell counts among control subjects carrying the same combinations of HLA-A alleles (0.47 ± 0.14; 0.45 ± 0.21 and 0.41 ± 0.17 × 10(6)/ml, respectively), therefore not supporting a direct effect of HLA specificity but rather an indirect association with a locus close to HLA-A. Multivariate analysis showed that the combination of the most common HLA-A alleles also have an impact on the clinical expression of HH in terms of iron stores, in males(p = 0.0009). CONCLUSION: The present study provides evidence supporting an inextricable link between extended HLA haplotypes, CD8+ T-lymphocyte numbers and severity of iron overload in hereditary hemochromatosis(HH). It gives additional information to better define a candidate region involved in the regulation of CD8+ T-lymphocyte numbers. A new evolutionary hypothesis concerning the inheritance of the phenotype of low CD8+ T-lymphocyte numbers associated with particular ancestral HLA haplotypes carrying the C282Y mutation and its implication on the clinical heterogeneity of HH is discussed

    Bacteriophages with Potential for Inactivation of Fish Pathogenic Bacteria: Survival, Host Specificity and Effect on Bacterial Community Structure

    Phage therapy may represent a viable alternative to antibiotics to inactivate fish pathogenic bacteria. Its use, however, requires the awareness of novel kinetics phenomena not applied to conventional drug treatments. The main objective of this work was to isolate bacteriophages with potential to inactivate fish pathogenic bacteria, without major effects on the structure of natural bacterial communities of aquaculture waters. The survival was determined in marine water, through quantification by the soft agar overlay technique. The host specificity was evaluated by cross infection. The ecological impact of phage addition on the structure of the bacterial community was evaluated by DGGE of PCR amplified 16S rRNA gene fragments. The survival period varied between 12 and 91 days, with a higher viability for Aeromonas salmonicida phages. The phages of Vibrio parahaemolyticus and of A. salmonicida infected bacteria of different families with a high efficacy of plating. The specific phages of pathogenic bacteria had no detectable impact on the structure of the bacterial community. In conclusion, V. parahaemolyticus and A. salmonicida phages show good survival time in marine water, have only a moderated impact on the overall bacterial community structure and the desired specificity for host pathogenic bacteria, being potential candidates for therapy of fish infectious diseases in marine aquaculture systems

    Antimicrobial Photodynamic Therapy: Study of Bacterial Recovery Viability and Potential Development of Resistance after Treatment

    Antimicrobial photodynamic therapy (aPDT) has emerged in the clinical field as a potential alternative to antibiotics to treat microbial infections. No cases of microbial viability recovery or any resistance mechanisms against it are yet known. 5,10,15-tris(1-Methylpyridinium-4-yl)-20-(pentafluorophenyl)-porphyrin triiodide (Tri-Py+-Me-PF) was used as photosensitizer. Vibrio fischeri and recombinant Escherichia coli were the studied bacteria. To determine the bacterial recovery after treatment, Tri-Py+-Me-PF (5.0 μM) was added to bacterial suspensions and the samples were irradiated with white light (40 W m−2) for 270 minutes. Then, the samples were protected from light, aliquots collected at different intervals and the bioluminescence measured. To assess the development of resistance after treatment, bacterial suspensions were exposed to white light (25 minutes), in presence of 5.0 μM of Tri-Py+-Me-PF (99.99% of inactivation) and plated. After the first irradiation period, surviving colonies were collected from the plate and resuspended in PBS. Then, an identical protocol was used and repeated ten times for each bacterium. The results suggest that aPDT using Tri-Py+-Me-PF represents a promising approach to efficiently destroy bacteria since after a single treatment these microorganisms do not recover their viability and after ten generations of partially photosensitized cells neither of the bacteria develop resistance to the photodynamic process

    Dietary inflammatory index and inflammatory biomarkers in adolescents from LabMed physical activity study

    Background/objectives The dietary inflammatory index (DII) is a tool to measure the diet’s inflammatory potential and has been used with adults to predict low-grade inflammation. The present study aims to assess whether this dietary score predicts low-grade inflammation in adolescents. Subjects/methods The sample comprises 329 adolescents (55.9% girls), aged 12–18 years, from LabMed Physical Activity Study. DII score was calculated based on a food-frequency questionnaire and categorized into tertiles. We collected blood samples to determine the follow inflammatory biomarkers: C-reactive protein (CRP), interleukin-6 (IL-6), complement component 3 (C3), and 4 (C4). In addition we calculated an overall inflammatory biomarker score. Odds ratios (OR) and 95% confidence intervals (95%CI) were computed from binary logistic regression models. Results DII score, comparing first with third tertile, was positively associated with IL-6 in crude model (OR = 1.88, 95% CI:1.09–3.24, ptrend = 0.011) and in fully adjusted (for biological and lifestyle variables) (OR = 3.38, 95%CI:1.24–9.20, ptrend = 0.023). Also, DII score was positively associated with C4, when fully adjusted (OR = 3.12, 95%CI:1.21–8.10, ptrend = 0.016). DII score was negatively associated with C3 in crude model, comparing first with second but not with third tertile, and no significant associations in fully adjusted model were observed, although a trend was found (OR = 1.71, 95% CI:0.63–4.66, ptrend = 0.044). No significant associations were observed between DII score and CRP. However, DII score was positively associated with the overall inflammatory biomarker score, when fully adjusted (OR = 5.61, 95% CI:2.00–15.78, ptrend = 0.002).
