    We thank Dr. Belsky (1) for his interest in our study (2), in which we investigated associations between physical activity and cognitive function in young people. He raises several important issues on the role of physical activity as a public health strategy in the prevention of cognitive decline. The issue of “neuroselection,” wherein individuals with better cognitive function are more likely to engage in healthy behaviors such as physical activity and refrain from unhealthy ones such as smoking, is a potential source of bias in observational studies. The alternative hypothesis is that of “neuroprotection,” in which engagement in healthy behaviors is likely to lead to enhanced cognitive development

    Effect of major school playground reconstruction on physical activity and sedentary behaviour: Camden active spaces.

    BACKGROUND: The physical school environment is a promising setting to increase children's physical activity although robust evidence is sparse. We examined the effects of major playground reconstruction on physical activity and sedentary time in primary schools using a quasi-experimental design (comparison group pre-test/post-test design). METHODS: Five experimental and two control schools from deprived areas of inner city London were recruited at baseline. Main outcome was physical activity and sedentary time measured from objective monitoring (Actigraph accelerometer) at one year follow up. Pupils' impressions of the new playground were qualitatively assessed post construction. RESULTS: A total of 347 pupils (mean age = 8 years, 55% boys; 36% Caucasian) were recruited into the study at baseline; 303 provided valid baseline Actigraph data. Of those, 231 (76%) completed follow-up (n = 169 intervention; n = 62 control) and 77.4% of the sample recorded at least 4 days of Actigraph wear. In mixed models adjusted for age, sex, ethnicity, ratio activity or sedentary/wear time at baseline, wear time at follow up, and school, no differences were observed in total moderate - vigorous activity (B = -1.4, 95% CI, -7.1, 4.2 min/d), light activity (B = 4.1, 95% CI, -17.9, 26.1), or sedentary time (B = -3.8, 95% CI, -29.2, 21.6 min/d) between groups. There were significant age interactions for sedentary (p = 0.002) and light intensity physical activity (p = 0.008). We observed significant reductions in total sedentary (-28.0, 95% CI, -1.9, -54.1 min/d, p = 0.037) and increases in total light intensity activity (24.6, 95% CI, 0.3, 48.9 min/d, p = 0.047) for children aged under 9 yrs. old in the intervention. CONCLUSION: Major playground reconstruction had limited effects on physical activity, but reduced sedentary time was observed in younger children. Qualitative data suggested that the children enjoyed the new playgrounds and experienced a perceived positive change in well-being and social interactions

    Fundus Autofluorescence and Spectral Domain OCT in Central Serous Chorioretinopathy

    Background. To describe the standard autofluorescence (FAF), the near infrared autofluorescence (NIA) and optical coherence tomography (OCT) patterns in central serous chorioretinopathy, correlating them with fluorescein angiography. Methods. Cross-sectional observational study, in which patients with at least seven months of CSC underwent ophthalmologic examination, fundus photography, FAF, NIA, fluorescein angiography (FA), and spectral-domain OCT. Results. Seventeen eyes of thirteen patients were included. the presentation features were a mottled hyperFAF in the detached area and areas with pigment mottling. NIA images showed areas of hyperNIA similar to FAF and localized areas of hypoNIA, which correlated with the points of leakage in the FA. OCT showed pigment epithelium detachment at the location of these hypoNIA spots. Discussion. FAF showed increased presence of fluorophores in the area of retinal detachment, which is believed to appear secondary to lipofuscin accumulation in the RPE or the presence of debris in the subretinal fluid. NIA has been related to the choroidal melanin content and there were areas of both increased and decreased NIA, which could be explained by damage ahead the retina, basically RPE and choroid.     Global metabolic response of Enterococcus faecalis to oxygen

    Oxygen and oxidative stress have become relevant components in clarifying the mechanism that weakens bacterial cells in parallel to the mode of action of bactericidal antibiotics. Given the importance of oxidative stress in the overall defense mechanism of bacteria and their apparent role in the antimicrobial mode of action, it is important to understand how bacteria respond to this stress at a metabolic level. The aim of this study was to determine the impact of oxygen on the metabolism of the facultative anaerobe Enterococcus faecalis using continuous culture, metabolomics and 13C-enrichment of metabolic intermediates. When E. faecalis was rapidly transitioned from anaerobic to aerobic growth, cellular metabolism was directed towards intracellular glutathione production and glycolysis was upregulated two-fold, which increased the supply of critical metabolite precursors (e.g. glycine and glutamate) for sulfur metabolism and glutathione biosynthesis as well as reducing power for cellular respiration in the presence of haemin. The ultimate metabolic response of E. faecalis to an aerobic environment was the upregulation of fatty acid metabolism and benzoate degradation, which was linked to important changes in the bacterial membrane composition as evidenced by changes in membrane fatty acid composition and the reduction of membrane-associated demethylmenaquinone. These key metabolic pathways associated with the response of E. faecalis to oxygen may represent potential new targets to increase the susceptibility of this bacterium to bactericidal drugs.This work was funded by the HRC (Health and Research Council of New Zealand) and the FCT (Portuguese Foundation for Science and Technology), with grant reference SFRH/BD/47016/2008

    Winter is coming:Hibernation reverses the outcome of sperm competition in a fly

Available from the publisher via the DOI in this record.Sperm commonly compete within females to fertilize ova, but research has focused on short-term sperm storage: sperm that are maintained in a female for only a few days or weeks before use. In nature, females of many species store sperm for months or years, often during periods of environmental stress, such as cold winters. Here we examine the outcome of sperm competition in the fruit fly Drosophila pseudoobscura, simulating the conditions in which females survive winter. We mated females to two males and then stored the female for up to 120 days at 4°C. We found that the outcome of sperm competition was consistent when sperm from two males was stored for 0, 1 or 30 days, with the last male to mate fathering most of the offspring. However, when females were stored in the cold for 120 days, the last male to mate fathered less than 5% of the offspring. Moreover, when sperm were stored long term the first male fathered almost all offspring even when he carried a meiotic driving sex chromosome that drastically reduces sperm competitive success under short-term storage conditions.     Investigation of faecal volatile organic compounds as biomarkers for the diagnosis of necrotising enterocolitis

    Necrotising enterocolitis (NEC) is the most prevalent and harmful illness in the neonatal intensive care unit. It primarily affects premature babies, with higher incidence in low birthweight patients. NEC usually occurs in the early days of life and may develop rapidly. Its diagnosis is based on medical observation because no early diagnostic tool is currently available. Over the last two decades, developments in analytical chemistry have allowed scientists to perform untargeted investigation of biological samples. Biomarkers are being sought, for a range of disorders, by investigating volatile organic compounds (VOCs) in bio-fluids. For the analysis of gas samples, solid phase micro-extraction (SPME) is a suitable technique to preconcentrate volatile compounds prior to analysis using devices such as gas chromatography – mass spectrometry (GC–MS). Recently, much attention has been directed towards the development of sensors and electronic noses to be used as diagnostic tools in hospitals, as they are generally more compact than a GC-MS and do not require specialised personnel. The work presented here is based on the hypothesis that faeces from patients suffering from necrotising enterocolitis show a specific pattern of volatile organic compounds in the days prior to diagnosis when compared with faeces from healthy patients. The objectives of this work were to develop two methods for the analysis of premature faeces using headspace–SPME–GC–MS and headspace – gas chromatography – sensor (HS–GC–Sensor), to analyse samples from healthy premature infants and premature infants affected by NEC using both instruments and to analyse the data collected. Methods were developed individually for each analytical technique. Two pipelines were applied for mass spectrometric data analysis while classification models were exclusively built using sensor data. Results obtained from HS–SPME–GC–MS data showed that the age at sampling had an influence on the number of compounds identified and on their intensities or relative abundance. Heptanal, 2-E-pentenal, hexanal and 2-methylbutanoic acid were identified as relevant compounds. Classifiers were built at days 1 to 6 prior to diagnosis. Accuracy, sensitivity and specificity of up to 74%, 62% and 79%, respectively, were obtained one day prior to diagnosis based on mass spectrometric data, while accuracy, sensitivity and specificity of 100% were obtained based on sensor data at two days prior to diagnosis. Therefore, classification of samples based on headspace analysis of faeces might have potential for the early diagnosis of necrotising enterocolitis

    Decrease in plasma miR-27a and miR-221 after concussion in Australian football players

    Introduction: Sports-related concussion (SRC) is a common form of brain injury that lacks reliable methods to guide clinical decisions. MicroRNAs (miRNAs) can influence biological processes involved in SRC, and measurement of miRNAs in biological fluids may provide objective diagnostic and return to play/recovery biomarkers. Therefore, this prospective study investigated the temporal profile of circulating miRNA levels in concussed male and female athletes. Methods: Pre-season baseline blood samples were collected from amateur Australian rules football players (82 males, 45 females). Of these, 20 males and 8 females sustained an SRC during the subsequent season and underwent blood sampling at 2-, 6- and 13-days post-injury. A miRNA discovery Open Array was conducted on plasma to assess the expression of 754 known/validated miRNAs. miRNA target identified were further investigated with quantitative real-time PCR (qRT-PCR) in a validation study. Data pertaining to SRC symptoms, demographics, sporting history, education history and concussion history were also collected. Results: Discovery analysis identified 18 candidate miRNA. The consequent validation study found that plasma miR-221-3p levels were decreased at 6d and 13d, and that miR-27a-3p levels were decreased at 6d, when compared to baseline. Moreover, miR-27a and miR-221-3p levels were inversely correlated with SRC symptom severity. Conclusion: Circulating levels of miR-27a-3p and miR-221-3p were decreased in the sub-acute stages after SRC, and were inversely correlated with SRC symptom severity. Although further studies are required, these analyses have identified miRNA biomarker candidates of SRC severity and recovery that may one day assist in its clinical management
