Pik3r1 Is Required for Glucocorticoid-Induced Perilipin 1 Phosphorylation in Lipid Droplet for Adipocyte Lipolysis.
Glucocorticoids promote lipolysis in white adipose tissue (WAT) to adapt to energy demands under stress, whereas superfluous lipolysis causes metabolic disorders, including dyslipidemia and hepatic steatosis. Glucocorticoid-induced lipolysis requires the phosphorylation of cytosolic hormone-sensitive lipase (HSL) and perilipin 1 (Plin1) in the lipid droplet by protein kinase A (PKA). We previously identified Pik3r1 (also called p85α) as a glucocorticoid receptor target gene. Here, we found that glucocorticoids increased HSL phosphorylation, but not Plin1 phosphorylation, in adipose tissue-specific Pik3r1-null (AKO) mice. Furthermore, in lipid droplets, the phosphorylation of HSL and Plin1 and the levels of catalytic and regulatory subunits of PKA were increased by glucocorticoids in wild-type mice. However, these effects were attenuated in AKO mice. In agreement with reduced WAT lipolysis, glucocorticoid- initiated hepatic steatosis and hypertriglyceridemia were improved in AKO mice. Our data demonstrated a novel role of Pik3r1 that was independent of the regulatory function of phosphoinositide 3-kinase in mediating the metabolic action of glucocorticoids. Thus, the inhibition of Pik3r1 in adipocytes could alleviate lipid disorders caused by excess glucocorticoid exposure
Emociones y salud actividades propiciadoras de emociones positivas
Se parte del concepto de emoción positiva, vivida en contextos, cuya experiencia subjetiva, es
auto-percibida como bienestar (Seligman & Csikszentmihalyi, 2000), razón por la que se
considera aquí como promotora de Salud. Así, este estudio de carácter cualitativo organiza,
con una “teoría basada en los datos”, las entrevistas semi-estructuradas, en una muestra
intencional de 60 profesores. Los contenidos discursivos obtenidos se sometieron a análisis de
contenido, de donde surgió un “árbol de categorías“ con 3 generaciones, alrededor de las
actividades que proporcionan emociones positivas. Las actividades identificadas que
proporcionan emociones positivas agregan un árbol de 13 categorías, que serán presentadas
por orden decreciente de unidades discursivas: “Amor”, “Amigos” “Sentir la Naturaleza”,
“Familia”, “Fare Niente”, “Pie Fuera de la Puerta”, “Musicalidad”, “Dormir”, “Lecturas”,
“Trabajar en lo que nos gusta”, “Convivir”, “Baño” y finalmente, la categoría “Tener Dinero”.
Se concluye que será posible proporcionar el desarrollo de estas actividades en ambiente
laboral, para experienciar el bienestar subjetivo
Evaluation of entropy and JM-distance criterions as features selection methods using spectral and spatial features derived from LANDSAT images
A study area near Ribeirao Preto in Sao Paulo state was selected, with predominance in sugar cane. Eight features were extracted from the 4 original bands of LANDSAT image, using low-pass and high-pass filtering to obtain spatial features. There were 5 training sites in order to acquire the necessary parameters. Two groups of four channels were selected from 12 channels using JM-distance and entropy criterions. The number of selected channels was defined by physical restrictions of the image analyzer and computacional costs. The evaluation was performed by extracting the confusion matrix for training and tests areas, with a maximum likelihood classifier, and by defining performance indexes based on those matrixes for each group of channels. Results show that in spatial features and supervised classification, the entropy criterion is better in the sense that allows a more accurate and generalized definition of class signature. On the other hand, JM-distance criterion strongly reduces the misclassification within training areas
An integrated software system for geometric correction of LANDSAT MSS imagery
A system for geometrically correcting LANDSAT MSS imagery includes all phases of processing, from receiving a raw computer compatible tape (CCT) to the generation of a corrected CCT (or UTM mosaic). The system comprises modules for: (1) control of the processing flow; (2) calculation of satellite ephemeris and attitude parameters, (3) generation of uncorrected files from raw CCT data; (4) creation, management and maintenance of a ground control point library; (5) determination of the image correction equations, using attitude and ephemeris parameters and existing ground control points; (6) generation of corrected LANDSAT file, using the equations determined beforehand; (7) union of LANDSAT scenes to produce and UTM mosaic; and (8) generation of output tape, in super-structure format

Life histories stories of homeless people, social exclusion situations and transforming encounters
Este trabalho apresenta, a partir de histórias de vida, características do processo de "encontro transformador" entre dois moradores de rua e uma professora, que foi "ponto de apoio" positivo em suas vidas. O "encontro transformador" é interação entre os seres humanos que possibilita a transformação dos envolvidos, no sentido de despertar suas potencialidades, a retomada do sentido da vida, promovendo-lhes a resiliência, que é a capacidade humana de fazer frente às adversidades da vida, superá-las e sair delas fortalecidos e, inclusive, transformados. O estudo longitudinal realizado envolveu o resgate de histórias de vida, através de entrevistas abertas, fotografias, registros em Diário de Campo e desenhos feitos pelos sujeitos de observação. Na interpretação dos dados contemplou-se o emprego de conceitos de determinadas teorias de: Psicologia, Geografia, Sociologia, Direito, Ciências da Educação, Complexidade e Sistêmica, em diálogo entre diferentes disciplinas. A análise do fenômeno - em que o morar na rua surgiu como situação existencial excludente - revelou nova configuração nas psiques dos moradores de rua, em movimento de transformação. No fenômeno observado - complexo - desvelou-se a dificuldade dos moradores de rua estudados de se manterem no processo resiliente sem o apoio efetivo da Sociedade Civil e do Estado, a partir de políticas públicas voltadas para esse tipo de população. Conclui-se pela importância dos resultados deste trabalho como contribuição para a ampliação de processos de formação, não só de profissionais que atuam com moradores de rua como de integrantes da sociedade em geral, norteados por uma visão solidária de busca de cidadania para todos.This work presents, based on life histories Stories, characteristics of a "transforming encounter" that took place between two homeless people and oneA teacher. and that was It was a positive "point of support" in their lives. "Transforming encounter" is an interaction between human beings that enables the transformation of the people involved, in the sense that their potentialities are awoken, they recover their direction of life, and their resilience is promoted. Resilience is the human capacity to deal with life's adversities, overcome them;so, and become stronger and even transformed. The longitudinal study that was conducted involved the obtention of life histories stories through open interviews, photographs, registers in a Field Diary and drawings made by the observation subjects. In the data interpretation, concepts from certain theories were used, p.from the following areas (e.g.: Psychology, Geography, Sociology, Law, Education Sciences, Complexity and Systemic Theory), in a dialog among different disciplines. The analysis of the phenomenon analysis - in which living on the street emerged as an excluding existential situation - revealed a new configuration in the homeless' psyches, which were undergoing a transformation movement. The observed phenomenon - awhich was complex one - showed the difficulty that the studied homeless individuals have into maintaining themselves in the resilient process without the effective support of the CCivil Society's and of the State's support, through public policies targeted at this kind of population. Specification and preliminary design of the CARTA system for satellite cartography
Digital imagery acquired by satellite have inherent geometrical distortion due to sensor characteristics and to platform variations. In INPE a software system for geometric correction of LANDSAT MSS imagery is under development. Such connected imagery will be useful for map generation. Important examples are the generation of LANDSAT image-charts for the Amazon region and the possibility of integrating digital satellite imagery into a Geographic Information System
Inter-And Transdisciplinary Action-Formation-Research with People Involved in the Homeless Issue
Este artigo apresenta reflexões teórico-metodológicas sobre processo de investigação de pós-doutorado que objetivava basicamente construir - na ação - estratégia de "psico-sócio-formação" de pessoas envolvidas com a questão do morador de rua; criar e aplicar um recurso metodológico operacional denominado "conto de encontro transformador". Do ponto de vista teórico, sob perspectivas inter e transdisciplinares de produção do conhecimento, essa "pesquisa-ação-formação" baseou-se no encontro dialógico entre os conhecimentos sobre "encontro transformador", "resiliência" e "ágape" e construtos teóricos da área da Educação, com ênfase no processo de autoformação. O projeto contou com vinte participantes: moradores de rua; trabalhadores de instituições de apoio a moradores de rua; técnicos das Secretarias de Assistência Social da Prefeitura de São Paulo e/ou da Secretaria da Saúde; e provenientes da Universidade de São Paulo e de outras Universidades do Brasil, França e Canadá.This paper presents theoretical and methodological reflections on a postdoctoral investigation process that aimed to build - on action - a psychological, social and formational strategy targeted at people involved in the question of homeless individuals; and to create and apply an operational methodological resource called "transforming encounter short story". From the theoretical point of view, under inter- and transdisciplinary perspectives of knowledge production, this action-formation-research was based on the dialogic encounter between the concepts of "transforming encounter", "resilience" and "agape"; and on theoretical constructs from the Education area, with emphasis on the self-formation process. The project had twenty participants: homeless people; workers from institutions that support the homeless; technicians with the Social Work Departments of the Municipal Government of São Paulo and/or with the Health Department; and participants from the University of São Paulo and from other Universities of Brazil, France and Canada
Clinical spectrum of females with HCCS mutation: from no clinical signs to a neonatal lethal form of the microphthalmia with linear skin defects (MLS) syndrome
Background: Segmental Xp22.2 monosomy or a heterozygous HCCS mutation is associated with the microphthalmia with linear skin defects (MLS) or MIDAS (microphthalmia, dermal aplasia, and sclerocornea) syndrome, an X-linked disorder with male lethality. HCCS encodes the holocytochrome c-type synthase involved in mitochondrial oxidative phosphorylation (OXPHOS) and programmed cell death. Methods: We characterized the X-chromosomal abnormality encompassing HCCS or an intragenic mutation in this gene in six new female patients with an MLS phenotype by cytogenetic analysis, fluorescence in situ hybridization, sequencing, and quantitative real-time PCR. The X chromosome inactivation (XCI) pattern was determined and clinical data of the patients were reviewed. Results: Two terminal Xp deletions of ≥11.2 Mb, two submicroscopic copy number losses, one of ~850 kb and one of ≥3 Mb, all covering HCCS, 1 nonsense, and one mosaic 2-bp deletion in HCCS are reported. All females had a completely (>98:2) or slightly skewed (82:18) XCI pattern. The most consistent clinical features were microphthalmia/anophthalmia and sclerocornea/corneal opacity in all patients and congenital linear skin defects in 4/6. Additional manifestations included various ocular anomalies, cardiac defects, brain imaging abnormalities, microcephaly, postnatal growth retardation, and facial dysmorphism. However, no obvious clinical sign was observed in three female carriers who were relatives of one patient. Conclusion: Our findings showed a wide phenotypic spectrum ranging from asymptomatic females with an HCCS mutation to patients with a neonatal lethal MLS form. Somatic mosaicism and the different ability of embryonic cells to cope with an OXPHOS defect and/or enhanced cell death upon HCCS deficiency likely underlie the great variability in phenotypes
