Construction of three new Gateway® expression plasmids for Trypanosoma cruzi

    Get PDF
We present here three expression plasmids for Trypanosoma cruzi adapted to the Gateway® recombination cloning system. Two of these plasmids were designed to express trypanosomal proteins fused to a double tag for tandem affinity purification (TAPtag). The TAPtag and Gateway® cassette were introduced into an episomal (pTEX) and an integrative (pTREX) plasmid. Both plasmids were assayed by introducing green fluorescent protein (GFP) by recombination and the integrity of the double-tagged protein was determined by western blotting and immunofluorescence microscopy. The third Gateway adapted vector assayed was the inducible pTcINDEX. When tested with GFP, pTcINDEX-GW showed a good response to tetracycline, being less leaky than its precursor (pTcINDEX).Fil: Alonso, Victoria Lucia. The reformer is used to produce pure hydrogen able to feed a Proton Exchange Membrane Fuel Cell. Mass and energy balances are used to obtain the nonlinear dynamic model of both the reforming and the separation stages. Constraints, system nonlinearities and flexible cost function are the main reasons to select an NMPC controller, which is tested against the ordinary differential equations as simulation model, and has an internal model based on the sample data technique.Accepted versio

Martín-Serra, A., Figueirido, B., Serrano, F., Palmqvist, P. MODULAR EVOLUTION OF THE CARNIVORAN PELVIC GIRDLE: A THREE-DIMENSIONAL MORPHOMETRIC APPROACH. 75th meeting of Vertebrate Palaeontology. Dallas (Texas). Journal of Vertebrate Palaeontology Supplement (Program and Abstracts) P. 176The pelvic girdle is a key skeletal structure within the appendicular skeleton in quadrupedal mammals. The pelvis anchors many important muscles and connects the hind limb to the axial skeleton. However, unlike other appendicular bones, each hemipelvis is composed of three different girdle bones: ilium, ischium and pubis. Here we investigate if the functional and developmental interactions among these bones accounts for the integration and modularity of the pelvis in mammalian carnivores. We use carnivorous mammals as a case study because our recent work has demonstrated that their appendicular skeleton is also integrated by functional reasons. A series of landmarks in 3D on one half of the pelvic girdle were digitized in a wide sample of living carnivorans. The landmarks were divided into four basic developmental units: illium, ischium, pubis and acetabulum. The latter was considered as a different unit because it interacts with the femoral head during development. Later, we tested different modularity hypotheses that consider all possible modules formed by the combination of these four developmental units. For each hypothesis, we calculated the RV coefficient, a proxy for morphological covariation. We compared each specific hypothesis with a distribution of RV coefficients resulting from randomly-defined modules to assess for statistical significance. One of the hypotheses with more statistical support separates the four original units as modules, which indicates a strong influence of development. Other supported hypotheses clearly point towards an association between the ischium and the pubis, with the illium and acetabulum more or less independent. However, these hypotheses cannot be unequivocally ascribed to functional interactions, because the ischium and the pubis also share some developmental processes. These results clearly indicate that the carnivoran pelvic girdle preserves a developmental modular structure with little modification attributable to functional adaptations, which agrees with previous studies that showed that the pelvis is conservative within each carnivoran family.Universidad de Málaga. The cardiovascular system in healthy individuals possesses an intrinsic rythm, which varies considerably throughout time. This propriety is fundamental to the adaptability of the system to constant changes, and is denominated as variability. Several theories and descoveries concerning the variability of signals of the cardiovascular systems have been published by several authors, over the last thirty years. These studies call into question the significance of homeostasis of the system. Throughout these investigations, several types of analysis and characterizations were proposed to corrolate how certain pathologies diminish the variability of cardiovascular parameters, leading to the emergence of periodical and poorly adaptable patterns, which took over this system. One of the clinical entities that has already been proven to affect the variability of cardiac frequency, is heart failure. This disease, which affects 1 to 2% of the World's population, presents, in Portugal, an intra-hospital mortality rate of 12,5%. Considering this problematic, an idea to train an artificial inteligence, the neuronal networks, was proposed in order to detect and evaluate the periodicity patterns in the evolution of the cardiovascular parameters (specifically, the cardiac frequency), in a heart failure background. With this analysis, the network transmits its evaluation of the vital prognostic of the patients. After the training and testing phases of the neuronal network, the prognostics were correct in 96,385% of the cases, with a percentage of response certainty of 81,07%. We concluded that this analysis of the variability of the cardiac frequency using a neuronal network, is possible. Moreover, we predict that this may prove to be a very useful tool in clinical practice in the future, and that it might also have several important applications.O sistema cardiovascular de indivíduos saudáveis apresenta um ritmo intrínseco que varia consideravelmente ao longo do tempo.

On the Metacognitive dimension of suspended syntactic constructions. A descriptive study on aphasic Spanish speakers

    Get PDF
By taking into consideration conversational outputs from aphasic Spanish speakers, a functional characterisation of suspended syntactic constructions will be provided here. Suspensions of syntactic constructions may be initially thought of as attributed to a language processing deficit in people with aphasia, which is, in fact, only partly the case. An examination of conversational data demonstrates, however, that a comprehensive explanation of syntactic suspensions requires a re-assessment of this phenomenon in the realm of meta-cognitive processes associated with language behaviour. Five general types of procedures and contexts for suspended syntactic constructions will be proposed and discussed in this paper. They differentially involve a series of metacognitive processes such as monitoring for anticipation and prevention of foreseeable mistakes, intersemiotic control of language production and communication by the use of gesturing, motivational and intentional aspects associated with language use, collaborative tasks in language production and theory of mind phenomena

Without the penetration of Portuguese language through African land, particularly in Angola, since the period of expansion by Portuguese navigators, sailors and clergy, it wouldn't have been possible to develop this work, that in a cultural perspective addresses the change that the written chronic produced in the paper at a given time would have caused, and the effects that occurred since then in the Angolan society and Portuguese Literature. First by the contact of direct players and then by the education released there, the Africans were able to take advantage of the Portuguese literary system that was offered, and changed a civilization paradigm, the passing of knowledge through writing. They demonstrated with this, being able not only to develop their culture, but also to pursue the creation of a literature with African roots capable to distinguish itself from the original. From this ability to accept and adapt to the new reality, the Angolans searched to create in their space, opposing to a form expressed by the colonizer, elements that would form the basis of their identity values, and with this, assert themselves in the common cultural area. We had watched the growth of the work of Ernesto Lara (Filho), that through chronic developed in the journalistic field and other elements, didn't lost the mark of a literary writing, participating in the natural process of creating literature. For these reasons is determined the importance of the study of chronic traditionally aimed at the newspaper. It had become one of the routes selected for the dissemination of Portuguese language and Angolan thought, thereby contributing to the definition of the elements that shape the Angolan cultural standard. The Portuguese language being accepted by the Angolan community passed also, to be part of a new space and provided a tribute that made it win new outlines in the enrichment generated from the meeting enjoyed between the two cultures

The reasons of this trend are clear: contributing to a reduction of gas emissions and dependency on fossil fuels. Hydrogen-based devices are of particular interest due to their significant efficiency and reliability. Reforming technologies are among the most economic and efficient ways of producing hydrogen. In this paper we consider the regulation of hydrogen outflow in an ethanol steam reformer (ESR). In particular, a fast model predictive control approach based on a finite step response model of the process is proposed. Simulations performed using a more realistic non-linear model show the effectiveness of the proposed approach in driving the ESR to different operating conditions while fulfilling input and output constraints.Peer ReviewedPostprint (author's final draft
