    Cherimoya, a subtropical fruit growing in sorne American countries and restricted areas of the south of Spain, is one of the richest sources of free amino acids. The effects of post-harvest temperatures on the texture and free amino acid profile of this fruit were studied. Fruit firmness was determined with a pressure tester, and free amino acids were measured by ion-exchange colum chromatography using an automatic amino acid analyzer. Cherimoya was characterized by high contents of proline and citrulline. Glutamic acid and glutamine were also present but at lower levels.     Changes in the content of free amino acids In cherimoya fruits at various ripening temperatures

    The variations in the content of free amino acids in fruits during ripening is a matter of interest since these compounds are related to their flavour, taste and structure. This study was designed to determine the changes of free amino acids in ripening cherimoya fruits (Annona cherimolia var. Fino de Jete, evaluating the influence of the storage temperature afier harvesting. Methanolic fruit extracts of epicarp and mesocarp were used for quantification of free amino acids which were analyzed by ion-exchange chromatography. Citrulline and proline were detected in relatively high amounts. Glutamate, glutamine, aspartate, asparagine and the non proteinogenic taurine, y-aminobutyric and y-aminoadipic acids were found in concentrations that varied between 2-6% of the total free amino acids content. Ripening increased the contents of amino acids, mainly proline, citrulline and glutamate when cherimoya storage at 20 oC or 80 oC. These increases did not occur when the storage temperature was maintained at 4 oC suggesting that chilling injury markedly affects the synthesis pathway of proline from glutamateLas variaciones en el contenido de aminoácidos libres en frutos durante la maduración es un tema de interés debido a la relación de estos compuestos con su olor, sabor y estructura. Este estudio se ha diseñado para determinar los cambios en aminoácidos libres durante la maduración de los frutos del chirimoyo (Annona cherimolia varo Fino de Jete), evaluando la influencia de la temperatura de almacenamiento después de la recolección. Extractos metanólicos de epicarpio y mesocarpio del fruto se usaron para la cuantificación de los aminoácidos libres, los cuales fueron analizados por cromatografia de intercambio iónico. Citrulina y prolina se detectaron en cantidades relativamente altas. Glutamato, glutamine, aspartato, asparragina y los no proteinogenéticos taurina, ácido y-aminobutírico y ácido Y-aminoadípico fueron encontrados en concentraciones que variaron entre 2-6% del total del contenido de aminoácidos libres. La maduración incrementó el contenido de aminoácidos, sobre todo de prolina, citrulina y glutamato, cuando los chirimoyos se almacenaron a 20 oC o 8 oc. Este incremento no ocurrió cuando la temperatura de almacenamiento se mantuvo a 4 oC, sugiriendo que el daño por frío afecta marcadamente a la vía de síntesis de la prolina a partir de glutamat

    A new method to trace colloid transport pathways in macroporous soils using X‐ray computed tomography and fluorescence macrophotography

    The fast and deep percolation of particles through soil is attributed to preferential flow pathways, and their extent can be critical in the filtering of particulate pollutants in soil. Particle deposition on the pore walls and transport between the pores and matrix modulate the preferential flow of particulate pollutants. In the present research, we developed a novel method of combining fluorescence macrophotography and X‐ray computed tomography (CT) to track preferential pathways of colloidal fluorescent microspheres (MS) in breakthrough experiments. We located accumulations of MS by fluorescence imaging and used them to delimit the deposition structures along the preferential colloid pathways by superimposing these images on the 3‐D pore network obtained from CT. Advection–diffusion with transport parameters from the dual‐porosity equation correlated with preferential pathway features across different soil management techniques. However, management did not influence the morphology of the MS preferential pathways. Preferential flow occurred in only a small fraction of the total pore network and was controlled by pores connected to the soil surface and by matrix density

    Mutation increasing β-carotene concentrations does not adversely affect concentrations of essential mineral elements in pepper fruit

    <div><p>Vitamin and mineral deficiencies are prevalent in human populations throughout the world. Vitamin A deficiency affects hundreds of millions of pre-school age children in low income countries. Fruits of pepper (<i>Capsicum annuum</i> L.) can be a major dietary source of precursors to Vitamin A biosynthesis, such as β-carotene. Recently, pepper breeding programs have introduced the orange-fruited (<i>of</i>) trait of the mutant variety Oranzheva kapiya, which is associated with high fruit β-carotene concentrations, to the mutant variety Albena. In this manuscript, concentrations of β-carotene and mineral elements (magnesium, phosphorus, sulphur, potassium, zinc, calcium, manganese, iron and copper) were compared in fruit from P31, a red-fruited genotype derived from the variety Albena, and M38, a genotype developed by transferring the orange-fruited mutation (<i>of</i>) into Albena. It was observed that fruit from M38 plants had greater β-carotene concentration at both commercial and botanical maturity (4.9 and 52.7 mg / kg fresh weight, respectively) than fruit from P31 plants (2.3 and 30.1 mg / kg fresh weight, respectively). The mutation producing high β-carotene concentrations in pepper fruits had no detrimental effect on the concentrations of mineral elements required for human nutrition.</p></div

    Condylar volume and surface in Caucasian young adult subjects

    <p>Abstract</p> <p>Background</p> <p>There have been no quantitative standards for volumetric and surface measurements of the mandibular condyle in Caucasian population. However, the recently developed cone-beam computed tomography (CBCT) system allows measurement of these parameters with high accuracy.</p> <p>Methods</p> <p>CBCT was used to measure the condylar volume, surface and the volume to surface ratio, called the Morphometric Index (MI), of 300 temporo-mandibular joints (TMJ) in 150 Caucasian young adult subjects, with varied malocclusions, without pain or dysfunction of TMJs.</p> <p>Results</p> <p>The condylar volume was 691.26 ± 54.52 mm<sup>3 </sup>in males and 669.65 ± 58.80 mm<sup>3 </sup>in, and was significantly higher (<it>p</it>< 0.001) in the males. The same was observed for the condylar surface, although without statistical significance (406.02 ± 55.22 mm<sup>2 </sup>in males and 394.77 ± 60.73 mm<sup>2 </sup>in females).</p> <p>Furthermore, the condylar volume (693.61 ± 62.82 mm<sup>3 </sup>) in the right TMJ was significantly higher than in the left (666.99 ± 48.67 mm<sup>3</sup>, <it>p </it>< 0.001) as was the condylar surface (411.24 ± 57.99 mm<sup>2 </sup>in the right TMJ and 389.41 ± 56.63 mm<sup>2 </sup>in the left TMJ; <it>t </it>= 3.29; <it>p </it>< 0.01). The MI is 1.72 ± 0.17 for the whole sample, with no significant difference between males and females or the right and left sides.</p> <p>Conclusion</p> <p>These data from temporomandibular joints of patients without pain or clinical dysfunction might serve as examples of normal TMJ's in the general population not seeking orthodontic care.</p

    The Neglected Tropical Diseases of Latin America and the Caribbean: A Review of Disease Burden and Distribution and a Roadmap for Control and Elimination

    The neglected tropical diseases (NTDs) represent some of the most common infections of the poorest people living in the Latin American and Caribbean region (LAC). Because they primarily afflict the disenfranchised poor as well as selected indigenous populations and people of African descent, the NTDs in LAC are largely forgotten diseases even though their collective disease burden may exceed better known conditions such as of HIV/AIDS, tuberculosis, or malaria. Based on their prevalence and healthy life years lost from disability, hookworm infection, other soil-transmitted helminth infections, and Chagas disease are the most important NTDs in LAC, followed by dengue, schistosomiasis, leishmaniasis, trachoma, leprosy, and lymphatic filariasis. On the other hand, for some important NTDs, such as leptospirosis and cysticercosis, complete disease burden estimates are not available. The NTDs in LAC geographically concentrate in 11 different sub-regions, each with a distinctive human and environmental ecology. In the coming years, schistosomiasis could be eliminated in the Caribbean and transmission of lymphatic filariasis and onchocerciasis could be eliminated in Latin America. However, the highest disease burden NTDs, such as Chagas disease, soil-transmitted helminth infections, and hookworm and schistosomiasis co-infections, may first require scale-up of existing resources or the development of new control tools in order to achieve control or elimination. Ultimately, the roadmap for the control and elimination of the more widespread NTDs will require an inter-sectoral approach that bridges public health, social services, and environmental interventions

    A Comparison of the Sensitivity and Fecal Egg Counts of the McMaster Egg Counting and Kato-Katz Thick Smear Methods for Soil-Transmitted Helminths

    Currently, in public health, the reduction in the number of eggs excreted in stools after drug administration is used to monitor the efficacy of drugs against parasitic worms. Yet, studies comparing diagnostic methods for the enumeration of eggs in stool are few. We compared the Kato-Katz thick smear (Kato-Katz) and McMaster egg counting (McMaster) methods, which are commonly used diagnostic methods in public and animal health, respectively, for the diagnosis and enumeration of eggs of roundworms, whipworms and hookworms in 1,536 stool samples from children in five trials across Africa, Asia and South America. The Kato-Katz method was the most sensitive for the detection of roundworms, but there was no significant difference in sensitivity between the methods for hookworms and whipworms. The sensitivity of the methods differed across the trials and magnitude of egg counts. The Kato-Katz method resulted in significantly higher egg counts, but these were subject to lack of accuracy caused by intrinsic properties of this method. McMaster provided more reliable estimates of drug efficacies. We conclude that the McMaster is an alternative method for monitoring large-scale treatment programs. It allows accurate monitoring of drug efficacy and can be easily performed under field conditions

    Safety and immunomodulatory effects of three probiotic strains isolated from the feces of breast-fed infants in healthy adults: SETOPROB study

    We previously described the isolation and characterization of three probiotic strains from the feces of exclusively breast-fed newborn infants: Lactobacillus paracasei CNCM I-4034, Bifidobacterium breve CNCM I-4035 and Lactobacillus rhamnosus CNCM I-4036. These strains were shown to adhere to intestinal mucus in vitro, to be sensitive to antibiotics and to resist biliary salts and low pH. In the present study, a multicenter, randomized, double-blind, placebo-controlled trial with 100 healthy volunteers in three Spanish cities was carried out to evaluate the tolerance, safety, gut colonization and immunomodulatory effects of these three probiotics. Volunteers underwent a 15-day washout period, after which they were randomly divided into 5 groups that received daily a placebo, a capsule containing one of the 3 strains or a capsule containing a mixture of two strains for 30 days. The intervention was followed by another 15-day washout period. Patients did not consume fermented milk for the entire duration of the study. Gastrointestinal symptoms, defecation frequency and stool consistency were not altered by probiotic intake. No relevant changes in blood and serum, as well as no adverse events occurred during or after treatment. Probiotic administration slightly modified bacterial populations in the volunteers’ feces. Intestinal persistence occurred in volunteers who received L. rhamnosus CNCM I-4036. Administration of B. breve CNCM I-4035 resulted in a significant increase in fecal secretory IgA content. IL-4 and IL-10 increased, whereas IL-12 decreased in the serum of volunteers treated with any of the three strains. These results demonstrate that the consumption of these three bacterial strains was safe and exerted varying degrees of immunomodulatory effects.Part of the research currently in progress in the authors' laboratory is funded by the company Hero Spain, S.     Short- and Long-Term Biomarkers for Bacterial Robustness: A Framework for Quantifying Correlations between Cellular Indicators and Adaptive Behavior

    The ability of microorganisms to adapt to changing environments challenges the prediction of their history-dependent behavior. Cellular biomarkers that are quantitatively correlated to stress adaptive behavior will facilitate our ability to predict the impact of these adaptive traits. Here, we present a framework for identifying cellular biomarkers for mild stress induced enhanced microbial robustness towards lethal stresses. Several candidate-biomarkers were selected by comparing the genome-wide transcriptome profiles of our model-organism Bacillus cereus upon exposure to four mild stress conditions (mild heat, acid, salt and oxidative stress). These candidate-biomarkers—a transcriptional regulator (activating general stress responses), enzymes (removing reactive oxygen species), and chaperones and proteases (maintaining protein quality)—were quantitatively determined at transcript, protein and/or activity level upon exposure to mild heat, acid, salt and oxidative stress for various time intervals. Both unstressed and mild stress treated cells were also exposed to lethal stress conditions (severe heat, acid and oxidative stress) to quantify the robustness advantage provided by mild stress pretreatment. To evaluate whether the candidate-biomarkers could predict the robustness enhancement towards lethal stress elicited by mild stress pretreatment, the biomarker responses upon mild stress treatment were correlated to mild stress induced robustness towards lethal stress. Both short- and long-term biomarkers could be identified of which their induction levels were correlated to mild stress induced enhanced robustness towards lethal heat, acid and/or oxidative stress, respectively, and are therefore predictive cellular indicators for mild stress induced enhanced robustness. The identified biomarkers are among the most consistently induced cellular components in stress responses and ubiquitous in biology, supporting extrapolation to other microorganisms than B. cereus. Our quantitative, systematic approach provides a framework to search for these biomarkers and to evaluate their predictive quality in order to select promising biomarkers that can serve to early detect and predict adaptive traits
